Objective. The effect of a proprietary formulation of trans-resveratrol (t-RSV) on manifestations of diabetic foot syndrome (DFS) was studied in type 2 diabetic patients with newly diagnosed diabetic foot ulcers. Method. Placebo-controlled, examiner-blinded, parallel-group randomized controlled pilot clinical trial (ACTRN Clinical Trial Registry number 12610000629033) involving 24 patients with DFS (15 males and 9 females, average age of 56.4 ± 9.1 years) divided into the placebo and RSV-treatment groups was performed. 50 mg of t-RSV or placebo capsules was given to each patient twice a day over a 60-day time period. Results. Reduction in the parameters reflecting diabetic ulcer size was more profound in the RSV group as compared to placebo. RSV-treated patients also had a marginally improved performance in the foot pressure test. A statistically significant decline in the plasma fibrinogen level, but not CRP, was also found in the RSV-treated patients. Some improvement in the plasma lipid profile and fasting glucose levels were not related to RSV-treatment, since they have been seen on both the RSV and placebo groups, revealing the effectiveness of medical supervision and education in the newly diagnosed patients with DFS. Conclusion. t-RSV supplementation promotes reduction of the foot ulcer size and reduces plasma fibrinogen level in type 2 diabetic patients.
The effect of inclusion of three antioxidants (Vitamin E, cysteine and glutathione) in ram semen extender on the release of antioxidant enzymes and free radicals was studied. A 3 × 3 factorial experiment was conducted to test effects of supplementing ram semen extender with Vitamin E (1, 5 and 10 IU), cysteine (1, 5 and 10 mM) or glutathione (0.5, 1 and 2 mM) on the sperm survival and release of superoxide dismutase (SOD), glutathione peroxidase (GPX) and thiobarbituric acid reactive substances (TBARS). Eighty ejaculates of eight fertile Najdi rams were collected, assessed for the gross examination and the good ejaculates (≥90% motility) were pooled and sperm count was assessed. Therefore ten extenders; control (C) and 9 treated (C plus antioxidant) were tested. Extended semen was stored at 5˚C for 96 h, examined for motility and survival and sperm cells were separated from plasma, sonicated, homogenized and exposed to the determinations of SOD, GPX and TBARS. The highest sperm survival was found in diluents containing 5 IU Vitamin E/ml, 1 and 2 mM glutathione (55.5% survival), while the lowest survival was found in 10 mM cysteine (11.1%). TBARS concentration was highest (P < 0.05) in control than other treatments, however, Vitamin E and glutathione exhibited low values. Contrariwise, activities of SOD and GPX increased (P < 0.05) within sperm cells and seminal plasma in diluents containing 5 IU E, 1 mM and 2 mM glutathione. The enzymatic activities were generally higher in seminal plasma than in sperm cells. It was concluded that supplementing ram semen extender during chilled storage with 5 IU Vitamin E per ml or 1-2 mM glutathione enhanced sperm survival and reduced free radicals.
Ammonia is a production limiting factor in the aquaculture media affecting fish production. A study was designed to scrutinize effects of ammonia on growth performance, survival, cortisol and hematological parameters of Tilapia fish. The study examined effects of 96 h-incubation of male and female Tilapia with 3 mg ammonium chloride per a liter of water compared to control. The study has been carried out in the physiology laboratory of the department of animal and fish production, Alexandria University. Fourteen aquaria were used (6 control and 8 ammoniated). Each aquarium contained 6 fish (half the population males and the other have females). Duration of the control reared fish was 30 days, however the duration for ammoniated group was 4 days. In all stressed fish, there found decreases in final body weight, average daily gain and specific growth rate as compared to controls. Hematological parameters revealed increases (P0.05) in total leukocyte counts in both males and females exposed to stressors. There were significant decreases (P0.05) in red blood cell, hematocrit value and hemoglobin concentration in both males and females. There were non-significant differences (P0.10) in these parameters between males and females. Exposing both male and female tilapia to ammonia, resulted in increases (P0.05) in mean corpuscular volume (MCV). Mean corpuscular hemoglobin (MCH) didn’t change in male tilapia, while females expressed increased MCH values in the ammonia condition. Mean corpuscular hemoglobin concentration (MCHC) decreased (P0.05) under ammonia with no differences between males and females. Differential leukocyte count exhibited increases (P0.05) in neutrophils in ammonia-exposed males and females and decreases (P0.05) in eosinophils and monocytes in males, but not in females. However, lymphocytes decreased (P0.05) in both females and males exposed to ammonia. Cortisol level increased (P0.05) by about 2 folds in both sexes of fish exposed to ammonia (2.95 and 2.72 vs6.40 and 6.48 ng/ml in control males and females vs ammonia-exposed males and females).Rearing tilapia fish in media containing high level (3mg/l water) of ammonium chloride not only deteriorated growth rate but it also negatively affected the health wellbeing.
This study was conducted to investigate the effects of 10 weeks supplementation of Quebracho tannins (QT; 0 [control], 100 [QT 100 ] or 200 g/[cow·d] [QT 200 ]) to 30 multiparous postpartum buffalo cows (10 cows per group) on milk yield and composition, blood metabolites and reproductive performance. Supplementation of QT 100 had no significant effect on milk yield, whereas QT 200 decreased ( P < 0.05) this trait. Compared with the control group, both QT levels decreased ( P < 0.05) fat-corrected milk (FCM) yield, but no significant effects were found on percentages of milk fat and protein. Contrariwise, yields of milk fat, lactose and milk protein were decreased ( P < 0.05) when QT 200 was supplemented. The solids nonfat (SNF) percentage and yield were decreased ( P < 0.05) with QT 100 supplementation. Moreover, QT tended to numerically reduce total number of ovarian follicles, number of small follicles, peripheral progesterone concentration and conception rate. Supplementation of QT 200 numerically increased number of large follicles, mean diameter of large follicle, number and diameters of corpora lutea. The inclusion of QT 200 shortened days open (DO) and decreased number of services per conception. Contrariwise, QT did not show significant effects on serum total protein, albumin, globulin, glucose, cholesterol and triglycerides concentrations. Supplementation of QT 100 caused an increase ( P < 0.05) of serum urea compared with that in control and QT200 groups. Generally, QT decreased ( P < 0.05) serum creatinine concentration. Therefore, the supplementation of a commercial QT to early lactating Egyptian buffalo cows displayed negative consequences on their productive and reproductive performances.
