Breast cancer is a complex disease which is found as the second cause of cancer-associated death among women. Accumulating of evidence indicated that various factors (i.e., gentical and envirmental factors) could be associated with initiation and progression of breast cancer. Diagnosis of breast cancer patients in early stages is one of important aspects of breast cancer treatment. Among of various diagnosis platforms, imaging techniques are main diagnosis approaches which could provide valuable data on patients with breast cancer. It has been showed that various imaging techniques such as mammography, magnetic resonance imaging (MRI), positron-emission tomography (PET), Computed tomography (CT), and single-photon emission computed tomography (SPECT) could be used for diagnosis and monitoring patients with breast cancer in various stages. Beside, imaging techniques, utilization of biochemical biomarkers such as proteins, DNAs, mRNAs, and microRNAs could be employed as new diagnosis and therapeutic tools for patients with breast cancer. Here, we summarized various imaging techniques and biochemical biomarkers could be utilized as diagnosis of patients with breast cancer. Moreover, we highlighted microRNAs and exosomes as new diagnosis and therapeutic biomarkers for monitoring patients with breast cancer.
Background and Objective:
Silica dust is present in almost all mining operations and can cause various health problems such as silicosis in workers. The present study aimed to determine the total and respirable dust levels and the crystalline silica content of the soil in the breathing zone of workers in Iranian mines.
Methods:
This cross-sectional study was carried out in 2016-2017 on six Iranian silica mines. Dust sampling was performed according to the National Institute of Occupational Safety and Health (NIOSH) method No. 0600. The samples were collected from the respiratory zone of workers at 114 stations in different units of the mines. The silica content was measured using the standard NIOSH method No. 7601. The crystalline silica content in bulk samples collected from the soil was determined by using the X-Ray Diffraction (XRD) spectroscopy method.
Results:
The highest concentration of crystalline silica dust was 2.81±0.49 mg/m3 and was observed in the air of crushing unit of mine no. 6, and the lowest was 0.08±0.208 mg/m3 and was measured in the management/administration unit of mine no.1. The mean silica content in the solid surface of the mines was 91%. The total and respirable dust levels in all units of the mines except management/administration were higher than the permissible limit. The free silica content of all collected samples was substantially higher than the permissible limits, and in some cases, it was as much as 100 times above the standard level, which reflects the extremely high risk of working in these mines.
Conclusion:
Exposure of workers with crystalline silica dust in all units was higher than the standard recommended limits. It is imperative to adopt immediate measures based on technical, managerial, and personal protection solutions to reduce the exposure of workers to silica.
Human adipose tissue has been identified as a viable alternative source for mesenchymal stem cells. SADSCs were isolated from human scalp biopsy and then were characterized by Flow cytometry. SADSC expressed CD90, CD44, and CD105 but did not express CD45 surface marker. Growth factors were used for chondrogenesis induction. Histology and immunohistology methods and gene expression by real-time PCR 14 days after induced cells have shown the feature of chondrocytes in their morphology and extracellular matrix in both inducing patterns of combination and cycling induction. Moreover, the expression of gene markers of chondrogenesis for example collagen type II aggrecan and SOX9 has shown by real-time PCR assay. Then, SADSCs were seeded alone on polycaprolatone (PCL) and with Freeze thaw Freeze (PCL+FTF) scaffolds and SADSCs differentiated toward the chondrogenic lineage and chondrogenesis induction were evaluated using scanning electron microcopy (SEM) and MTT assay. Our results showed that SADSCs were also similar to the other adipose-derived stem cells. Using TGF-beta3 and BMP-6 were effective for chondrogenesis induction. Therefore using of TGF-beta3 and BMP-6 growth factors may be the important key for in vitro chondrogenesis induction. The bio-composite of PCL+FTF nanofibrous scaffolds enhance the chondroblast differentiation and proliferation compared to PCL scaffolds .Therefore, our model will make it possible to study the mechanism of transition from chondroblast to chondrocyte.
