Plant carnivory represents an exceptional means to acquire N. Snap traps of Dionaea muscipula serve two functions, and provide both N and photosynthate. Using (13)C/(15)N-labelled insect powder, we performed feeding experiments with Dionaea plants that differed in physiological state and N status (spring vs. autumn plants). We measured the effects of (15)N uptake on light-saturated photosynthesis (A(max)), dark respiration (R(D)) and growth. Depending on N status, insect capture briefly altered the dynamics of R(D)/A(max), reflecting high energy demand during insect digestion and nutrient uptake, followed by enhanced photosynthesis and growth. Organic N acquired from insect prey was immediately redistributed, in order to support swift renewal of traps and thereby enhance probability of prey capture. Respiratory costs associated with permanent maintenance of the photosynthetic machinery were thereby minimized. Dionaea's strategy of N utilization is commensurate with the random capture of large prey, occasionally transferring a high load of organic nutrients to the plant. Our results suggest that physiological adaptations to unpredictable resource availability are essential for Dionaea's success with regards to a carnivorous life style.

The systemic fungal endophyte of the grass Achnatherum inebrians, Epichloë gansuensis, has important roles in enhancing resistance to biotic and abiotic stresses. In this work, we first evaluated the effects of E. gansuensis on nitrogen metabolism, nitrogen use efficiency, and stoichiometry of A. inebrians under varying nitrogen concentrations. The results demonstrated that E. gansuensis significantly improved the growth of A. inebrians under low nitrogen conditions. The fresh and dry weights, nitrogen reductase, nitrite reductase, and glutamine synthetase activity, NO, NH, N, and P content, and also the total N accumulation, N utilization efficiency, and N uptake efficiency were all higher in leaves of A. inebrians with E. ganusensis (E+) plants than A. inebrians plants without this endophyte (E-) under low nitrogen availability. In conclusion, E. gansuensis has positive effects on improving the growth of A. inebrians under low-nitrogen conditions by modulating the enzymes of nitrogen metabolism and enhancing nitrogen use efficiency.

To better utilize native pasture at the high altitude region, three-consecutive-year feeding experiments and a total of seven metabolism trials were conducted to evaluate the impact of three forage stages of maturity on the chemical composition, nutrient digestibility, and energy metabolism of native forage in Tibetan sheep on the Qinghai-Tibetan Plateau (QTP). Forages were harvested from June to July, August to October, and November to December of 2011 to 2013, corresponding to the vegetative, bloom, and senescent stages of the annual forages. Twenty male Tibetan sheep were selected for each study and fed native forage ad libitum. The digestibility of DM, OM, CP, NDF, ADF, DE, DE/GE, and ME/GE were greatest (P< 0.01) from the vegetative stage, intermediate (P < 0.01) from the bloom stage, and least (P < 0.01) from the senescent stage. Nutrient digestibility and energy parameters correlated positively (linear, 0.422 to 0.778; quadratic, 0.568 to 0.815; P < 0.01) with the CP content of forage but correlated negatively with the content of NDF (linear, 0.343 to 0.689; quadratic, 0.444 to 0.777; P ≤ 0.02), ADF (linear, 0.563 to 0.766; quadratic, 0.582 to 0.770; P < 0.01), and ether extract (EE, linear, 0.283 to 0.574; quadratic, 0.366 to 0.718; P ≤ 0.04) of forage. For each predicted variable, the prediction of DMI expressed as grams per kilogram of BW (g/kg BW·d) yielded a greater R2 value (0.677 to 0.761 vs. 0.616 to 0.711) compared with the equations of DMI expressed as g/kg metabolic BW by step-wise regression. The results suggest that parameters of forage CP, NDF, and ADF content were most closely related to nutrient digestibility. Contrary to previous studies, in this study, ADF content had a greater linear relationship (0.766 vs. 0.563 to 0.732) with OM digestibility than the other parameters of nutrient digestibility. The quadratic relationship between forage CP content and CP digestibility indicates that when forage CP content exceeds the peak point (9.7% DM in the present study), increasing forage CP content could decrease CP digestibility when Tibetan sheep were offered native forage alone on the QTP. Additionally, using the forage CP, EE, NDF, and ADF content to predict DMI (g/kg BW·d) yielded the best fit equation for Tibetan sheep living in the northeast portion of the QTP.

