Neosporosis primarily affects cattle and dogs and is not currently considered a zoonotic disease. Toxoplasmosis is a zoonosis with a worldwide distribution that is asymptomatic in most cases, but when acquired during pregnancy, it can have serious consequences. The seropositivity rates determined by the indirect fluorescent antibody test for Neospora caninum (N. caninum) and Toxoplasma gondii (T. gondii) were 24.3% (49 samples) and 26.8% (54 samples), respectively. PCR positivity for N. caninum was observed in two samples of cord blood (1%) using the Nc5 and ITS1 gene, positivity for T. gondii was observed in 16 samples using the primer for the B1 gene (5.5% positivity in cord blood and 2.5% positivity in placental tissue). None of the samples showed structures characteristic of tissue cysts or inflammatory infiltrate on histopathology. Significant associations were observed only between N. caninum seropositivity and the presence of domestic animals (p = 0.039) and presence of dogs (p = 0.038) and between T. gondii seropositivity and basic sanitation (p = 0.04). This study obtained important findings regarding the seroprevalence and molecular detection of N. caninum and T. gondii in pregnant women; however, more studies are necessary to establish a correlation between risk factors and infection.
The present study evaluated the behavior of Rhipicephalus microplus naturally infesting Brangus cattle raised with Nellore animals in the municipality of Água Clara, state of Mato Grosso do Sul, Central Brazil. For the field experiment composed of a group with 15 Brangus animals and 15 Nellore animals and a control group with 30 Brangus cattle, all at the post-weaning stage. The 2 groups were kept for 6 months in 2 adjacent paddocks, naturally infested with ticks, at a stocking rate of approximately 0.6 of an animal unit per hectare (AU/ha). Every 18 days, the animals were weighed, and ticks on both sides of each bovine were counted. Every 36 days, blood was collected for hematocrit measurement and molecular diagnosis of the 3 pathogens that cause tick fever. The Brangus animals had 6.8 times more ticks than the Nellore cattle. No significant difference was observed in the weight gain of the Brangus and Nellore animals, and no correlation between the number of ticks and weight was observed in either group. During the 6 months of the study, all animals presented normal hematocrit, were positive for Babesia bigemina and Anaplasma marginale, and were negative for Babesia bovis. Animals in Group 1 produced 27% fewer ticks than the animals in Group 2. The production rate of engorged female ticks in Nellore and Brangus cattle was 0.83% and 2.01%, respectively. Considering the infestation pattern required to produce 10 engorged females/day/animal, the Nellore and Brangus animals required 1,204 and 497 larvae/day, respectively. Our estimates showed that Brangus cattle need only 11.3% of the larvae in the pastures with Nellore cattle to maintain the infestation level observed in the present study. Brangus animals raised with Nellore cattle do not have a lower parasitic load and continue to suffer from tick infestation. Finally, Nellore cattle raised with Brangus cattle do not act as a cleaner because this grouping does not contribute to a reduced infestation in the Brangus animals. Resumo: Este trabalho teve o objetivo de avaliar o comportamento da infestação de carrapatos (Rhipicephalus microplus) em bovinos da raça Brangus associado com Nelore no Brasil central em rebanho infestado naturalmente na região do município de Água Clara, MS, Brasil. Para o experimento de campo um grupo de 15 animais em inicio de recria da raça Brangus e 15 animais da raça Nelore e outro grupo controle com 30 animais Brangus, foram mantidos em dois piquetes geminados com lotação de aproximadamente 0,6 unidade animal por hectare (Ua/ha). A cada 18 dias os carrapatos foram contados dos dois lados do corpo dos bovinos após a pesagem dos mesmos. A cada 36 dias foi colhido sangue para realizar o hematócrito e o diagnóstico molecular para os três agentes da Tristeza Parasitária Bovina (TPB). The objective of this work was to evaluate the diversity of ticks associated with free-living animals and to investigate new host records for ticks. Ticks were collected from animals rescued during the flood of the Jamari River in the municipality of Ariquemes, state of Rondônia, North Region of Brazil. A total of 39 animals were captured, out of which 10 were amphibians, 19 were reptiles and 10 were mammals. A total of 127 ticks of the Amblyomma genus were collected from these animals, distributed among seven species: Amblyomma dissimile, Amblyomma geayi, Amblyomma humerale , Amblyomma longirostre, Amblyomma nodosum , Amblyomma rotundatum and Amblyomma varium. In addition, one specimen of Rhipicephalus (Boophilus) microplus was collected. Among these specimens, 85 were adults and 42 were nymphs, with A. rotundatum being the most prevalent species. An Amblyomma spp. larvae was also collected from a lizard (Uranoscodon superciliosus), and one Amblyomma calcaratum and one Amblyomma dubitatum were recovered from the environment, thus totaling 130 ticks. Among the Ixodidae collected from different hosts, we provide the first report for the species A. rotundatum parasitizing Rhinella major, U. superciliosus, Leptophis ahaetulla, Chironius multiventris, and Mastigodryas boddaerti, as well as of A. humerale parasitizing U. superciliosus, A. geayi parasitizing Choloepus didactylus, and Rhipicephalus (B.) microplus parasitizing Alouatta puruensis.
