ABSTRACT. Genetic diversity in crops is essential to make improvements related to superior germplasms. Implementation of molecular markers to identify suitable genotypes speeds up the breeding progress by enhancing selection efficiency. This study was carried out to probe genetic diversity among 21 maize genotypes using 20 inter simple sequence repeat (ISSR) markers. We identified a total of 190 polymorphic bands with an average of 9.5 alleles per primer. The highest number of polymorphic bands (17) was found using ISSR marker UBC-10, whereas the lowest number of polymorphic bands (4) was found using UBC-809. The coefficient of genetic similarity ranged from 0.888 to 0.118%. The highest similarity was found between accessions 12 (015224) and 9 (015114), whereas the lowest similarity was found between genotypes 20 (EV-5098) and 14 (015030). The polymorphism information content ranged from 0.17 to 0.47. A dendrogram was generated based on Jaccard's distance matrix. The genotypes were found to group into two major clusters that could be further partitioned into two sub-clusters. Genotypes located within the same cluster are genetically more closely related to each other. The present study efficiently identified diverse genotypes that may be used for creating new varieties with distinct characteristics. The identified genotypes could be used as parents for future development of diverse populations.
Objective:The present study aimed to investigate the association between the rs10757274 SNP (present on locus 9p21 in the gene for CDKN2B-AS1) and coronary artery disease (CAD) in a local population of Pakistan.Methods:It was a case-control study. An allele-specific PCR-based strategy was used for the identification of genotypes. A total of 350 samples were used for the investigation, out of which 220 samples were CAD patients and 130 samples were normal healthy individuals. Effects of parameters, like family history of CAD, smoking, presence of diabetes, and hypertension, in changing the chances of CAD were studied. Odds ratio was estimated with 95% confidence interval.Results:A strong association was observed between CAD and factors, like smoking (OR: 1.666; 95% CI: 1.042-2.664), presence of hypertension (OR: 26.55; 95% CI: 15.95-44.20), diabetes (OR: 3.009; 95% CI: 1.841-4.920), and family history of CAD (OR: 4.9; 95% CI: 2.965-8.099). Results for the association between the genotype on the basis of rs10757274 showed a strong association between the GG genotype and the occurrence of CAD (OR: 9.603; 95% CI: 5.746-16.05).Conclusion:The present results suggest the importance of the 9p21 locus in modulating the chances of CAD.
The Kyoto Protocol includes agroforestry practices as a suggested approach for mitigating global atmospheric CO2. Agroforestry systems are a desirable option for mitigating atmospheric CO2, as they provide numerous secondary benefits, including food, fodder, fuel, increased farm income, biodiversity maintenance, and soil conservation. This research was planned to assess the current carbon storage status and future potential of agroforestry systems in Pakistan through a nondestructive approach (allometric equations) in 14 subdivisions (tehsils) of three selected districts located in the irrigated plains of Punjab, Pakistan. A total of 1750 plots of 0.405 ha each were selected in a randomized, unbiased sampling to estimate the total number of trees, tree species diversity, diameters at breast height (DBHs), and tree heights. Soil carbon was also measured at 0–30 cm using the Walkley–Black method in a subset of plots. It was found that the current number of trees in farms in the study area varied from 18 to 51 trees/ha, which can be increased to 42–83 trees/ha if all the farmers plant the maximum permissible number of trees along with their crops. The estimated total tree carbon stock ranged from 0.0003 to 8.79 Mgha−1, with the smallest mean value of 0.39 Mgha−1 for tehsil Faisalabad, and the largest mean value of 1.41 Mgha−1 for tehsil Chiniot. The whole study area had an estimated woody vegetation carbon stock of 950,470 Mg and a soil carbon stock of 22,743,008 Mg. If farmers in the study area all increased tree stocking to their average maximum desired stocking, the total tree carbon stock would more than double to 2,497,261 Mg. These results highlight both the current and potential carbon sequestration potential of agroforestry in Pakistan and can be further used in devising strategies for implementing tree planting programs on agricultural lands and designing future carbon sequestration projects in Pakistan.
Reduced production of nitric oxide due to rs1799983 single nucleotide polymorphism in nitric oxide synthase 3 gene (NOS3) may enhance the risk of coronary artery disease. The association of rs1799983 polymorphism with coronary artery disease was investigated in the local population of Pakistan. Study consisted of 376 individuals, out of which 198 were coronary artery disease patients and 178 were normal healthy individuals. Allele-specific polymerase chain reaction (PCR) based strategy was used for the detection of different genotypes of rs1799983 polymorphism. PCR amplification results were obtained for 354 samples. Frequency of T allele was higher as compared to G allele in our population. Strong association between rs1799983 and coronary artery disease was observed (p < 0.01). TT genotype was found to enhance 5.717 times the risk of coronary artery disease (odds ratio (OR): 5.717; 95% confidence interval (95% CI) 3.586-9.115). On the basis of present results, it can be concluded that rs1799983 is strongly associated with coronary artery disease in our population and TT genotype of this polymorphism enhanced the risk of coronary artery disease in Pakistani population.
