Merck, Darmstadt.]Die ausgezeichneten Untersuch ungen von R u zi c k a und Mitarbeitern haben nicht nur zu einer Synthese des Androsterons (VII) gefiihrt, sie haben dariiber hinaus einen vollstandigen Einblick in die Konstitution und den sterischen Aufbau dieses wichtigen Naturstoffes gegeben. Die von Ruzickal) durchgefiihrte Synthese geht vom Cholesterin aus, das nach Hydrierung und Umlagerung der Hydroxylgruppe durch energische Oxydation seiner Seitenkette beraubt wird. Es erschien uns von Interesse zu untersuchen, ob sich diese Entfernung der Seitenkette auch auf systematischem W ege erreichen l a t . Als Ausgangsmaterial fur d e r d g e Versuche kommen native Gallensauren nicht in Frage, da diese, wie bereits Ruzicka2) zeigen konnte, sterisch dem Epi-koprosterin-Typ zuzuordnen sind. Der raumlichen Anordnung des Androsterons entspricht jedoch die 3-Oxy-allocholansaure (I), die nach den Arbeiten von Windauss) und Wieland4) zugiinglich ist. Wir haben uns eine groRere Menge dieses Materials bereitet und die Verkiirzung der Seitenkette in den ersten Stufen nach der bekannten Methode von Wielands) durchgefiihrt. Der von Wieland und Mitarbeitern an der Cholansaure vorgenommene Grign a r d-Abbau lieferte, wenigstens in den letztenstufen, geringe Ausbeuten. Die aertragung einer derartigen Reaktionsfolge auf eine hydroxyl-haltige Saure erschien zunachst wegen der mit der Einfiihrung einer Oxygruppe verbundenen Gefahr einer Ringsprengung bei oxydativen Eingriffen wenig verlockend. Es zeigte sich jedoch, d& sich nach Blockierung der Hydroxylgruppe durch den Acetylrest trotzdem leidliche Ergebnisse erhalten lassen. Wir gelangten so vom Methylester der 3-Oxya 11 o c h ol ans a ur e durch Umsetzung mit P h enyl-magnesium b r o mi d iiber das Diphenyl-carbinol I1 zur 3-Acetoxy-nor-allocholansaure (111). Von dieser Saure aus lie13 sich der gleiche Abbau zur 3-Acetoxyb i s n o r -a1 1 o c h o 1 an s a u r e (IV) glatt durchfiihren.Die bei dem weiteren, mit dem Verlust von 2 Kohlenstoffatomen verbundenen Abbau von Wieland, Schlichting und Jacobi6) und auch von Tscheschea) in der Cholansaure-bzw. Allo-cholansaure-Reihe gemachten Erfahrungen lieRen unsere Aussichten, in der Oxy-allocholansaure-Reihe die entsprechende Umwandlung zu erzielen, recht gering erscheinen. Erfreulicherweise gelang es jedoch nach einer Reihe von Versuchen, die 3-Acetoxya t i oa 11 o c h o 1 an s a u r e (V) bei der Oxydation des Diphenyl-carbinols aus dern 3-Acetoxy-bisnor-allocholansaure-methylester in einer Ausbeute von 20% d. Th. zu gewinnen. Zur ijberfiihrung der so erhaltenen 3-Oxy-atiol) Ruzicka, Goldberg, Meyer, Briingger, Eichenberger, Helv. chim. Acta z, Helv. chim.
Die antiskorbutische Wirksamkeit der Ascorbinsäure tierischer und verschiedener pflanzlicher Herkunft ist durch zahlreiche Untersuchungen absolut sichergestellt. Die Identität dieses Stoffes mit dem Vitamin C, die kaum mehr zweifelhaft war, ist abschließend dadurch bewiesen, daß auch die synthetisch hergestellte l-Ascorbinsäure nach Eeichstein und Mitarbeitern l ) die gleiche Wirksamkeit zeigt wie das natürliche Material. Diese Autoren haben außer der l-Ascorbinsäure die entsprechende bisher in der Natur nicht aufgefundene d-Form synthetisiert und biologisch prüfen lassen. Nach der vorliegenden kurzen Notiz hat diese Verbindung auch bei viermal größeren Dosen als bei der 1-Form noch keine deutliche antiskorbutische Wirkung erkennen lassen. Dieses Resultat, das noch durch die Prüfung in wesentlich höheren Dosen ergänzt werden soll, beleuchtet die Abhängigkeit der Vitamin C-Wirksamkeit von der Feinstruktur des Moleküls. Es liegt vom theoretischen und praktischen Standpunkt aus nahe, die Fragestellung dahin zu verallgemeinern, ob es noch andere, insbesondere der Ascorbinsäure chemisch nahestehende Verbindungen gibt, die antiskorbutische Wirksamkeit besitzen. Vor einigen Monaten hat Ohle 2 ) über die Synthese eines Stoffes von der gleichen Summenformel wie Ascorbinsäure -C 6 H 8 O e -kurz berichtet. Er erhielt durch alkalische Behandlung des Methylesters der d-Glucosonsäure unter Verseifung und gleichzeitiger Wasserabspaltung die genannte Verbindung. Sie zeigt die charakteristischen chemischen Keaktionen in gleicher Weise wie die Ascorbinsäure, ist aber nicht mit ihr identisch, was ohne weiteres aus den physikalischen Konstanten und z. B. aus dem Verhalten bei der Acetonierung hervorgeht. -Eine entsprechende Synthese wurde kürzlich von Maurer und Schiedt 8 ) ausführlicher beschrieben. Diese Autoren erhielten, gleichfalls ausgehend von dem Methylester der 2-Ketogluconsäure, durch Anwendung von Natriummethylat in Pyridin oder Methanol unter Verseifung und Wasserabspaltung einen Stoff der Zusammensetzung C e H 8 0 6 , der nach seinen Eigenschaften und seiner Herkunft identisch mit der von Ohle gewonnenen Verbindung sein dürfte.
