Abstract:In an effort to develop new antimicrobial agents, 3-(4-methylcoumarin-7-yloxyacetylamino)-2-thioxo-1,3-thiazolidin-4-one (4) was synthesized by reaction of thiocarbonylbisthioglycolic acid with ethyl (4-methyl-2-oxo-2H-chromen-7-yloxy)acetohydrazide (3), which was prepared in turn from 7-hydroxy-4-methylcoumarin (1). The condensation of compound 4 with different aromatic aldehydes afforded a series of 5-(arylidene)-3-(4-methylcoumarin-7-yloxyacetyl-amino)-2-thioxo-1,3-thiozolidin-4-one analogs 5a-h. The structures of these synthetic compounds were elucidated on the basis of IR, 1 H-NMR and 13 C-NMR spectral data and ESI-MS spectrometric analysis. Compounds 5a-h were examined for their antibacterial activity against several strains of Gram-positive and Gram-negative bacteria.
Kết quả nghiên cứu phổ hồng ngoại của DES điều chế cho thấy có sự hình thành tương tác liên kết hydrogen liên phân tử giữa L-proline và acid para-toluenesulfonic. Điều kiện thực hiện phản ứng tốt nhất thu được sau khi khảo sát: nhiệt độ phản ứng 80oC, thời gian phản ứng 120 phút, tỉ lệ benzaldehyde:o-phenylenediamine=2:1 (mmol) và lượng xúc tác sử dụng là 30% mol so với o-phenylenediamine. Xúc tác điều chế có thể sử dụng 4 lần mà không thay đổi hoạt tính đáng kể sau mỗi lần sử dụng. Các nhóm thế halogen làm giảm hiệu suất phản ứng trong khi những nhóm thế cho điện tử không ảnh hưởng đến hiệu suất phản ứng tổng hợp một số dẫn xuất benzimidazole. DES điều chế cũng cho thấy sự chọn lọc khi ưu đãi hình thành sản phẩm benzimidazole thế hai lần so với benzimidazole thế một lần.
The synthesis of three new 5-bromo-N’-(1-arylethylidene)-2-hydroxybenzohydrazide compounds was accomplished by the use of acid 2-hydroxybenzoic (acid salicylic) as the starting material. Refluxing the hydrazides with acetic anhydride gave three corresponding new compounds namely 2-(4-acetyl-5-aryl-5-methyl-4,5-dihydro-1,3,4-oxadiazol-2-yl)-4-bromophenyl acetates. The structures of the compounds were confirmed by their IR, 1H, 13C-NMR and HR-MS spectral data.
