The collection and characterization of tomato germplasm are of relevant importance for agriculture. This study aimed to collect and characterize, by way of morphological description and statistical tools for the composition of groups, tomato accesses from the Southern and Serrana regions of the State of Espírito Santo, as well as to establish the tomato germplasm bank of the Ifes - Campus de Alegre. Thirty-seven accessions were collected from different commercial locations of Espírito Santo. The experiment was conducted in the Ifes Campus de Alegre in the Agroecology sector. For the morpho-agronomic characterization, sixteen essential descriptors were used, nine quantitative and seven qualitative. In the color of the ripe fruit, there was the formation of five distinct groups with the colors yellow, green, orange, pink and red. In the format of the fruit, three groups were formed, slightly flattened, flattened and rounded. The ANOVA showed that there was a significant difference for all the quantitative characteristics evaluated. According to the Ward-MLM procedure, due to the high likelihood function in group seven (from 130.71), it was possible to group the 37 genotypes into seven groups. The greatest dissimilarity was observed in groups V and VII with a distance of 330.02, and the least dissimilar ones, groups IV and VI, with 8.21. Genetic variability was detected in tomato germplasm for the morpho-agronomic characteristics. Many of the accessions are promising sources of phenotypes of interest to the Ifes germplasm bank.
The effects of the cryoprotectants dimethyl sulfoxide, glycerol and methyl glycol at different concentrations on Austrolebias minuano sperm quality parameters were evaluated in this study. The cellular kinetic parameters, determined using flow cytometry, indicated the best results with the samples cryopreserved with 7.5% methyl glycol. Dimethyl sulfoxide concentrations of 7.5% and glycerol concentrations of 10%, 12.5% and 15% demonstrated the least benefit across all evaluated sperm kinetic parameters. When assessed using flow cytometry, a concentration of 7.5% methyl glycol similarly showed better sperm kinetic parameters than 12.5% dimethyl sulfoxide. We conclude that cryoprotectants, especially methyl glycol, are effective for the preservation of sperm quality in A. minuano. However, high sensitivity of spermatozoa against glycerol was observed in these studies; thus, it is not recommended for cryopreservation purposes.

The definition of species boundaries constitutes an important challenge in biodiversity studies. Cynopoecilus Regan, 1912 encompasses several endangered species of annual fish, occurring in temporary ponds in a restricted area of Southern Brazil and Uruguay. Divergences about the taxonomic status of Cynopoecilus species highlight the importance of species delimitation studies. Therefore, we address here the phylogenetic structure of Cynopoecilus, while assessing its taxonomic implications. For this, fragments of the mitochondrial COI and nuclear RAG1 genes were characterized and analyzed for a set of 275 and 280 specimens, respectively. DNA barcoding and phylogenetic analyses detected subdivision of these specimens in 8–10 clusters, which comprise the six previously described species, and suggest one invalid taxon and at least 3–5 putative new species. The phylogenetic structure also suggests that the Jacuí River and the Patos Lagoon historically acted as effective barriers to gene flow between populations, although some isolated dispersal events across these water bodies could be evidenced, especially for C. melanotaenia Regan, 1912. In general, the results highlight the need of independent conservation strategies within the distribution area of each of the endemic allopatric killifish clusters, while questioning several taxonomic boundaries and distribution data.
