Aim of the research: to identify changes in the value of indicators of the state of the prooxidant-antioxidant system (PAS) in the tissues of edible parts of agricultural plants under the influence of temperature changes. Methodology: Quantitative determination of indicators of the state of PAS was performed on tissue samples of edible parts of the following plants: Solánum lycopérsicum L., Сucumis sativus L., Capsicum annuum L., Solanum melongena L., Solanum tuberosum L., Allium sativum L., Allium cepa L., Daucus carota L., Beta vulgaris L., Cucurbita pepo var. Giraumontia L. The concentration of superoxide anion radical (•O2-), TBA-active products, superoxide dismutase (SOD) activity, catalase, the concentration of ascorbic acid (AA), glutathione (GSH) were determined. The results of the research show that hypothermia activates both parts of the PAS, however, cooling is accompanied by more powerful both low molecular weight and enzymatic antioxidant (AO) protection. The research of AO can be divided according to the degree of reduction of the protective value in hypothermia in the following series: SOD, catalase, GSH, AA. The most resistant in terms of changes in PAS to hypothermia is Solanum tuberosum L., Allium sativum L., Beta vulgaris L.; the least resistant is Capsicum annuum L., and Solánum lycopérsicum L. The generative organs of plants are less resistant to hypothermia than the vegetative ones. Practical consequences. As a result of the conducted biochemical analysis, it is established which method of storage of plant products is more effective in terms of preservation of AO activity: cooling or freezing.
Reactive Oxygenspecies (ROS) cause free radical peroxidation of macromolecules (FRPO). The formation of malonic dialdehyde (MDA) is the marker of FRPO, and the consequences of changes in prooxidant-antioxidant system (PAS) are evidenced by changes in the activity of the enzyme cytochrome oxidase. The study of the role of ROS in anti-infective protection of animals, oxidative explosion processes, mechanisms of aging and apoptosis has opened up the prospects for the search for analogues in the plant world. Quantitative determination of PO and FRPO products was carried out on grains of Zea mays L. taken from plants of the following cultivars: "DK Burshtyn" (high disease-resistant cultivar), "DN Demeter" (medium disease-resistant cultivar) and "DK Veles" (low resistance). Evaluation of the level and sources of ROS generation was performed by spectrophotometric HBT test. The level of FRPO was assessed by the concentration of background and stimulated malonic dialdehyde (MDA). To assess the effects of PAS changes, the cytochrome oxidase activity was determined. As a result of the conducted researches, it was established that the tissues of grains of highly resistant variety Zea mays L. "DK Burshtyn" have the lowest background level and the highest level of stimulated by yeast and NaF generation of •O2-, which indicates the presence of a powerful activating ability, and especially calcium and NADPH oxidase signalling systems. It was found that with increasing disease resistance of Zea mays L. there is a decrease in the background and stimulated levels of MDA, which indicates a low degree of FRPO membrane lipids and may be due to increased AO of PAS. It is proved that during the transition from low-resistant to disease "DK Veles" cultivar to medium-resistant "DN Demeter" and high-resistant "DK Burshtyn" cultivar, the cytochrome oxidase activity increases, which may be explained by a decrease in the intensity of peroxide degradation of mitochondrial membranes as a result of the increased AOD. It is concluded that the prooxidant link of PAS is involved in maintaining the resistance of plant cultivars to disease, but requires a powerful compensatory antioxidant mechanism to protect against peroxide destruction of macromolecules.Keywords: prooxidants, superoxide, malonic dialdehyde, reactive oxygen species, free radical peroxidation, cytochrome oxidase. Активні форми Оксигену (АФО) спричинюють вільнорадикальне перекисне окиснення макромолекул (ВРПО). Маркером ВРПО є утворення малонового діальдегіду (МДА), а про наслідки зміни прооксидантно-атиоксидантного балансу свідчить зміна активності ферменту цитохромоксидази. Дослідження ролі АФО у протиінфекційному захисті тварин, процесах окисного вибуху, механізмах старіння та апоптозу відкрило перспективи пошуку аналогів у рослинному світі.Кількісне визначення ПО та продуктів ВРПО здійснювали на зернівках Zea mays L. взятих від рослин наступних сортів: «ДК Бурштин» (високостійкий до хвороб сорт), «ДН Деметра» (середньостійкий до хвороб сорт) та «ДК Велес» (малостійкий). Оцінку рівня та джерел генерації АФОздійснювали за спектрофотометричниим НСТ-тестом.Оцінку рівня ВРПО здійснювали за концентрацією фонового та стимульованого малонового діальдегіду(МДА).Для здійснення оцінки наслідків зміни ПАС проводили визначення активності цитохромоксидази. В результаті проведених досліджень встановлено, що тканини зернівок високостійкого сорту Zea mays L. «ДК Бурштин» мають найнижчий фоновий рівень та найвищийрівень стимульованої дріжджами та NaF генерації •О2-, що свідчить про наявність потужної активуючої здатності каскаду передачі сигналу для збудження експресії захисних генів, а особливо кальцієвої та НАДФН оксидазної сигнальної систем. Виявлено, що зі збільщенням стійкості сорту Zea mays L. до хвороб спостерігається зниження фонового та стимульованого рівня МДА, що свідчить про низький ступінь ВРПО ліпідів мембран та можливо пояснюється посиленою АО ланки ПАС. Доведено, що при переході від малостійкого до хвороб сорту «ДК Велес» до середньостійкого «ДН Деметра» та високостійкого сорту «ДК Бурштин» активність цитохромоксидази зростає,що, можливо, пояснюється зниженням інтенсивності перекисної деструкції мембран мітохондрій в результаті посилення АОЗ. Здійснено висновок про те, що прооксидантна ланка ПАС бере участь у підтримці стійкості сорту рослин до хвороб, однак потребує потужного компенсаторного антиоксидантного механізму для захисту від перекисної деструкції макромолекул.Ключові слова: прооксиданти, супероксид, малоновий діальдегід, активні форми Оксигену, вільнорадикальне перекисне окиснення, цитохромоксидаза.
