This article comprises detailed information about L-asparaginase, encompassing topics such as microbial and plant sources of L-asparaginase, treatment with L-asparaginase, mechanism of action of L-asparaginase, production, purification, properties, expression and characteristics of l-asparaginase along with information about studies on the structure of L-asparaginase. Although L-asparaginase has been reviewed by Savitri and Azmi (2003), our effort has been to include recent and updated information about the enzyme covering new aspects such as structural modification and immobilization of L-asparaginase, recombinant L-asparaginase, resistance to L-asparaginase, methods of assay of L-asparagine and L-asparaginase activity using the biosensor approach, L-asparaginase activity in soil and the factors affecting it. Also, side-effects of L-asparaginase treatment in acute lymphoblastic leukemia (ALL) have been discussed in the current review. L-asparaginase has been and is still one of the most widely studied therapeutic enzymes by researchers and scientists worldwide.

Pesticides, due to their lucrative outcomes, are majorly implicated in agricultural fields for crop production enhancement. Due to their pest removal properties, pesticides of various classes have been designed to persist in the environment over a longer duration after their application to achieve maximum effectiveness. Apart from their recalcitrant structure and agricultural benefits, pesticides also impose acute toxicological effects onto the other various life forms. Their accumulation in the living system may prove to be detrimental if established in higher concentrations. Thus, their prompt and accurate analysis is a crucial matter of concern. Conventional techniques like chromatographic techniques (HPLC, GC, etc.) used for pesticides detection are associated with various limitations like stumpy sensitivity and efficiency, time consumption, laboriousity, requirement of expensive equipments and highly trained technicians, and many more. So there is a need to recruit the methods which can detect these neurotoxic compounds sensitively, selectively, rapidly, and easily in the field. Present work is a brief review of the pesticide effects, their current usage scenario, permissible limits in various food stuffs and 21st century advancements of biosensor technology for pesticide detection. Due to their exceptional performance capabilities, easiness in operation and on-site working, numerous biosensors have been developed for bio-monitoring of various environmental samples for pesticide evaluation immensely throughout the globe. Till date, based on sensing element (enzyme based, antibody based, etc.) and type of detection method used (Electrochemical, optical, and piezoelectric, etc.), a number of biosensors have been developed for pesticide detection. In present communication, authors have summarized 21st century's approaches of biosensor technology for pesticide detection such as enzyme-based biosensors, immunosensors, aptamers, molecularly imprinted polymers, and biochips technology. Also, the major technological advancements of nanotechnology in the field of biosensor technology are discussed. Various biosensors mentioned in manuscript are found to exhibit storage stability of biocomponent ranging from 30-60 days, detection limit of 10(-6) - 10(-16) M, response time of 1-20 min and applications of developed biosensors in environmental samples (water, food, vegetables, milk, and juice samples, etc.) are also discussed. Researchers all over the globe are working towards the development of different biosensing techniques based on contrast approaches for the detection of pesticides in various environmental samples.

A biosensor is an analytical device that consists of an immobilized biocomponent in conjunction with a transducer, and represents a synergistic combination of biotechnology and microelectronics. This review summarizes the use of biosensors for detecting and quantifying heavy metal ions. Heavy metal contamination is of serious concern to human health since these substances are non-biodegradable and retained by the ecological system. Conventional analytical techniques for heavy metals (such as cold vapour atomic absorption spectrometry, and inductively coupled plasma mass spectrometry) are precise but suffer from the disadvantages of high cost, the need for trained personnel and the fact that they are mostly laboratory bound. Biosensors have the advantages of specificity, low cost, ease of use, portability and the ability to furnish continuous real time signals. The analysis of heavy metal ions can be carried out with biosensors by using both protein (enzyme, metal-binding protein and antibody)-based and whole-cell (natural and genetically engineered microorganism)-based approaches.

Thallium concentration reached up to 534 μg L(-1) in the Reigous acid mine drainage downstream from the abandoned Pb-Zn Carnoulès mine (Southern France). It decreased to 5.44 μg L(-1) in the Amous River into which the Reigous creek flows. Tl(I) predominated (>98% of total dissolved Tl) over Tl(III), mainly in the form of Tl(+). Small amounts of Tl(III) evidenced in Reigous Creek might be in the form of aqueous TlCl(2)(+). The range of dissolved to particulate distribution coefficients log K(d) = 2.5 L kg(-1) to 4.6 L kg(-1) indicated low affinity of Tl for particles, mainly ferrihydrite, formed in the AMD-impacted watershed. The low retention of Tl(+) on ferrihydrite was demonstrated in sorption experiments, the best fit between experimental and modeled data being achieved for surface complexation constants log K(ads) = -2.67 for strong sites and log K(ads) = -3.76 for weak sites. This new set of constants allowed reasonable prediction of the concentrations of aqueous and particulate Tl resulting from the mixing of water from Reigous Creek and the Amous River water during laboratory experiments, together with those measured in the Amous River field study.

Various aquatic plant species are known to accumulate heavy metals through the process of bioaccumulation. World's most troublesome aquatic weed water hyacinth (Eichhornia crassipes) has been studied for its tendency to bio-accumulate and bio-magnify the heavy metal contaminants present in water bodies. The chemical investigation of plant parts has shown that it accumulates heavy metals like lead (Pb), chromium (Cr), zinc (Zn), manganese (Mn) and copper (Cu) to a large extent. Of all the heavy metals studied Pb, Zn and Mn tend to show greater affinity towards bioaccumulation. The higher concentration of metal in the aquatic weed signifies the biomagnification that lead to filtration of metallic ions from polluted water. The concept that E. crassipes can be used as a natural aquatic treatment system in the uptake of heavy metals is explored.

Applications of solid phase microextraction for the determination of metallic and organometallic speciesThis paper reviews recent developments of solid phase microextraction (SPME) and its application to the analysis of organometallic species of lead, arsenic, mercury, tin, and selenium by hyphenation with HPLC-GC-atomic spectrometry. In the first part, a background of the technique is given in terms of derivatization, fibers used, extraction and desorption conditions. The second part summarizes typical SPME applications to the determination of organometallic species and the main experimental conditions with the aid of specific examples. Most of the applications comprise alkylation with NaBEt 4 and headspace extraction followed by gas chromatographic separation with a suitable detector.
