Collectively, these data showed that HeECo could blunt the postprandial glycemic surge in rats; possibly through inhibition of alpha-glucosidase and positive modulation of antioxidant enzymes. Our findings confirmed the anti-hiperglycemic activity of HeECo in STZ- diabetic rats. Cedrela odorata is effective in diminishing glucose levels in vitro and in vivo and in ameliorating oxidative damage that occurs in diabetes and/or due to hyperglycemia in rats. According to our results, the efficacy of Cedrela odorata preparation could be due to the presence of active principles with different mode of actions at the molecular level, including α-glycosidases and glucose transporter inhibitors and antioxidant property.

This study assessed the effect of whey protein substitution with isolated soy protein in protein bar (PB) formulations at 25% (PB2), 50% (PB3), or 75% (PB1) weight/weight on the proximate and mineral composition, sensory, and antidiabetic properties. Sensory evaluation was conducted within diabetic (DB) and non-diabetic (NDB) consumers by preference ranking and acceptance test. The formulations were analysed in terms of moisture, ash, protein, lipid, carbohydrates, fibers and mineral content. The consumers did not distinguish the formulations by preference ranking test. However, the acceptability test showed a rating of 9 most frequent for PB1 (36.30%), followed by PB2 and PB3 (both 34.09%), among DB consumers. The PB1 and PB3 showed higher content of total, soluble and insoluble fibers and, PB 2 presented higher carbohydrate content. Potassium, sodium and calcium showed the highest mineral content in the formulations. PB3 was assessed for glycaemic and lipidemic control in diabetics and non-diabetics female Wistar rats, for this 20% of PB was added in the ration consumed ad libitum, besides, the rats received 100 mg/kg b. w. by gavage daily. The treatment did not reduce significantly fasting glucose, lipid profile, or peripheral glucose disposal in DB or NDB rats. However, it significantly improved insulin tolerance test values in diabetic rats. The results suggest that the formulations showed good acceptance and potentially ameliorate insulin resistance both in control group and in animal model of type II diabetes.

Introduction Hibiscus sabdariffa (Hs) has been widely used for weight loss and in the fight against obesity-associated comorbidities. Objective To evaluate the effects of Hs and physical training on the functional capacity of normal-weight and obese rats. Methods Wistar rats were distributed into eight experimental groups: control (C, n = 8) , Hibiscus Sabdariffa (Hs, n = 8), high-intensity interval training (IT, n = 8), high-intensity interval training + Hibiscus Sabdariffa (ITHs, n = 8), obese (O, n = 8), obese + continuous aerobic training (OAT, n = 8), obese + Hibiscus Sabdariffa (OHs, n = 8), , and obese + continuous aerobic training + Hibiscus Sabdariffa (OATHs, n = 8). Hibiscus Sabdariffa extract was administered for 60 days in a dose of 150 mg/kg of body weight. The maximum progressive effort test (MPET) was performed on a treadmill at the beginning and end of the study. The variables analyzed were maximum speed Vmáx time, and distance covered. Lactate was measured immediately after the MPET. Functional capacity was evaluated by the distance/adiposity index. The ANOVA with Bonferroni post hoc and Pearson’s correlation tests were used at a 5% significance level. Results After both types of training, moderate-intensity continuous and high-intensity interval performed on the treadmill, final body weight, weight gain, and the adiposity index decreased, and Vmax, time, and distance covered in the MPET increased, in addition to an improvement in functional capacity. Hs supplementation reduced the adiposity index in normal-weight and obese rats. Hs associated with aerobic training reduced final body weight and increased functional capacity. Conclusion Hs supplementation promoted a reduction in the adiposity index in normal-weight and obese rats and an increase in the functional capacity of trained obese rats. Obesity is associated with other diseases such as diabetes and cancer. Botryosphaeran, a fungal (1→3)(1→6)‐β‐d‐glucan, is described to present antimutagenic, hypoglycemic, hypocholesterolemic, and antitumor activities when administered by gavage over 15 days in rats and mice. Thus, the present study aims to analyze the metabolic effects of Botryosphaeran (12 mg/kg body weight/day) treatment over 30 days in obese Wistar male rats. Obesity was induced in the rats by a high‐fat/high‐sugar diet for 8 weeks. Control rats received a standard diet. On the 5th week, Botryosphaeran treatment commenced. Groups: control, obese, and obese+Botryosphaeran 30 days. In the 8th week, obesity was characterized. Feed intake, glucose and lipid profiles, glucose tolerance, and insulin sensitivity were analyzed. Obese rats showed accumulation of visceral adipose tissue, reduction of muscle mass, glucose intolerance, insulin resistance, hyperglycemia, and dyslipidemia. Botryosphaeran effectively reduced weight gains and the accumulation of retroperitoneal adipose tissue, corrected the levels of glucose, triglycerides, and very low‐density lipoprotein‐cholestrol, and improved insulin sensitivity. Treatment for 30 days was effective in maintaining the beneficial effects demonstrated by this β‐glucan when administered for 15 days without promoting side effects. Treatment with (1→3)(1→6)‐β‐ d‐glucan presented anti‐obesogenic and beneficial metabolic effects in Wistar rats; important for the treatment of obesity and its comorbidities.

Digoxin is a cardiotonic that increases the cardiac output without causing deleterious effects on heart, as well as improves the left ventricular performance during physical exercise. We tested whether the association between chronic digoxin administration and aerobic interval training (AIT) promotes beneficial cardiovascular adaptations by improving the myocardial contractility and calcium (Ca2+) handling. Male Wistar rats were randomly assigned to sedentary control (C), interval training (T), sedentary digoxin (DIGO) and T associated to digoxin (TDIGO). AIT was performed on a treadmill (1h/day, 5 days/week) for 60 days, consisting of successive 8-min periods at 80% and 20% of VO2máx for 2 min. Digoxin was administered by orogastric gavage for 60 days. Left ventricle samples were collected to analysis of Ca2+ handling proteins; contractility and Ca2+ handling were performed on isolated cardiomyocytes. TDIGO group had a greater elevation in fractional shortening (44%) than DIGO, suggesting a cardiomyocyte contractile improvement. In addition, T or TDIGO groups showed no change in cardiomyocytes properties after Fura2-acetoxymethyl ester, as well as in sarcoplasmic reticulum Ca2+-ATPase (SERCA2a), phospholamban and calcineurin expressions. The main findings indicate that association of digoxin and aerobic interval training improved the cardiomyocyte contractile function, but these effects seem to be unrelated to Ca2+ handling.
