ViÖn C«ng nghÖ sinh häcM«i tr−êng biÓn lµ mét nguån cung cÊp c¸c s ¶n phÈm tù nhiªn cã ho¹t tÝnh sinh häc lín vµ hiÕm trong ®ã nhiÒu s ¶n phÈm cã c¸c ®AEc tÝnh cÊu tróc mµ c¸c s ¶n phÈm tù nhiªn ë trªn c¹n kh«ng cã. Nghiªn cøu vÒ ®AEc ®iÓm d−îc häc cña c¸c s ¶n phÈm nµy còng ® kÐo theo c¸c ph¸t hiÖn vÒ nhiÒu t¸c nh©n cã ho¹t tÝnh tiÒm tµng cã thÓ øng dông trong y häc l©m sµng [13]. èc cèi (Conidae) lµ mét trong nh÷ng hä ®éng vËt th©n mÒm lín ë biÓn (®AEc biÖt vïng biÓn nhiÖt ®íi) víi kho ¶ng h¬n 700 loµi, cã näc ®éc, bao gåm c ¶ c¸c loµi ¨n thÞt ®éng vËt, ¨n c¸c loµi èc kh¸c, ¨n s©u vµ thËm chÝ c ¶ c¸ [12,13]. Chóng kh«ng chØ ®−îc biÕt ®Õn bëi vÎ ®Ñp cña vá (cã thÓ mua ®−îc ë nhiÒu cña hµng b¸n ®å l−u niÖm ë kh¾p n¬i trªn thÕ giíi) mµ chóng cßn ®−îc quan t©m bëi c¸c ®AEc tÝnh sinh häc cña c¸c chÊt cã trong näc ®éc cña chóng. ViÖc t¸ch chiÕt còng nh− lµm s¸ng tá cÊu tróc cña mçi thµnh phÇn tõ näc cña chóng ® ®−îc b¾t ®Çu tõ ®Çu nh÷ng n¨m 1980 vµ chóng kh«ng chØ gióp Ých cho con ng−êi trong viÖc khai th¸c nguån d−îc liÖu tù nhiªn mµ cßn ®ãng gãp rÊt ®¸ng kÓ cho ngµnh khoa häc thÇn kinh [1].Conotoxin lµ lo¹i ®éc tè cã trong näc ®éc cña loµi èc cèi. Chóng lµ hçn hîp c¸c ®éc tè protein/peptide, enzyme hoAEc c¸c ph©n tö cã khèi l−îng thÊp mµ loµi èc dïng ®Ó b¾t måi, c¹nh tranh vµ tù vÖ. §AEc tÝnh cña conotoxin lµ chóng cã t¸c dông khãa chän läc ®iÖn ¸p hoAEc phèi tö cña c¸c kªnh ion trong qu¸ tr×nh dÉn truyÒn tÝn hiÖu cña tÕ bµo thÇn kinh nh− ω-MVIIA tõ Conus magus bao v©y c¸c kªnh Ca ++ type N; µO-conotoxin, δ-PVIA øc chÕ sù bÊt ho¹t c¸c kªnh Na + , κ-PVIIA t−¬ng t¸c víi c¸c kªnh K + ... nªn chóng kh«ng chØ cã vai trß quan träng trong s¨n b¾t con måi mµ cßn lµ c«ng cô h÷u Ých trong khoa häc thÇn kinh ®Ó x¸c ®Þnh ®−îc c¸c thô thÓ nhê ¸i lùc cao vµ ®AEc hiÖu cña m×nh [1,3,9]. Trong c¸c hä conotoxin, ω-conotoxin lµ nhãm ®−îc sö dông nhiÒu trong ngµnh khoa häc thÇn kinh vµ nh÷ng lÜnh vùc nghiªn cøu vÒ chøc n¨ng cña c¸c d¹ng kªnh Ca ++ nh− ω-conotoxin MVIIA tõ Conus magus ® ph¸t triÓn thµnh thuèc cho ®iÒu trÞ c¸c bÖnh gi ¶m ®au [1,4]. Ngoµi t¸c dông gi ¶m ®au, conotoxin cßn rÊt h÷u Ých cho chØ ®Þnh kh¸c trong l©m sµng. ThÝ dô κ-conotoxin PVIIA ® ®−îc chøng minh lµ lµm gi ¶m møc ®é nhåi m¸u c¬ tim trong bÖnh thiÕu m¸u côc bé thö nghiÖm trªn m« h×nh chuét, thá vµ chã in vivo. Trong thµnh phÇn näc ®éc cña mçi loµi èc cèi cã thÓ chøa ®Õn h¬n 200 lo¹i conopeptide do ®ã −íc tÝnh sÏ cã kho ¶ng tõ 50.000 ®Õn 100.000 conopeptide kh¸c nhau cã ho¹t tÝnh d−îc häc ®−îc t×m thÊy tõ tÊt c ¶ c¸c loµi Conus nµy [1]. ChÝnh v× vËy mµ c¸c conopeptide ph ¶i rÊt kh¸c nhau vÒ tr×nh tù amino acid còng nh− ®AEc tÝnh d−îc häc. Dùa vµo tr×nh tù amino acid, conotoxin cã thÓ ®−îc ph©n nhãm thµnh hai lo¹i lín lµ c¸c protein/peptide giµu liªn kÕt disulfide vµ kh«ng giµu disulfide. Trong nhãm peptide giµu disulfide còng ®−îc ph©n lo¹i thµnh c¸c siªu hä phô thuéc vµo liªn kÕt disulfide. Following the strategy of cloning ω-conotoxin MVIIA (ω-CTX) from Conus magus (C. magus), in this article, the expression and purification of the peptide is reported. For that, recombinant plasmid CTXpET32c was designed containing fusion gene encoding thioredoxin (as original) and ω-CTX (Trx-CTX) that is linked by enterokinase restriction site, was transformed into the E. coli BL21 strain. The fusion protein, Trx-CTX combined to 6xHis-tag, is about 21 kDa in size, was expressed by induction with 1 mM IPTG and analyzed by SDS-PAGE. The recombinant protein was isolated and purified by Sepharose chelating affinity chromagraphy. SDS-PAGE results were an evidence to confirm the successful construction and expression of ω-CTX in the fusion form with Trx in E. coli with vector CTXpET-32c. When it was treated by enterokinase to remove thioredoxin, ω-CTX peptide was purified and obtained by using 5 kDa cutoff centrifugal filters.
