Microsatellite markers or SSR (Simple Sequence Repeats) have proved to be an excellent tool for cultivar identification, pedigree analysis and the evaluation of genetic distance among organisms. Soybean cultivars have been characterized mainly by morphological and biochemical traits. However, these traits have not been sufficient to characterize the large number of cultivars eligible to receive protection under the Brazilian Cultivar Protection Act. In order to define new soybean cultivar markers, the alleles of twelve SSR loci of 186 Brazilian soybean cultivars were studied by estimating the variation in their size range and their respective frequencies. On average, 5.3 alleles per locus were detected, with a mean genetic diversity of 0.64 ± 0.12. These loci were used to distinguish morphologically similar groups, presenting a mean similarity coefficient of 0.46; their use allowed to determine 184 profiles for the 186 cultivars. A dendrogram based on the SSR loci profiles showed good agreement with the cultivar pedigree information.

Para avaliar os efeitos do tamanho de semente sobre o crescimento inicial das plantas, a produtividade e a qualidade fisiológica foram testadas três cultivares de soja, BRSMG 752S, BRSMG 790A e BRSMG 750SRR originadas de três tamanhos de semente (peneiras 4,0 mm, 5,0 mm e 6,0 mm). A semeadura foi efetuada em 06/12/2007 em plantio direto e o desbaste aos 21 dias após a semeadura, deixando-se treze plantas por metro. A colheita manual foi realizada em 11/04/08. Após a colheita, as sementes foram classificadas em três tamanhos, usando-se peneiras manuais de 6,0 mm, 6,5 mm e 7,0 mm. O delineamento experimental foi em blocos ao acaso para o experimento de campo e inteiramente casualizado para as análises de laboratório, ambos em esquema fatorial 3 (cultivar) x 3 (peneira), com quatro repetições. Os dados obtidos foram submetidos à análise de variância pelo teste F e as médias foram comparadas pelo teste de Tukey a 5% de probabilidade. Para as avaliações de campo, foram determinadas altura de plantas e produtividade. No laboratório, os tratamentos foram avaliados pelos testes de germinação e de vigor (envelhecimento acelerado, classificação do vigor da plântula, comprimento e massa seca de raiz). Foi verificado que sementes menores (peneira 4,0 mm) produzem plantas com menor altura na colheita e menor produtividade, em relação às sementes maiores (peneira 6,0 mm). As sementes classificadas em diferentes tamanhos apresentam diferenças em qualidade fisiológica. Sementes maiores (peneira 7,0 mm) apresentam maiores porcentagens de germinação e de vigor.Termos para indexação: Glycine max, peneira, viabilidade, vigor. Soybean [Glycine max (L.) Merr.] has been studied and enhanced for most of its economically important traits. Previous research has studied the association among them and the effect of the genotype × environment interaction; however, less is known about their correlation considering absolute maturity, as well as the use of multiple selection indices to study the genotype × environment interaction and select superior cultivars. Regarding this, the aim of the present study was to identify lines that associate precocity, good yield performance, and high oil and protein contents in the grains, as well as to estimate the correlation among these traits and study the effect of genotype × environment interaction using a standardized multiple selection index. Trials were conducted in two crop seasons in the state of Minas Gerais, Brazil, with 39 lines in 13 evaluation environments. The experiments were conducted in a randomized complete block design with four replications, and the grain yield, absolute maturity, and oil and protein contents in the grains were evaluated. The results indicated high experimental precision and accuracy, with significant differences among lines for all traits. High‐magnitude correlations between evaluated traits were found, highlighting the negative correlation between absolute maturity and protein content in the grains. The genotype × environment interaction was also significant, and the use of the multiple selection index was efficient to identify superior and stable inbred lines by the genotype + genotype × environment (GGE) biplot method, which explained 82.23% of the genotype × environment interaction effect. Lines 27 and 31 stood out from the others because they associated stability and good performance for all evaluated traits.
Heterodera glycines Ichinohe, the soybean cyst nematode (SCN) was detected in Brazil in 1992 and since then it has been causing losses to Brazilian soybean crop yields. SCN populations have great genetic diversity which makes it difficult to manage this disease. The objectives of this research were to investigate the genetic variation of 16 SCN populations sampled in middle western and southeastern Brazil, utilizing random amplified polymorphic DNA (RAPD) techniques, and to establish useful and specific RAPD markers for SCN race 3. RAPD markers demonstrated genetic variability among and within SCN populations, and they could be used for monitoring nematode population dynamics. The OPA-07 primer was a reliable molecular marker for race 3, while electrophoretic profile analysis of DNA fragments amplified with OPA-10 primer detected slight variation within those populations identified as race 3. The SCN population from Chapadão do Céu, GO - sample 2 - was the most genetically distant from the other populations.
