Localization in vacuoles, the lysosome-like organelle of yeast, was checked for several newly detected proteolytic enzymes. While arninopeptidase Co and carboxypeptidase S were found in vacuoles, proteinase D and proteinase E as well as a variety of other proteolytic activities detectable with the aid of chromogenic peptide substrates do not reside in this cell compartment.
Yeast Mutant Compartmentation
Toxicity correlated patterns like feeding behaviour, temperature and resulting feeding rate (pellets/h) were examinated. UV‐ and Cobalt‐irradiated Bacillus thuringiensis var. kurstaki preparations were found to be significantly toxic. Compared to active toxin with living spores an inactivated sporal preparation is less active, but not ineffective: LT50 26–60 h (UV‐inactivated, still some living spores) and 43–58 h (gamma‐inactivated, no active spores). Simultaneously it can be demonstrated that the reduction of feeding rate in test is still comparable to material with living spores.
Zusammenfassung
Über Einflußfaktoren der Toxizität von Bacillus thuringiensis gegenüber dem Einbindigen Traubenwickler, Eupoecilia ambiguella Hb. (Lep., Cochylidae)
Es wurde der Einfluß verschiedener Faktoren, wie Temperatur, Fraßverhalten und Fraßrate, auf die Toxizität von Bacillus thuringiensis gegenüber dem Einbindigen Traubenwickler untersucht. Das Optimum der Fraßrate wird unter B. t.‐Einfluß von 20 °C auf höhere Temperaturen verschoben. Ein Temperatur‐Anstieg um 3 °C verkürzt die LT50 um den Faktor 1,4. Eine Fraßzeit von nur 1 Minute ist ausreichend für die Abtötung; je länger jedoch die Fraßdauer, umso kürzer ist die LT50. Durch UV oder Gamma‐Strahlen behandelte Sporen ergeben bei verlängerter LT50 eine ausreichende Abtötung der Traubenwickler‐Raupen, womit die Wasserschutzgebietsauflage für solchermaßen behandelte B. t.‐Präparate im Weinbau entfallen könnte.
