The dependence on pH of the disproportionation of the flavosemiquinone has been investigated by ESR. For 3-alkylated lumiflavin [R(3) in aqueous solution p i (for the dissociation of the neutral flavosemiquinone) is 8.4; PKred is 6.3. Hydrolytic ring cleavage in position 2, 8,8'-dimerization, and 10-dealkylation interfere with the formation of the flavosemiquinone anion. However, in non-aqueous dimethylformamide and with stoichiometric amounts of strong base ( t -B u O K ) , 100 Olio flavosemiquinone anion is formed without interfering side reactions. Its light absorption spectrum is practically identical to that of the "red flavoprotein radical".From the ESR spectra the individual isotropic hyperfine coupling constants have been determined.Since the discovery of flavin radicals by Michaeljs and co-workers [1,2] there has been doubt as to the correlation of visible absorption spectra and protonation state of the flavosemiquinone. Beinert [3] was the first to interpret the light absorption spectra of the half-reduced flavin system as a function of pH. I n a previous paper [4] we have made further correlations between structure, light absorption and other physical properties of flavin species.Within the range of these previous investigations two essential aspects remained : (a) determination of the light absorption of the flavosemiquinone anion, and (b) investigation of the tautomer structure of the neutral flavosemiquinone that absorbs in the range of 560-600 nm, as revealed by Beinert [3].The biochemical importance of these two points is stressed by Massey and Palmer [5] who found two classes of radical intermediates at half reduction of flavoproteins. The first type of radical, which shows red colour, has been found e.g. in D-amino acidoxidase, glucose oxidase a t high pH, oxynitrilase, and L-amino acid oxidase. The other type, the blue flavin radical, was found in the Azotobacter vinelandii flavoprotein, glucose oxidase a t low pH, and thioredoxin reductase. To be sure, these radicals, which are quantitatively verified by electron spin resonance (ESR), must not be confused with other coloured intermediates that have Non-Standard Abbreviations. An attempt is made to create a basis for the understanding offlavin-(Vitamin B2-) dependent oxidations in terms of molecular structure. The main points of this survey can be summarized as follows: a) Theflavins must be differentiated according to their redox states (quinone state, radical state, hydroquinone state), since configuration, energy content of the tautomeric forms, metalaffinity etc., alter drastically during reduction. -6 ) The methylgroup on C-8 of the flavoquinone is reactive. -cj The electron transfer properties of the flavin system frequently depend on the coupling of secondary redox systems to the apoprotein. This interaction thermodynamically stabilizes the half-reduced state of the flavin. -d) Numerous novel ffavin types, among them stabilized free radicals, have become availabIe through syntheses at the dihydro stage. -ej Reaction of the flavin radical with 0 2 is slow as long as no disproportionation occurs, i. e. the fast flavin autoxidation proceeds via the dihydro state.f l The flavin nucleus gains affinity for heavy metal ions only by the acceptance of one, and only one, electron. -g) Redox activity and contact with the ffavin coenzyme have been confirmed for Fe and Mo, which occur in flavoproteins. The properties of the metal coordination sphere are determined primarily by sulfur-containing groups of the apoprotein.
Von den Naturst'offen, die Strukturen aufweisen, von denen die Fahigkeit zur komplexen Bindung von Netallionen bekannt ist, ist das Riboflavin, das die fiir das Oxin charaktsristische Chelatgruppe besitzt, von besonderem Interessel) 2 ) . Es darf nach H . R. Mahler e,t al. 3, vermutet werden, dass beim R,iboflavin die biologische Wirkung in Flavoproteinen mit der Ausbildung eines solchen Metdl-Komplexes verbunden ist, indem hierbei Sgsteme mit spezifischem Redox-Potential entstehen.Von den bisher bekanntgewordenen, die Wirkung des Riboflavins auf das Wachstum von Mikroorganis~nen~) und Zellgewebe5) hemmenden Verbindungen sei neben dem Lumiflavin 6, das von WooZZey7) synthetisierte 7,8-Dimethyl-l0-ribity1-2,4-diaminophenazin erwiihnt,.Lumiflavin (6,7,9-Trimethylisoalloxazin) (XII) entsteht beim photolytischen Abbau des Riboflavinss). Seine Loslichkeitseigenschaften unterscheiden sich nur relativ wenig von denen ties Ri'boflavins.Im folgenden berichten wir uber Synthesen von Verbindungen, die als Strukturanaloge des Lumiflavins und damit des Isoalloxazin-Syst'enis im Riboflavin aufgefasst werden konnen, wobei darauf geachtet wurde, dass diese Verbindungen in den folgenden Eigenschaften mit der heterozyklischen Kornponente im Riboflavin ubereinstimmen: Eine dem Isoalloxazin verwandte Mesomerie, intaktes Redoxsystem, intakte Chelatgruppe, ahnliche Molekeldimension, Bhnliche Liislichkeitsvorhaltnisse.Fur Flavinsynthesen sind drei Wege bekannt : Kondensation von primiir-sekundaren aromatischen o-Diaminen mit Alloxan unter Rorsaurezusatz9) oder mit 5-Halogenbarbitursauren in PyridinlO),
Es wird versucht, aus neueren chemischen und biochemischen Daten eine Basis fur das Verstandnis Flavin-( Vitamin-B2-)abhangiger Oxydation aufzuzeigen. Diese Ubersicht mochte folgendes herausstellen: Die chemischen Eigenschaften der Flavine miissen nach Redoxstufen (Chinonstuje, Radikalstufe, Hydrochinonstuje) difSerenzievt werden, denn bei der Reduktion andern sich Planaritat, Energieinhalt der Tautomerjormen, Metall-Affinitat usw. drastisch. -Die Methylgruppe an C-8 der Flavochinone ist reaktionsfahig. -Die Elektronentransfer-Eigenschaften des Flavin-Systems hangen oft von der Kopplung an sekundare Redox-Systeme im Apoprotein ab. Diese Wechselwirkung Jtabilisiert die halbreduzierte Stufe des Flavins thermodynamisch. -Synthesen in der Dihydrostufe machen zahlreiche neue Flavin-Typen, u. a. stabilisierte jreie Radikale, zuganglich. -Das Flavinradikal reagiert nur Iangsam mit 0 2 , solange es nicht disproportionieren kann, d. h. die Jchnelle Flavin-Autoxydation verlauft iiber die Dihydrostufe. -Der Flavinkern gewinnt erst durch Aufnahme eines und nur eines Elektrons Affinitat zu Schwermetallionen. -Redox-Aktivitat und Kontakt mit dem Flavocoenzym sind fur die in Fiavoproteinen vorkommenden Metalle Fe und Mo sichergestellt. Die Eigenschaften der Metall-Koordinationssphare werden in erster Linie durch schwefelhaltige Gruppen des Apoproteins bestimmt I. Einleitung 1. Die saure Hydrolyse laBt daher den Polyhydroxy-Substituenten am Flavin-Kern intakt und fuhrt zum Riboflavin. Dieses alteste bekannte Flavin-Derivat ( Warburg und Christian 1932) [3] ging zunachst als Lactoflavin (Vitamin B2) in die Literatur ein. ~ [2] P .