Spring black stem and leaf spot of lucerne (alfalfa, Medicago sativa L.), caused by Phoma medicaginis, is an important disease in temperate regions of the world. It is now a serious disease threatening global lucerne production. This experiment was designed to test the combined effects of the arbuscular mycorrhizal (AM) fungus Funneliformis mosseae and the rhizobium Sinorhizobium medicae on growth, nutrient uptake and disease severity in lucerne. The results showed that F. mosseae increased plant phosphorus and nitrogen uptake and plant dry weight, and this beneficial effect was enhanced when in association with S. medicae. Rhizobial and AM fungal effects were mutually promoting; inoculation with AM fungus significantly increased the formation of root nodules, and inoculation with rhizobium increased the percentage of root length colonised by AM fungus (P < 0.05). After infection with P. medicaginis, typical leaf spot symptoms with the lowest disease incidence and disease index occurred on plants that were host to both F. mosseae and S. medicae. Plants with both symbiotic microorganisms had higher activities (concentrations) of phenylalanine ammonia-lyase, chitinase, β-1,3-glucanase, lignin, hydroxyproline-rich glycoprotein and jasmonic acid. Therefore, the tested AM fungus (F. mosseae) and rhizobium (S. medicae) have the potential to reduce damage and yield loss in lucerne from spring black stem and leaf spot caused by P. medicaginis.

Leaf spot of perennial ryegrass (Lolium perenne) caused by Bipolaris sorokiniana is an important disease in temperate regions of the world. We designed this experiment to test for the combined effects of the arbuscular mycorrhizal (AM) fungus Claroideoglomus etunicatum and the grass endophyte fungus Epichloë festucae var. lolii on growth and disease occurrence in perennial ryegrass. The results show that C. etunicatum increased plant P uptake and total dry weight and that this beneficial effect was slightly enhanced when in association with the grass endophyte. The presence in plants of both the endophyte and B. sorokiniana decreased AM fungal colonization. Plants inoculated with B. sorokiniana showed the typical leaf spot symptoms 2 weeks after inoculation and the lowest disease incidence was with plants that were host to both C. etunicatum and E. festucae var. lolii. Plants with these two fungi had much higher activity of peroxidases (POD), superoxide dismutase (SOD) and catalase (CAT) and lower values of malondialdehyde (MDA) and hydrogen peroxide (HO). The AM fungus C. etunicatum and the grass endophyte fungus E. festucae var. lolii have the potential to promote perennial ryegrass growth and resistance to B. sorokiniana leaf spot.

SummaryCarnivorous Dionaea muscipula operates active snap traps for nutrient acquisition from prey; so what is the role of D. muscipula's reduced root system?We studied the capacity for nitrogen (N) acquisition via traps, and its effect on plant allometry; the capacity of roots to absorb NO 3 À , NH 4 + and glutamine from the soil solution; and the fate and interaction of foliar-and root-acquired N. Feeding D. muscipula snap traps with insects had little effect on the root : shoot ratio, but promoted petiole relative to trap growth. Large amounts of NH 4 + and glutamine were absorbed upon root feeding. The high capacity for root N uptake was maintained upon feeding traps with glutamine. High root acquisition of NH 4 + was mediated by 2.5-fold higher expression of the NH 4 + transporter DmAMT1 in the roots compared with the traps. Electrophysiological studies confirmed a high constitutive capacity for NH 4 + uptake by roots. Glutamine feeding of traps inhibited the influx of 15 N from root-absorbed 15 N/ 13 C-glutamine into these traps, but not that of 13 C. Apparently, fed traps turned into carbon sinks that even acquired organic carbon from roots. N acquisition at the whole-plant level is fundamentally different in D. muscipula compared with noncarnivorous species, where foliar N influx down-regulates N uptake by roots.
