O trigo semeado na região meridional do Brasil tem apresentado sérios problemas de qualidade da semente, sendo que na literatura não são encontrados, ainda, procedimentos padronizados para a determinação do vigor de sementes de trigo. O trabalho foi realizado com objetivo de estudar a metodologia do teste de envelhecimento acelerado para identificar níveis de vigor de lotes de sementes de trigo. Foram utilizadas sementes das cultivares CD 104 e BRS 220, representadas por cinco lotes cada. As seguintes determinações foram efetuadas: grau de umidade (estufa, 105±3 ºC, 24 h); teste de germinação (8x50 sementes, 20 ºC); emergência de plântulas em campo; envelhecimento acelerado (41 ºC / 48, 60 e 72 horas; e 43 ºC / 48 horas). A comparação de médias foi realizada pelo teste de Tukey, a 5% de probabilidade; para o estudo sobre a metodologia do teste de envelhecimento acelerado utilizou-se o coeficiente de correlação de postos de Spearman, onde se buscou obter uma medida de intensidade da associação entre os dados do teste de vigor com os dados obtidos na emergência de plântulas em campo. Pelos resultados obtidos conclui-se que o teste de envelhecimento acelerado utilizando a temperatura de 43 ºC, por 48 h fornece informações consistentes que permitem diferenciar lotes de sementes de trigo.
-The tetrazolium test is used to control seed quality of various plant species since it allows a rapid evaluation of viability. Freshly harvested barley seeds show dormancy that can make the germination test ineffective for an immediate evaluation. Therefore, the development of more efficient methods, such as the tetrazolium test, is necessary. The objective of this research work was to study various procedures for performing the tetrazolium test on barley seeds. Five lots of cv. BRS 195 barley seeds were used and subjected to the following treatments: two different methods of seed preconditioning (direct immersion in H 2 O and between sheets of moistened paper towels); two types of preparation for staining (longitudinal cross-section of the seed through the embryo with immersion of one half in a 2,3,5 triphenyl tetrazolium chloride solution or placing both halves on top of filter paper moistened with the tetrazolium salt solution); two methods of staining (on top of filter paper and direct immersion in the tetrazolium salt solution). Three concentrations of the tetrazolium salt solution (0.1%, 0.5%, and 1.0%) were used. It was concluded that the tetrazolium test on barley seeds may be accomplished with preconditioning by direct immersion in H 2 O and staining by immersing in a 0.1% or 0.5% concentration of tetrazolium salt solution or staining on top of filter paper moistened with such solution at a 1.0% concentration.Index terms: Hordeum vulgare, germination, dormancy.Viabilidade de sementes de cevada pelo teste de tetrazólio RESUMO -O teste de tetrazólio é empregado no controle da qualidade de sementes de várias espécies, pois permite estimar rapidamente a sua viabilidade. Sementes de cevada apresentam dormência quando recém-colhidas, o que pode tornar o teste de germinação ineficaz para avaliação imediata, sendo necessário o desenvolvimento de métodos mais eficientes como o de tetrazólio. O objetivo desse trabalho foi estudar vários procedimentos para a condução do teste de tetrazólio em sementes de cevada. Foram utilizados cinco lotes de sementes da cv. BRS 195, testando duas formas de pré-condicionamento da semente (imersão direta em água e entre folhas de papel toalha umedecidas); dois tipos de preparo para coloração (corte longitudinal da semente através do embrião com imersão de uma das metades em solução de cloreto de 2,3,5 trifenil tetrazólio ou com colocação das duas metades sobre papel de filtro umedecido com a solução do sal de tetrazólio); duas formas de coloração (sobre papel de filtro e com imersão direta na solução) e três concentrações de solução do sal de tetrazólio (0,1%, 0,5% e 1,0%). Foi concluído que o teste de tetrazólio em sementes de cevada pode ser conduzido mediante pré-umedecimento por imersão direta em água, coloração por imersão na solução de sal de tetrazólio nas concentrações de 0,1% e 0,5%, ou coloração sobre papel de filtro umedecido com essa solução, na concentração de 1,0%.Termos para indexação: Hordeum vulgare, germinação, dormência.
