Kantitatif RT-PCR (RT-qPCR) ile MikroRNA (miRNA) Ekspresyon Profillemesi Özet: Gen ekspresyonunun post-transkripsiyonel düzenleyicisi olarak bilinen miRNA'lar ökaryotik canlılarda çeşitli fizyolojik ve patolojik süreçlerde rol oynamaktadır. miRNA'ların rollerinin ortaya konulması ile birlikte miRNA'lar üzerine yapılan çalışmaların sayısı da gün geçtikçe artmaktadır. Yapılan çalışmalar ile birlikte miRNA'ların bütün hücre ve doku tiplerinde eksprese olduğu ortaya konmuştur. miRNA ekspresyon profilinin ortaya konması için çeşitli yöntemler önerilmesine rağmen, yüksek duyarlılığı ve özgüllüğü nedeniyle RT-qPCR altın standart olarak kabul edilmektedir. Bu derlemede; RT-qPCR ile miRNA ekspresyon profillemesi sürecindeki adımlar detaylı olarak özetlenmiş ve konu ile ilgili literatür bilgisine yer verilmiştir. Anahtar kelimeler: Ekspresyon, miRNA, normalizasyon , RT-qPCR MicroRNA (miRNA) Expression Profiling by Quantitative RT-PCR (RT-qPCR) Summary: miRNAs, known as post-transcriptional regulators of gene expression, play a crucial role in various physiological and pathological processes in eukaryotic organisms. As the roles of miRNAs have been revealed, the number of studies on miRNAs is increasing day by day. Studies have shown that miRNAs are expressed in all cell and tissue types. Although various methods have been proposed to demonstrate miRNA expression profile, RT-qPCR is considered as a gold standard technique due to its high sensitivity and specificity. In this review; the steps in the miRNA expression profiling process by RT-qPCR were summarized in detail and relevant literature knowledge was given.
Özet: Gen ekspresyon çalışmalarında farklı deneysel veya klinik koşullar altında genlerin ekspresyon seviyelerinin karşılaştırılması için kullanılan en yaygın teknik Kantitatif Gerçek Zamanlı Polimeraz Zincir Reaksiyonu (RT-qPCR)'dur. RT-qPCR'da güvenilir sonuçlar elde etmek için verilerin housekeeping genler (HKG) ile normalizasyonunun yapılması gerekmektedir. Bununla birlikte; çalışma tasarımına uygun en stabil HKG'nin kullanılması, çalışma sonuçlarının güvenilirliğini artırmaktadır. Güvenilir veri elde etmek için çalışma tasarımına uygun stabil HKG'nin seçimi ve validasyonu önemli bir adımdır ki bu ekspresyon bazlı çalışmalarda bütünleyici bir adım olmuştur. HKG seçimi için farklı yazılım programları geliştirilmiştir. Normalizasyon için seçilecek en stabil HKG'nin seçilmeden önce deneysel çalışmalar ile dikkatli bir şekilde değerlendirmesinin yapılması gerekmektedir. Bu derlemede; Kantitatif Gerçek Zamanlı PCR Deneylerinin Yayınlanması için Asgari Bilgi (MIQE), HKG'lerin genel özellikleri, RT-qPCR çalışmalarında kullanımları ile çalışma dizaynına göre en uygun ve stabil olan HKG'lerin seçimi ve validasyonu hakkında bilgiler özetlenmiştir. Anahtar kelimeler: Gen ekspresyonu, housekeeping gen, kantitatif RT-PCR, referans gen Selection and Validation of Suitable Housekeeping Genes (HKGs) in Quantitative RT-PCR (RT-qPCR) Studies Summary: The quantitative Real-time Polymerase Chain Reaction (RT-qPCR) is the most common technique used to compare the expression levels of genes under different experimental or clinical conditions. Data should be normalized with housekeeping genes (HKGs) to obtain reliable results in RT-qPCR. Likewise, the use the most stable and suitable HKG for the study design improves the reliability of the study results. The selection and validation of a stable HKG according to the study design to obtain reliable data is an essential and integral step in expression-based studies. The several software programs have been developed for HKG selection. The most stable HKG to be selected for normalization needs to be carefully evaluated by experimental studies before selection. In this review, summarized Minimum Information for Publication of Quantitative Real-Time PCR Experiments (MIQE), characteristics of HKGs, selection and validation of most suitable and stable HKGs according to study design and, usage in RT-qPCR.
The aim of this study was to investigate the effects of using presynch-10 and ovsynch synchronization protocols on endometrial expression and serum levels of insulin-like growth factor-1 (IGF-1) that thought to play roles during endometrial healing process in cows diagnosed with repeat breeder syndrome (RBS) and subclinical endometritis (SE). A total of 40 Holstein-Friesian cows identified as having RBS and SE were used. The animals in Group 1 (G1, n=20) were applied presynch-10 and ovsynch furthermore, biopsy and blood samples were collected both before and after administration. Biopsy and blood samples were taken from the animals in Group 2 (G2, n=20) without performing any synchronization. The IGF-1 gene expression profiles were determined in all samples using RT-qPCR. Even though the level of IGF-1 expression increased in samples taken after synchronization in G1, it was not statistically significant (P>0.05). There was no statistical difference in serum IGF-1 levels between G1 and G2 (P>0.05). In conclusion, the expression of the IGF-1 gene in the endometrial tissue of RBS cows with SE was identified, but the evidence suggests that the synchronization protocols in question had no significant effect on the gene expression profile being investigated.