Neospora caninum is an obligate intracellular protozoan with canids (Canis domesticus, Canis lupus dingo, Canis latrans, Canis lupus) as its definitive hosts. The objective of this study was to detect anti-N. caninum antibodies in pregnant women seen at referral center for prenatal screening in the state of state Mato Grosso do Sul, Brazil. A total of 188 serum samples from pregnant women provided by the Instituto de Pesquisa, Ensino e Diagnósticos da APAE de Campo Grande (IPED/ APAE) were subjected to IFA test and western blot analysis. The samples were divided into three groups: 23/99 samples from the seropositive group for toxoplasmosis were positive for anti-N. caninum IgG antibodies, and 9/99 positive for IgM; in the HIV group, 7/33 were positive for IgG; and in the HIV+toxoplasmosis group, 13/56 were positive for IgG and two positive for IgM. The seropositivity for IgG was assessed by western blot by testing 43 IFA test positive samples using rNcSRS2 (Nc-p43) as antigen. The serological results of the present study suggest that exposure of these pregnant women to the parasite N. caninum and presence of IgM antibodies are indicative of recent infection. Further studies are needed to establish the possibility of active infection.
More than 70 tick species are found in Brazil, distributed over five genera and including main vectors of infectious disease agents affecting both animals and humans. The genus Amblyomma is the most relevant for public health in Brazil, wherein Amblyomma aureolatum, Amblyomma ovale and Amblyomma sculptum have been incriminated as vectors of Rickettsia and Borrelia pathogens. The objective of this study was to investigate the presence of Rickettsia spp. and Borrelia spp. in ticks in the Brazilian mid-western savannah. DNA extraction, PCR for Borrelia spp. (flgE gene) and Rickettsia spp. (ompA and gltA genes) and subsequent sequencing were performed. A total of 1875 ticks were collected and identified as A. sculptum except for two Amblyomma coelebs ticks. Molecular evidence for Borrelia spp. and Rickettsia parkeri was found in A. sculptum. This is the first molecular evidence for R. parkeri in A. sculptum ticks in the Midwest region and Borrelia spp. circulating in a tick of the Amblyomma genus in Brazil.
The objective of this work was to evaluate the efficacy of two cypermethrin-and chlorpyrifos-based acaricides in controlling Rhipicephalus microplus in a naturally infested bovine herd and in in vitro tests, as well as to monitor the animals for tick fever. Male bovines in the rearing phase were used, with 30 Brangus and 30 Nellore animals naturally infested. The groups were composed as follows: 15 Nellore treated, 15 Nellore control, 15 Brangus treated and 15 Brangus control. Every 18 days, the animals were monitored for tick count, acaricide treatment, weight, blood pack cell volume, and clinical signs. For in vitro tests, the larval packet test, adult immersion test and DNA amplification for tick fever diagnosis were performed. In the first animal treatment period, product 1 (cypermethrin, 15 g + chlorpyrifos, 25 g + citronellal, 1 g) was used; in the second period, product 2 (cypermethrin, 15 g + chlorpyrifos, 30 g + fenthion, 15 g) was used. In Brangus animals, the mean efficacy was 35.1% and 95.8% in the first and second periods, respectively, for the same animals. For Nellore animals, the efficacy in periods one and two was 51% and 97.1%, respectively. The in vitro results showed efficacy above 95% for the two challenged acaricides. The Brangus animals showed a high production of ticks associated with the presence of tick fever agents, which could generate risks for the disease's enzootic stability
Neonatal sepsis is a clinical syndrome defined by systemic signs of infection in newborns accompanied by bacteremia. Can be responsible for serious consequences for the newborn child, characterized at the birth as early sepsis or late onset sepsis, with high rate of neonatal morbidity and mortality. Pathological agents such as Escherichia coli (E. coli), Streptococcus agalactiae (S. agalactiae), Ureaplasma urealyticum and Mycoplasma hominis are most often responsible for intrauterine infections. The objective of this study is to evaluate the factors of neonatal sepsis predisposition in pregnant women through histopathological examination and the apoptotic index of placental tissues and detect DNA of E. coli and S. agalactiae using the Polymerase Chain Reaction (PCR). Histopathological analyses were made and the apoptotic index was determined to verify the levels of possible inflammatory infiltrates and cell death. Placenta samples were collected from November 2013 to May 2014. After DNA extraction, a PCR was performed amplifying the target fragment from the conserved regions of the rpoB (beta-RNA polymerase) polymorphism of E. coli and the factor 1 of S. agalactiae. The apoptosis index was tested with Acridine Orange and the histological procedure with Hematoxylin-Eosin staining. Among 100 samples of placental tissues analyzed by PCR, 48 represented the control group and did not present a risk factor associated with neonatal sepsis, and 52 samples representing the study group had at least one risk factor. Among these 52 samples, 7 (13.4%) had a PCR positive for E. coli. No placenta samples showed a positive PCR for S. agalactiae. The quantification of the apoptotic index did not show statistical significances between the groups and no inflammatory infiltrates were observed. However, histological sections showed fibrinoid necrosis, infarct areas and areas of calcification in all samples. Therefore, the results allow to conclude that the seven patients of experimental group with positive PCR for E. coli had eminent risk factors of neonatal sepsis, and the infection of the urinary tract (UTI) is the main aggravating circumstance. The histopathological examination demonstrated that the risk factors caused significant alterations, producing fibrinoid necrosis and infarcted areas in the placenta, contrary to apoptotic index that didn't differ from the group with unprecedented risk.