-Freshly harvested triticale seeds are usually dormant, making the immediate evaluation of the physiological potential of seed lots difficult. We evaluated different triphenyl tetrazolium chloride (TTC) test methods for rapidly determining the viability of four seed lots of x.Triticosecale Wittmack cultivar IPR111. The test variables were: Preconditioning, (i) placing whole seeds between moistened paper towels or (ii) directly soaking the seeds in water, both procedures being conducted at 20 °C for 18 hours; Post-conditioning seed preparation, (i) longitudinal bisection of the seed through the embryo with one half being stained and the other discarded or (ii) longitudinal bisection with both halves being stained; Staining for three and four hours, in the dark, with 0.1%, 0.5% or 1.0% (w/v) TTC according to the preconditioning method described above, (i) both halves of each seed were placed on filter paper moistened with TTC and maintained at 40 °C or (ii) one half of each seed was immersed in 5 mL of TTC solution in a 100 mL glass beaker at 30 °C. -The assessment of the germination test in wheat seeds varies from 4 to 15 days, because the species normally presents dormancy in freshly harvested seeds. The tetrazolium test can characterize seed viability in less than 24 hours including lots with dormancy seeds. The objective of this study was to develop a practical and efficient procedure for evaluating the viability of wheat seeds using the tetrazolium test. Five seed lots of the BRS 208 cultivar were used, where the following were tested: a) pre-conditioning between moist paper towels or direct immersion in water for 18 hours, at 20 °C; b) longitudinal section of the embryo and the endosperm; c) coloration on paper or by immersion for 2 and 3 hours, at 30 and 40 °C; and d) concentrations of tetrazolium solution at 0.075%, 0.1%, 0.5% and 1.0%. The tetrazolium test may be efficiently used to evaluate wheat seed viability by pre-conditioning the seeds between paper towels (18 hours, at 20 °C) and adopting the following combinations of preparation and coloration: coloration of both halves of the seed on paper (2 hours, at 30 °C), in a 1.0% tetrazolium solution; or coloration of one half of the seed by immersion (3 hours, at 30 °C), in a 0.1% tetrazolium solution; or coloration of one half of the seed by immersion (2 hours, at 40 °C), in a 0.075% tetrazolium solution. O trabalho objetivou propor procedimento prático e eficiente para avaliação da viabilidade de sementes de trigo pelo teste de tetrazólio. Utilizaram-se cinco lotes de sementes da cultivar BRS 208, testando-se: a) pré-condicionamento entre papel toalha umedecido ou por imersão direta em água, por 18 horas, a 20 °C; b) corte em bissecção longitudinal ao longo do embrião e do endosperma; c) coloração sobre papel ou por imersão, durante 2 e 3 horas, a 30 e 40 °C; e d) concentrações de solução de tetrazólio a 0,075%, 0,1%, 0,5% e 1,0%. RESUMO -As espéciesPiper hispidinervum e Piper aduncum destacam-se como produtoras de óleos essenciais ricos em compostos de interesse às indústrias de perfumaria e produtos fitossanitários, como o safrol e dilapiol. Essas espécies encontram-se em processo de domesticação e estudos sobre o processo germinativo de suas sementes são escassos. Assim, objetivou-se com o presente trabalho avaliar a viabilidade de sementes de P. hispidinervum e P. aduncum, buscando determinar um protocolo eficiente para a realização do teste de germinação para cada espécie estudada. O trabalho foi conduzido no período de julho a agosto de 2008, sendo as sementes submetidas a diferentes combinações de temperatura e de luz: 20 °C e 25 °C sob luz branca e difusa; e 20-30 °C alternando escuro (16 h) e claro (8 h), totalizando cinco tratamentos. O tratamento utilizando a temperatura de 25 °C com incidência de luz branca foi o que produziu melhores resultados, com maiores valores de porcentagem e velocidade de germinação sendo, portanto, indicado para avaliação da qualidade fisiológica das sementes de pimenta longa e pimenta-de-macaco.Termos para indexação: Piperácea, luz, temperatura, qualidade fisiológica. LIGHT AND TEMPERATURE IN THE SEED GERMINATION OF LONG PEPPER (Piper hispidinervum) AND MONKEY PEPPER (Piper aduncum)ABSTRACT -The species Piper hispidinervum and Piper aduncum stand out as producers of essential oils, rich in compounds, such as saffrol and dillapiol, which are of interest to the fragrance and pesticide industries. These species are being domesticated and studies about their seed germination process are scarce and the aim of this study was to develop an efficient protocol for germination tests with which to evaluate seed viability in P. hispidinervum and P. aduncum. The study was carried out in July and August, 2008, when the seeds were submitted to different combinations of temperature and light: 20 °C and 25 °C with diffuse, white light; and 20-30 °C with alternating dark (16 h) and light (8 h) periods, totaling 5 treatments. The treatment with 25 °C and white light gave the best results in terms of higher percentages and germination rates, and therefore, these conditions are recommended for evaluating seed physiological quality of long pepper and monkey pepper.
