INDIRECT SPECTROPHOTOMETRIC DETERMINATION OF CAPSAICINOIDS IN Capsicum PEPPERS USING THE REACTION OF COMPLEX Co(II) WITH 4-(2-PYRIDYLAZO) RESORCINOL. Capsaicinoids (CAPS) are substances responsible for pungency in Capsicum. It is important to quantify these types of compounds owing to their broad application in food, pharmaceuticals, cosmetics and chemical weapons. In this work, we developed an indirect spectrophotometric method based on the colorimetric reaction between CAPS, Co(II) 3.10×10-5 mol L -1 and 4-(2-pyridylazo) resorcinol (PAR) 6.23×10 -5 and, in cachaça:water 92:8v/v solutions, for quantification of total CAPS in Capsicum peppers. The product of the reaction is CoPAR 2 CAPS 2 and its absorption in aquo-ethanolic solution at 510 nm is proportional to the total CAPS concentration from 0.60 to 17.94 mg L -1. The values of limit of detection and limit of quantification were 0.0004 and 0.001 mg of CAPS/g of pepper, respectively, with 4% relative standard deviation. The developed method yielded similar results to those obtained from high performance liquid chromatography, with 95% of confidence.Keywords: capsaicinoids; peppers; spectrophotometric quantification.
INTRODUÇÃOCapsaicinoides (CAPS) são alcaloides muito encontrados em vegetais, nos quais atuam nos mecanismos de defesa das espécies, conferindo picância (ardência, pungência) aos frutos e sementes. CAPS também correspondem aos principais agentes terapêuticos naturais com ação anestésica, analgésica e também atuam como psicoestimulantes e neurodepressores.1,2 Pimentas e pimentões Capsicum representam importantes fontes naturais de CAPS, 3 sendo que capsaicina (CAP) e dihidrocapsaicina (DHCAP) representam cerca de 90% dos CAPS mais comumente encontrados nessas espécies. 4 O gênero Capsicum engloba cerca de 20 espécies, normalmente classificadas de acordo com o nível de domesticação e cerca de 10 espécies silvestres. As pimentas já disseminadas pelos continentes são classificadas pelos botânicos em cinco espécies: as primeiras Capsicum baccatum e Capsicum pubescens, depois Capsicum annuum, Capsicum chinense e Capsicum frutescens. Essas três últi-mas passaram a liderar a comercialização atual. De todas as várias espécies conhecidas, essas cinco são as mais cultivadas.
5Pimentas Capsicum apresentam propriedades químicas distintas e sem parentesco com outras pimentas, como a pimenta-do-reino ou pimenta-preta (Piper nigrum L., da família Piperaceae), a pimenta--rosa (Schinus molle L., da família Anacardiaceae) e a pimenta-dajamaica (Pimenta officinalis Lindl., da família Myrtaceae). Entre as Capsicum, os pimentões diferem das pimentas apenas por sua picância ou pungência reduzida, às vezes nem notada. 6 A picância das pimentas Capsicum é enganosa, pois embora as pimentas pequenas sejam frequentemente consideradas as mais ardidas, pimentas grandes nem sempre são muito suaves.7 Os CAPS acumulam-se na vesícula da epiderme na parte interna dos frutos e nas sementes em níveis que variam com o genótipo, maturidade e condições de crescimento, sendo liberados na...
