Cancer is the second cause of death after cardiovascular diseases in the world. Anticancer prevention used can cause undesirable things. Flavonoids are secondary metabolites derived from natural products that are useful for anticancer treatment. This study was performed to observe the cytotoxic activity of alpinumisoflavone from Erythrina poeppigiana, toward cervical cancer (Hela), colon cancer (WiDr), and hepatoma cancer (HepG2) cells. The cytotoxic activity of alpinumisoflavone was tested using (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyltetrazolium bromide) assay. The percentage of cell mortality was calculated and the IC50 was calculated using probit analysis. The result shown that alpinumisoflavone has antiproliferative effect to colon cancer (WiDr), cervical cancer (Hela), and hepatoma cancer (HepG2) cells with the value of IC50 are 5.63, 7.18, and 18.08 µg/ml, respectively. Based on the value of IC50 alpinumisoflavone is very cytotoxic to colon cancer WiDr cell.
Abstrak: Teknik pemisahan sangat diperlukan dalam mengisolasi senyawa-senyawa yang mempunyai bioaktivitas untuk memperoleh senyawa murni dalam suatu riset pengembangan di bidang kimia organik bahan alam hayati. Senyawa yang diisolasi dari jaringan tumbuhan secara alamiah berada dalam keadaan tercampur dengan senyawa lainnya, oleh karena itu perlu dipilih teknik pemisahan dan pemurnian yang sesuai. Kromatografi Cair Kinerja Tinggi (KCKT) Preparatif merupakan salah satu instrumen pemisahan modern yang digunakan untuk memisahkan dan memurnikan campuran senyawa dengan daya pisah tinggi, cepat (throughput tinggi). Tujuan dari penelitian ini adalah untuk menetapkan kondisi optimum pemisahan senyawa flavonoid dari fraksi polar Erythrina poeppigiana menggunakan sistem KCKT preparatif. Penelitian ini dilakukan dengan tahapan : Ekstraksi dan Fraksionasi, proses pemisahan dan pemurnian, analisis KCKT analitik fraksi polar E. poeppigiana, penentuan kondisi optimum KCKT preparatif dengan menggunakan kolom C18 meliputi; konsentrasi sampel yang diinjeksikan, pemilihan fasa gerak, dan laju alir pada panjang gelombang deteksi 254 nm dan 365 nm. Dari hasil penelitian diperoleh kondisi pemisahan yang optimum dari fraksi polar E. poeppigiana yaitu pada konsentrasi sampel 3000 ppm dan fase gerak campuran metanol dan air (7:3,v/v) dengan kecepatan laju alir 5,28 mL/menit kromatogram hasil KCKT yang dihasilkan menunjukkan satu puncak. Kondisi optimum yang diperoleh tersebut selanjutnya digunakan oleh peneliti untuk memisahkan senyawasenyawa dalam fraksi polar E.poeppigiana yang dikerjakan di laboratorium.Abstract: Separation techniques are very necessary in isolating compounds that have bioactivity to obtain pure compounds in development research in the field of organic chemistry of biological natural materials. Compounds isolated from plant tissue are naturally mixed with other compounds, therefore the appropriate separation and purification techniques need to be chosen. High Performance Liquid Chromatography (HPLC) Preparative is one of the equipment in the laboratory that is used to separate and purify compound mixtures with high separability, fast (high throughput). The purpose of this study was to determine the optimum conditions for the separation of flavonoids from the polar fraction of Erythrina poeppigiana using the preparative HPLC system. This research was carried out in the following stages analyzing KCKT analytical polar fraction of E. peppigiana, determining the optimum conditions of preparative HPLC using C18 columns including the concentration of the injected sample, mobile phase selection, and flow rate at 254 nm and 365 nm detection wavelengths. The results showed that the optimum separation conditions of the polar E. poeppigiana fraction were at a sample concentration of 3000 ppm and the mobile phase of a mixture of methanol and water (7:3 v/v) with a velocity flow rate of 5.28 mL / min.
