The principal physiological function of Iipoic acid appears to 02 its action as coenzyme in the oxidative decarboxylation of a-0x0 acids. In order to find out whether Iipoic acid is essential to higher organisms, structurally modified lipoic acids were prepared as potential antagonists. Only 4-oxalipoamide promotes the growth of S. faecalis 8043; the other substances tested are inactive or else inhibit growth. In experiments to determine the substrate specificity of lipoamide oxidoreductase and its inhibition by structurally modified lipoic acids, no strong inhibitors were discovered. -Ring strain, spectra, and ring cleavage of compounds containing the 1,2-dithiolane ring are discussed in relation to the biological function and the reactivity of Iipoic acid.
Enolather und Enolester symmetrischer Cyclanone lassen sich in praparativem MaBstab mit Ozon zu w-Aldehydsauren bzw. deren Estern aufspalten, die ohne Isolierung acetalisiert und verestert werden konnen. Bei der konkurrierenden Ozonisierung eines Vinylathers und eines Olefins wird die polare Doppelbindung im Vinylather schneller gespalten. -Die o-Aldehydesteracetale lassen sich in guter Ausbeute mit Lithiumaluminiumhydrid zu o-Hydroxyacetalen reduzieren. o-Hydroxq-octanal wurde mit Malonsaure zu lO-Hydroxy-A2-decensaure kondensiert, einem Inhaltsstoff des Weiselsaftes. OZONISIERUNG CYCLISCHER VINYLATHERVinylather sind bisher nur vereinzeltim Zusammenhang mit analytischen Fragenozonisiert worden 2.3) : teils zur Strukturaufklarung der zugrundeliegenden Ketone, teils zur Bestimmung der Enolisierungsrichtung bei unsymmetrischen Carbonylverbindungen. Wir fanden, da13 sich die Ozonspaltung der Enolather cyclischer Ketone vorziiglich zur praparativen Darstellung von a-Aldehydsauren eignet. w-Formylcarbonsauren, die bei Naturstoffsynthesen eingesetzt wurden4-6), hat man durch RosENMuND-Reduktion von Dicarbonsaure-ester-chloriden oder durch Ozonspaltung. ungesattigter Carbonsauren7) hergestellt. Abgesehen von der Schwierigkeit, ROSENMUND-Reduktionen gut reproduzierbar durchzufiihrenheute wiirde man deshalb wohl die Reduktion mit Lithiumtributoxyaluminiumhydrid vorziehen -, sind die Estersaurechloride meist nur in grol3en Ansatzen rein und in guter Ausbeute m gewinnen, da bei kleinen Mengen die exakte Trennung von Dicarbonsanre, Estersaure und Diester oft schwierig ist. -Zur Her-1) Kurze Mitteilung: U. Fessenden [142] hat verschiedene experimentelle Methoden beschrieben, nach denen derartig schnelle Reaktionen verfolgt werden konnen. Er konnte die Lebensdauer T = 7,4 & 0,l msec. des durch Bestrahlung von k h a n niit 2,s MeV-Elektronen gebildeten khylradikals bei -170 "C messen. Aus der Temperaturabhangigkeit von T konnte aufierdem die Aktivierungsenergie der [142] R. W'. Fessenden, J. physic. Chem. 68, 1508(1964). Rekombinationsreaktion zu 830 cal/Mol berechnet werden. Hiiufig ist bereits der Nachweis, dal3 bei einer Reaktion iiberhaupt Radikale beteiligt sind, von Bedeutung. Das trifft vor allem fur biochemische Probleme zu, bei denen der Reaktionsablauf noch nicht geklart ist. Die EPR-Spektren auftretender freier Radikale bestehen meist aus einer einzigen breiten Linie bei g = 2, so daB nur Linienbreite und Intensitat quantitative Aussagen ermoglichen [143]. Die physiologische Aufgabe der Liponsaure scheint hauptsachlich die Coenzymfunkticn bei der oxydativen Decarboxylierung von u-Oxosauren zu sein. Um zu priifen, ob Lipomaure ein essentieller Wirkstoff fur den hoheren Organismus ist, wurdcn strukturveranderte Liponsauren hergestellt, in der Hofftiung, Antagonisten zu erhalten. Nur 4-Oxa-liponsaureamid fordert das Wachstum von S. faecalis 8043; die anderen gepriijten Substanzen verhalten sQh inert oder hemmen das Wachstum. Bei Versuchen zur Hemmung und Substratspezijitat der Lipoamid-oxydoredirktase rnit strukturveranderten Liponsauren wurden keine stark wirksamen Inhibitoren gefunden. -Im Hinblick auf die biologische Funktion der Liponstiure und ihre Reaktionsfahigkeit werden Ringspannung, Spektren und offriung des Rings bei Verbindungen mit dem I .2-Dithiolanring diskutiert . Anlagerung von Adipinslure-athylester-chlorid an Acetylen, Umsetzung mit alkoholischer Lauge und Hydrieren des P-Dicarbonylderivates lieferte 6-Hydroxy-8-methoxy-octanslure-( I)-athylester (XIII) (Syntheseweg A). -Umsetzung von Athyl-vinyl-ather mit dem Acetal des Adipinaldehydsaure-Ithylesters und folgende Hydrierung fiihrte zu 6.8-Diathoxy-octansaure-(I)-ester (XXIV) (Syntheseweg B). -XI11 und XXIV wurden uber das 6.8-Bisisothiuroniumsalz in dJu-Liponsaure umgewandelt. Die guten Ausbeuten beim Einsatz von in 6-Stellung acetylierten 6.8-Dihydroxy-octanslure-Derivaten gestatteten RiickschlIisse auf den Chemismus der Solvolyse sekundlrer Ester. Forschungen der letzten Jahre haben die d,l-u-Liponsaure (I) 1) als Wachstumsfaktor fur Bakterien undin hoherer Konzentrationfiir Krebsgewebez) erkannt. Ihr Vorkommen in einigen Enzymsystemen und damit in vielen Nahrungsmitteln und Organen lassen in ihr einen wichtigen Nahrungsfaktor' (Vitamin?) vermuten. -Umfangreiche Anstrengungen haben fur die a-Liponshre viele Synthesen gebracht, die uber die verschiedenartigsten Wege verlaufen3-'3). Eine einfache und zugleich billige Darstellungsmethode dieses wichtigen Naturstoffes ist jedoch bisher nicht bekannt geworden. -Der erste der von uns unten beschriebenen neuen Wege (A) hat sich auch in halbtechnischen Ansatzen bewahrt und ermoglicht die Her-1) L.
