Objective: This study aimed to produce data on sperm quality while maintaining the integrity of sperm DNA samples taken from patients submitted to in vitro fertilization (IVF) procedures at our center, and determine whether increased levels of histones were associated with sperm DNA damage and decreased fertilization, cleavage, and pregnancy rates. Such findings might shed light on the physiology and outcomes of pregnancy. Methods: Semen samples from 27 patients divided into two groups were analyzed. The case group included individuals offered IVF; the control group had subjects with normal spermograms. Sperm DNA structure was assessed through phosphorylated histone H2AX analysis by flow cytometry. Results:The patients with altered sperm parameters had more histones in sperm chromatin than the individuals with normal sperm parameters. Conclusion: Results indicated that increased levels of histone in sperm chromatin do not affect embryo production, but affect the cleavage rate, embryo quality, and might thus reduce pregnancy rates. The integrity of the paternal genome is of paramount importance in the initiation and maintenance of a viable pregnancy in patients treated with assisted reproduction technology procedures. Further studies on sperm diagnostic tests at a nuclear level might improve the treatment offered to infertile couples.
Objetivo: Comparar os crioprotetores Dimetil Sulfoxido (DMSO), Etileno Glicol (EG) e sua associação, para criopreservação do córtex ovariano de ovelha. Metodologia:Os fragmentos ovarianos foram divididos em 3 partes: 1. Uma parte foi destinada a análise do material fresco. 2. Uma segunda parte foi incubada com as soluções de congelamento contendo 1,5M EG ou 1,5M DMSO ou 1,5M EG + 1,5M DMSO e lavada para diluição dos crioprotetores. 3 . A terceira foi submetida à ação dos crioprotetores (EG 1,5M; DMSO 1,5M e EG+ DMSO 1,5M), e criopreservada. Em todos os grupos, uma parte da amostra foi submetida ao isolamento dos folículos pré-antrais e o restante foi destinado à análise ultra-estrutural. Resultado:Após o isolamento dos folículos primordiais (controle), a porcentagem de folículos viáveis foi 78,9%. A percentagem de folículos viáveis apenas na exposição dos crioprotetores 1,5M EG, 1,5M DMSO e 1,5M EG + 1,5M DMSO foi de 77,1%, 68,4% e 60,7% respectivamente. Após a criopreservação foi de 75%, 60% e 55,6% respectivamente. Na análise ultra-estrutural os folículos primordiais oriundos de fragmentos ovarianos frescos ou expostos aos crioprotetores apresentaram morfologia semelhante. No entanto, nas amostras congeladas observou-se alteração das mitocôndrias em todos os grupo. Apesar disso a integridade das demais organelas foi preservada nos folículos criopreservados com EG, enquanto que nos outros grupos (DMSO e associação) foi observado excesso de vesiculação no citoplasma dos oócitos e dilatação da membrana nuclear indicando degeneração. Conclusão: O Etileno Glicol parece ser o crioprotetor mais adequado para criopreservação do tecido ovariano de ovino.
Objective:The main objective of this study was to evaluate viability, embryo implantation rate and clinical pregnancy in patients undergoing controlled ovarian stimulation. material and methods: Viable embryos of patients deriving from in vitro -zing and thawing of embryos were evaluated during the periods from January 2010 to December 2012 in the Clinical Reproduction, Presidente Prudente -SP. Results: Total of 176 embryos was frozen. The average number of frozen/thawed embryos per patient was 5, totaling 140 thawed embryos. These thawed embryos, 78,6% (110/140) displayed viable morphologically (at least 50% from intact blastomeres) and 75% (105/140) of thawed embryos were transferred. In 28 cycles in which there was transference, an average of 3.75 embryos were transferred per patient, from which resulted in 17 biochemical pregnancies and of these 8 pregnancies reached term (28,6%) and embryonic implantation rate was 20%. Conclusion: It can be concluded that the method of survival rate, allowing the storage of surplus embryos with satisfactory pregnancy rates in thawing cycles. key words: RESUmO Objetivo: implantação e gestação com intuito de introduzir como procedimento de rotina em pacientes com hiper-estímulo ovariano. materiais e métodos provenientes de fertilização in vitro a dezembro de 2012 na Clínica Reproduction, Presidente Prudente -SP. Resultados: sentaram-se morfologicamente viáveis (pelo menos 50% descongelados foram transferidos. nos 28 ciclos em que transferidos por paciente, dos quais resultaram em 17 gesta-(28,6%) e taxa de implantação embrionária foi de 20%.
