This study aimed to obtain a microbial active compound as a novel antimalarial drug from Indonesian isolates. Target-based assays were used to screen for antimalarial activity against the parasite mitochondrial, Plasmodium falciparum malate:quinone oxidoreductase (PfMQO) enzyme. In total, 1600 crude extracts, composed from 800 fungi and 800 actinomycetes extracts, were screened against PfMQO, yielding six active extracts as primary hits. After several stages of stability tests, one extract produced by Aspergillus sp. BioMCC f.T.8501 demonstrated stable PfMQO inhibitory activity. Several purification stages, including OCC, TLC, and HPLC, were performed to obtain bioactive compounds from this active extract. All purification steps were followed by an assay against PfMQO. We identified the active compound as nornidulin based on its LC-MS and UV spectrum data. Nornidulin inhibited PfMQO activity at IC50 of 51 µM and P. falciparum 3D7 proliferation in vitro at IC50 of 44.6 µM, however, it had no effect on the growth of several mammalian cells. In conclusion, we isolated nornidulin from Indonesian Aspergillus sp. BioMCC f.T.8501 as a novel inhibitor of PfMQO, which showed inhibitory activity against the proliferation of P. falciparum 3D7 in vitro.
Prosiding Use Cases Artificial Intelligence Indonesia adalah buku yang mengumpulkan hasil-hasil kajian dan liputan 26 use cases inovasi dan 4 inisiatif pemanfaatan kecerdasan artifisial yang kemudian dipetakan menjadi lima klaster bidang kecerdasan artifisial, yakni: riset industri dan hankam, layanan publik dan kesehatan, kota cerdas dan kebencanaan, ketahanan pangan dan maritim, serta klaster inisiatif pemanfaatan kecerdasan artifisial. Materi buku diperoleh dari para kontributor seluruh anggota quadhelix dan para narasumber pegiat kecerdasan artifisial di Indonesia. Buku ini akan membantu masyarakat dalam mendapatkan pengetahuan dan pencerahan tentang seluruh teknologi kecerdasan artifisial yang membantu sektor-sektor terkait dalam hal otomatisasi, alat bantu untuk menganalisis, membuat rekomendasi serta keputusan, memprediksi dan sebagainya.
Dalam rangka mengetahui potensi keragaman mikroba Indonesia sebagai sumber senyawa alami, Balai Bioteknologi-BRIN mengoleksikan mikroba dari berbagai lokasi di Indonesia. Selanjutnya, BRIN bekerja sama dengan mitra riset dari Indonesia dan Jepang dalam proyek Science and Technology Research Partnership for Sustainable Development Program (SATREPS) dalam fase I dari tahun 2015 sampai 2020 dan dalam fase II dari tahun 2021 hingga 2026 untuk penemuan senyawa obat dari mikroba. Alur pencarian senyawa obat dari mikroba dimulai dari kegiatan eksplorasi mikroba (isolasi dan identifikasi mikroba) dari berbagai jenis sampel alami (tanah, tumbuhan, serasah, hewan laut, dan lain-lain) melalui waktu yang cukup panjang, yakni antara 10–20 tahun. Salah satu tahap kritis dalam proses penemuan senyawa obat (drug discovery) dari mikroba adalah tahap identifikasi mikroba. Secara umum, proses identifikasi mikroba dilakukan secara molekuler. Namun, proses ini membutuhkan waktu dan biaya yang besar, terlebih bila menangani mikroba dalam jumlah yang banyak dan khususnya mikroba dari golongan fungi yang memiliki bentuk morfologi mikroba tertentu. Proses identifikasi berbasis morfologi dilakukan melalui pengamatan dengan mikroskop. Namun, identifikasi mikroba ini sangat tergantung dari pengalaman yang dimiliki personel pelaksana dan akan menyulitkan apabila mengidentifikasi mikroba yang baru. Kecerdasan artifisial (KA) dapat membantu mengidentifikasi mikroba berbasis morfologi. Dalam hal ini, KA membantu personel dalam menganalisis gambar morfologi suatu mikroba, sehingga proses ini dapat dilakukan secara cepat dan akurat. TKA juga diharapkan dapat membantu personel dalam menemukan mikroba baru yang belum pernah ditemukan sebelumnya, sehingga dapat meningkatkan nilai dari mikroba asli Indonesia tersebut.
