Les pesticides organochlorés, considérés comme contaminants environnementaux, sont connus pour induire des perturbations d'un certain nombre de fonctions physiologiques dont la fonction de reproduction. L'impact de l'hexachlorobenzène (HCB) sur la fonction de reproduction mâle chez Meriones unguiculatus a été étudié sur trois lots d'animaux recevant par gavage quotidien 1,6, 4 et 16 mg/kg PC/j pendant 30 j. Sur un plan macroscopique, le HCB entraîne une diminution significative du poids relatif testiculaire, alors que le poids relatif des vésicules séminales ne semble pas être affecté par le traitement. L'étude cytoarchitecturale montre qualitativement une diminution du nombre de spermatozoïdes, suggérant une modification du déroulement de la spermatogenèse et une perturbation de la phase de différentiation particulièrement visible avec la dose la plus forte. L'état de l'activité testiculaire a été estimé par une détermination morphométrique du diamètre des tubes séminifères et par la quantification de l'activité spermatique. Aucune modification significative du diamètre des tubes séminifères n'est observée ; en revanche, l'activité spermatique des testicules des animaux traités avec 16 mg/kg/j est significativement affectée : elle passe de 88 ± 4,89% chez les animaux témoins à 60 ± 3,16%, soit une diminution de l'ordre de 32%. Sur le plan endocrinien, une diminution significative de la testostéronémie est observée chez les animaux des lots II et III (respectivement 0,48 ± 0,08 ng/ml et 0,54 ± 0,07 ng/ml) comparativement à ceux du lot témoin (1,08 ± 0,1 ng/ml), p < 0.01.
L'effet de l'hexachlorobenzene (HCB) sur la fonction hépatique et thyroïdienne a été recherché chez Mériones unguiculatus. Quatre lots d'animaux ont été gavés avec 0, 1,6, 4 et 16 mg/kg/j de HCB pendant 30 j. Le HCB n'a pas d'effet sur le poids corporel. Cependant, il entraîne chez les animaux des deux sexes une hypertrophie hépatique très accentuée, particulièrement chez les animaux traités par la plus forte dose. Des modifications de l'architecture hépatique sont également observées. Une nécrose cellulaire apparaît déjà chez les animaux recevant 1,6 mg/kg/j, accompagnée de congestions centrolobulaires et périportales et de vacuolisations cytoplasmiques chez les animaux traités avec 4 mg/kg/j. Ces mêmes lésions sont accentuées avec la dose la plus forte. L'intoxication au HCB perturbe certaines activités enzymatiques hépatiques. À forte dose, ce pesticide entraîne une augmentation de l'activité ALAT, dont le taux plasmatique est triplé chez les mâles par rapport aux témoins (170 ± 24,7 U/l vs. 52,66 ± 8,29 U/l) et doublé chez les femelles (120 ± 12,47 U/l vs. 56 ± 5 U/l). Ce même traitement augmente l'activité ASAT uniquement chez les femelles (259 ± 29 U/l vs. 244,66 ± 18 U/l). L'impact de cette même dose de HCB sur les hormones thyroïdiennes s'est manifesté par une baisse très significative de la TT4 (21,95 ± 7,46 nmol/l vs. 40,59 ± 1,08 nmol/l) chez les mâles et de la TT3 (1,42 ± 0,11 nmol/l vs. 3,96 ± 0,48 nmol/l) chez les femelles comparativement aux témoins.
Thyroid hormones play a complex role in the toxicity of hexachlorobenzene (HCB) and related compounds. Time-course and dose-response experiments for free- and total thyroxine (T4) and triiodothyronine (T3) plasma levels for thyroid-stimulating hormone (TSH) and thyroid gland histomorphology were determined in male Wistar rats. Also, we examined the possible reversibility of changes noted after removal of HCB. Rats treated with this organochlorine compound resulted in a hypertrophy of the thyroid gland and altered thyroid function by decreasing significantly the levels of total- and free T4 in a dose-dependent manner (total T4: 28 and 51%; free T4: 21 and 37%), and this decrease was seen as early as 21 days and thereafter. Free T3 was also decreased by 21% with the highest dose starting from day 21. No significant changes were observed in the circulating levels of total T3 In response to the decrease of thyroid hormones, a dose-dependent increase of TSH levels (27 and 31%, respectively, for 4 mg and 16 mg/kg of HCB body weight) was observed after 21 days of HCB treatment. We have observed a hypertrophy and hyperplasia of follicular cells and a decrease in colloid volume in histological picture. When HCB was removed and changed by vehicle, the thyroid relative weight and plasma TSH continued to rise and serum thyroid hormones remained suppressed. These findings suggest that subchronic exposure of rats to HCB induced an irreversible hypothyroidism state.
