Synthetic hexaploid (SH) wheat derived from crossing tetraploid durum wheat and diploid Aegilops tauschii provide germplasm for wheat improvement as the conventional wheat varieties possess very low genetic diversity. This study aims to identify diverse SH lines which can be used in breeding programs for transferring the desired traits into bread wheat. The study was conducted on 24 SH lines using 10 pairs of randomly amplified polymorphic DNA (RAPD) markers. Application of RAPDs showed the presence of some diagnostic bands in SH wheats that were absent in durum parents suggest that these bands are donated by D genome of the wild relative Ae. tauschii.

Studies on the insect pollinators diversity and their relative abundance in Eruca sativa Mill. (Arugula) and Brassica rapa L. (field mustard) was carried out during spring season from February to April consecutively during all the three years of 2016–18. Insect pollinators observed belonged to four orders i.e. Hymenoptera, Diptera, Lepidoptera, and Coleoptera. A total of 20 major species of insect pollinators were recorded. The highest abundance of pollinator species belonged to Hymenoptera. The most prominent insect pollinator species were Apis mellifera followed by other three honey bee species of A. cerana, A. florea, and A. dorsata respectively. Some species of solitary bees were also recorded. From Diptera, four species of syrphid fly and one species from Muscidae family were also recorded. Insect pollinators recorded from order Lepidoptera were Pieris brassicae, Vanessa cardui, and Papilio demoleus. Lady bird beetle Coccinella septempunctata was recorded from Coleoptera order as occasional visitor. It was noticed that E. sativa attracted more insect pollinators than B. rapa which may be attributed to different amount and chemical properties of nectar, with number of pollen grains, and flower canopy of both crops. Further studies are needed to confirm the reasons for higher pollinator visitation to E. sativa than B. rapa through chemical analysis of nectar, amount of pollens, flower physiology and phenology of both crops.

During the growing seasons of 2014 through 2016, a total of 336 leaf samples from bell pepper (showing leafroll and interveinal yellowing) and arable weeds were collected from Riyadh region, Saudi Arabia. The use of a polerovirus generic reverse transcription (RT)-PCR assay confirmed their presence in the bell pepper samples. Sequencing of the generic amplicon revealed high similarity (87.6 to 98.1% in nt) with four poleroviruses; Tobacco vein distorting virus, Pepper vein yellows virus, Pepper yellows virus, and Pepper yellow leaf curl virus. To further characterize one of these isolates (105D), a larger part of the genome (∼1,300 nt) spanning approximately from the 3′ end of ORF2 to the middle of ORF3, was amplified and sequenced. Blasting the resulting sequence revealed the low amino acid and nucleotide identity percentages in the coat protein and movement protein partial genes with viruses deposited in GenBank. Next-generation sequence was used to acquire a larger part of the genome, which resulted in the reconstruction of isolate 105D’s partial genome (5,496 nt). Sequence similarity analysis revealed the presence of a divergent polerovirus isolate belonging to a new species that was tentatively named Pepper leafroll chlorosis virus (PeLRCV). Using a specific RT-PCR assay for this isolate confirmed the presence of this new viral species in the symptomatic peppers. Aphid transmission experiments showed that PeLRCV is vectored by Aphis gossypii and that it can infect at least five out of the 15 different plants species tested. Based on our findings, PeLRCV is a new member of genus Polerovirus in the family Luteoviridae.