Heterodera glycines Ichinohe, o nematóide de cisto da soja (SCN), foi detectado no Brasil em 1.992 e desde então tem provocado perdas consideráveis à produtividade da soja brasileira. Populações de SCN apresentam grande diversidade genética, o que dificulta o manejo dessa cultura em áreas infestadas. Os objetivos desta pesquisa foram investigar a possível variação genética de dezesseis populações de SCN amostradas nas regiões Centro Oeste e Sudeste do Brasil, utilizando a técnica do RAPD (amplificação casualizada de DNA polimórfico), bem como estabelecer marcadores RAPD específicos para a raça 3 do SCN. Nossos resultados mostraram que os marcadores RAPD podem ser utilizados em estudos de variabilidade genética entre populações de SCN e dentro de raças deste nematóide, bem como podem ser usados no monitoramento de dinâmica populacional deste patógeno. O "primer" OPA-07 foi considerado satisfatório como marcador molecular para a raça 3, enquanto a análise eletroforética dos fragmentos de DNA amplificados com a utilização do "primer" OPA-10 detectou ligeira variação nas populações de SCN identificadas como raça 3. A população do SCN oriunda de Chapadão do Céu, GO - amostra 2 - foi a geneticamente mais distante, quando comparada às demais usadas neste estudo
Os objetivos deste trabalho foram selecionar progênies superiores de soja e avaliá-las, em casa de vegetação, quanto à resistência ao nematóide tipo 3 de cisto da soja (Heterodera glycines). Foram avaliadas 222 progênies segregantes de soja, em ensaios conduzidos em campo, nos anos 1999/2000, 2000/2001 e 2001/2002, sob delineamento de blocos aumentados de Federer, e no ano 2002/2003 em blocos ao acaso, com duas repetições, tendo sido avaliados dez atributos agronômicos. No ano de 2003 foi conduzido em casa de vegetação um ensaio com 11 progênies superiores, para a avaliação de resistência ao nematóide de cisto, adotando-se delineamento inteiramente casualizado, com cinco repetições. Com relação aos atributos agronômicos, as progênies JAB 99-17-4-9-1 e JAB 99-40-12-1-2 destacaram-se das demais por possuir médias adequadas para a maioria dos atributos. Por sua vez, na avaliação de resistência em casa de vegetação, seis progênies revelaram-se resistentes ao nematóide de cisto da soja tipo 3.
-The difficulty on identifying, lack of segregation systems and absence of suitable standards for coexistence of non trangenic and transgenic soybean are contributing for contaminations that occur during productive system. The objective of this study was to evaluate the efficiency of two methods for detecting mixtures of seeds genetically modified (GM) into samples of non-GM soybean, in a way that seed lots can be assessed within the standards established by seed legislation. Two sizes of soybean samples (200 and 400 seeds), cv. BRSMG 810C (non-GM) and BRSMG 850GRR (GM), were assessed with four contamination levels (addition of GM seeds, for obtaining 0.0%, 0.5%, 1.0%, and 1.5% contamination), and two detection methods: immunoassay of lateral flux (ILF) and bioassay (pre-imbibition into 0.6% herbicide solution; 25 ºC; 16 h). The bioassay is efficient in detecting presence of GM seeds in seed samples of non-GM soybean, even for contamination lower than 1.0%, provided that seeds have high physiological quality. The ILF was positive, detecting the presence of target protein in contaminated samples, indicating test effectiveness. There was significant correlation between the two detection methods (r = 0.82; p ≤ 0.0001). Sample size did not influence efficiency of the two methods in detecting presence of GM seeds.Index terms: Glycine max, glyphosate, transgenic.Detecção de presença adventícia de semente geneticamente modificada em lotes de soja não transgênica RESUMO -A dificuldade de identificação, a falta de sistemas de segregação e a ausência de normas adequadas à coexistência de soja não transgênica e transgênica têm contribuído para que ocorram contaminações durante as etapas do processo produtivo. O objetivo deste trabalho foi verificar a eficiência de dois métodos de detecção de misturas de sementes geneticamente modificadas (GM) em amostras de soja não transgênica, para avaliar lotes de sementes quanto aos limites da legislação. Foram avaliados dois tamanhos de amostra (200 e 400 sementes), das cultivares BRSMG 810C (não transgênica) e BRSMG 850GRR (GM), com quatro níveis de contaminação (sementes GM adicionadas para obter contaminações de 0,0%, 0,5%, 1,0% e 1,5%), e dois métodos de detecção: imunoensaio de fluxo lateral (IFL) e bioensaio (pré-embebição em solução do herbicida a 0,6%; 25 ºC; 16 h). O bioensaio é eficiente na detecção da presença de sementes GM em amostras de semente não transgênica, mesmo para contaminação inferior a 1,0%, desde que apresentem alta qualidade fisiológica. O IFL foi positivo, detectando a presença da proteína alvo nas amostras com contaminação, indicando a eficiência do teste. Houve correlação significativa entre os dois métodos utilizados (r = 0,82; p ≤ 0,0001). O tamanho da amostra não influenciou a eficiência dos dois métodos na detecção da presença de sementes GM.Termos para indexação: Glycine max, glifosato, transgênicos.
Four microsatellite sequences were tested in soybean DNA from cultivars and segregating genotypes. Three of them were close to the resistance locus rhg1 on molecular linkage group G (Satt309, Sat_168, Sat_163) and one was close to Rhg4 locus on group A2 (Sat_162). Progenies previously classified as cyst nematode (SCN) resistant and others with unknown reaction were tested, using the resistant cultivars Liderança, and Renasscença and the susceptibles 'Cristalina' and 'OCEPAR-4' as control. The best primer for resistance to SCN was Sat_162. Twenty segregant progenies tested with Sat_162 presented a 150 bp band for homozygous resistant genotypes and 200 bp for susceptible ones, and both for the heterozigous genotypes. Previous studies have shown that this microsatellite marker is efficient to select genotypes carrying Peking-derivative resistance. 'Peking' takes part in the in the genealogy of all segregant progenies evaluated in this study. Thus, the obtained results showed that Sat_162 distinguished resistant and susceptible soybean genotypes to SCN, race 3.

Merr.) cultivar BRS 217 Flora was developed by Embrapa and released for production in the states of Goiás, Minas Gerais, Bahia, Mato Grosso and the Distrito Federal, Brazil. It is resistant to stem canker, frog-eye leaf spot, bacterial pustule, and partially resistant to powdery mildew.