Xanthomonas citri subsp. citri, a causative agent of the citrus canker (CC) disease, belongs to one of the essential groups of the bacterial phytopathogen family, Xanthomonadaceae. It has been a potential threat to the globally significant citrus fruit crop, which has remained under investigation for disease management and epidemiology since the 1980s. In Pakistan, the average yield of citrus is 11 t/ha, which is lower than other countries, including China, Brazil, and India, having average productions of 27, 26, and 22 tons/hectare, respectively. Citrus canker is one of the most devastating diseases, posing a significant threat to crop yield and fruit quality. To date, five distinct types (or forms) of the citrus canker have been recognized; the Asiatic (Canker A) form is most destructive and affects most citrus cultivars. Severe infection outcomes include dieback, defoliation, severely blemished fruit, premature fruit drop, and reduced fruit quality. The infection increases under humid, warm, cloudy climate, wind, and heavy rainfall. The analysis of plasmid and chromosomal DNA of X. citri subsp. citri depicted an evolutionary relationship among pathovars of Xanthomonas. The extensive study on the genome of X. citri subsp. citri has contributed to the current knowledge of plant host recognition of pathogens, host specificities, dissemination, and propagation. Regulatory programs, i.e., quarantine or exclusion, continued to be practiced, prohibiting infected citrus plant material into the existing stock. Other measures include removal of inoculums sources, resistant hosts, protective copper-containing sprays, and windbreak systems. In this review, we explored the latest trends in the areas of epidemiology, pathogenome, detection, host–pathogen interaction, biofilm formation, and management of X. citri subsp. citri.

Plants in nature are under the persistent intimidation of severe microbial diseases, threatening a sustainable food production system. Plant-bacterial pathogens are a major concern in the contemporary era, resulting in reduced plant growth and productivity. Plant antibiotics and chemical-based bactericides have been extensively used to evade plant bacterial diseases. To counteract this pressure, bacteria have evolved an array of resistance mechanisms, including innate and adaptive immune systems. The emergence of resistant bacteria and detrimental consequences of antimicrobial compounds on the environment and human health, accentuates the development of an alternative disease evacuation strategy. The phage cocktail therapy is a multidimensional approach effectively employed for the biocontrol of diverse resistant bacterial infections without affecting the fauna and flora. Phages engage a diverse set of counter defense strategies to undermine wide-ranging anti-phage defense mechanisms of bacterial pathogens. Microbial ecology, evolution, and dynamics of the interactions between phage and plant-bacterial pathogens lead to the engineering of robust phage cocktail therapeutics for the mitigation of devastating phytobacterial diseases. In this review, we highlight the concrete and fundamental determinants in the development and application of phage cocktails and their underlying mechanism, combating resistant plant-bacterial pathogens. Additionally, we provide recent advances in the use of phage cocktail therapy against phytobacteria for the biocontrol of devastating plant diseases.

BACKGROUND Bacillus species synthesize antifungal lipopeptides (LPs) making them a sustainable and eco‐friendly management option to combat Fusarium wilt of chickpea. RESULTS In this study, 18 endophytic Bacillus strains were assessed for their antifungal activity against Fusarium oxysporum f. sp. ciceris (FOC) associated with Fusarium wilt of chickpea. Among them, 13 strains produced significant inhibition zones in a direct antifungal assay while five strains failed to produce the inhibition of FOC. Bacillus thuringiensis CHGP12 exhibited the highest inhibition 3.45 cm of FOC. The LPs extracted from CHGP12 showed significant inhibition of the pathogen. Liquid chromatography–mass spectrometry (LC–MS) analysis confirmed that CHGP12 possessed the ability to produce fengycin, surfactin, iturin, bacillaene, bacillibactin, plantazolicin, and bacilysin. In an in vitro qualitative assay CHGP12 exhibited the ability to produce lipase, amylase, cellulase, protease, siderophores, and indole 3‐acetic acid (IAA). IAA and gibberellic acid (GA) were quantified using ultra‐performance liquid chromatography (UPLC) with 370 and 770 ng mL−1 concentrations of IAA and GA respectively. Furthermore, the disease severity showed a 40% decrease over control in CHGP12 treated plants compared to the control in a glasshouse experiment. Moreover, CHGP12 also exhibited a significant increase in total biomass of the plants namely, root and shoot growth parameters, stomatal conductance, and photosynthesis rate. CONCLUSION In conclusion, our findings suggest that B. thuringiensis CHGP12 is a promising strain with high antagonistic and growth‐promoting potential against Fusarium wilt of chickpea. © 2022 Society of Chemical Industry.
