Mosses growing on the surface of the Borobudur Temple will affect the aesthetic value of the temple. Interaction between moss and actinomycetes may trigger the growth of moss that can cause an increase in biodeterioration of stone. The purpose of this study was to isolate and identify the actinomycetes associated with moss on the surface of decayed stone of Borobudur Temple as well to assess their ability for phosphate solubilizing. Actinomycetes were isolated using serial dilution method and were identified based on 16S rRNA gene sequences. A total of 37 actinomycetes were isolated from three sampling sites. The isolates found belong to five genera (Gordonia, Microbacterium, Micromonospora, Nocardia, and Streptomyces) and distributed among four families (Microbacteriaceae, Micromonosporaceae, Nocardiaceae, and Streptomycetaceae). Isolates of actinomycetes composed of 19 Streptomyces Group and 18 Rare Actinomycetes Group. Nineteen isolates (51.35%) were identified as genus Streptomyces. Seventeen isolates (45.94%) showed abilities to release soluble phosphate and most of the isolates belong to the genus Streptomyces. The isolates have been collected will be deposited to Indonesian Culture Collection (InaCC) to enrich the collection of actinomycetes from ancient stone in Indonesia and will be used as a source of reference material research, taxonomic, or as source for further study.
Penelitian ini bertujuan untuk mengidentifikasi mikroorganisme (kapang dan bakteri) yang terdapat pada gambar cadas dan berpotensi sebagai biodeteriogen gambar cadas. Metode penelitian meliputi pengambilan sampel dan identifikasi molekuler. Identifikasi molekuler mengunakan primer 9F (5’-GAG TTT GAT CIT IGC TCAG-3’) dan 1510R (5’-GGT TAC CTT GTT ACG ACTT-3’) untuk bakteri, ITS1F (5’-CTT GGT CAT TTA GAG GAA GTAA-3’) dan ITS4R (5’-TCC TCC GCT TAT TGA TAT GC-3’) untuk kapang. Siklus PCR yang digunakan pada penelitian ini sebanyak 35 siklus, meliputi Denaturasi (95oC selama 15 detik), annealing, dan elongation (72oC selama 10 detik). Berdasarkan hasil identifikasi molekuler di peroleh tiga belas jenis mikroorganisme yang terdiri dari empat jenis Bakteri dan sembilan jenis Kapang.
Penelitian ini bertujuan untuk mengetahui efektivitas penggunaan menthol sebagai bahan konsolidasi sementara cagar budaya pada temuan ekskavasi yang rapuh. Pengujian yang dilakukan meliputi karakteristik morfologi menthol, penetrasi kering dan basah menthol pada ketiga jenis sampel (Arang, Bata, Batu), durabilitas konsolidasi sementara menthol pada sampel, dan kuat tekan. Hasil penelitian menunjukkan bahwa proses solidifikasi menthol dimulai pada bagian tepi dan membentuk bentukan seperti jarum (whisker). Penetrasi menthol pada sampel sangat dipengaruhi oleh bentuk dan ukuran partikel sampel serta kandungan air yang terdapat pada sampel. Untuk pengujian durabilitas, pengaruh suhu sangat besar terhadap durabilitas konsolidasi menthol. Semakin tinggi suhu lingkungan, maka semakin cepat pula proses sublimasi menthol. Hasil uji kuat tekan sangat dipengaruhi oleh ukuran partikel sampel, semakin kecil ukuran partikel sampel yang terkonsolidasi menthol, maka semakin tinggi pula nilai kuat tekan yang dihasilkan. Dari hasil penelitian ini dapat disimpulkan bahwa menthol dapat digunakan sebagai bahan konsolidan sementara cagar budaya arang. This study aims to determine the effectiveness of the use of menthol as temporary consolidant for fragile heritage findings on the excavation site. Test includes morphological characteristic of menthol, dray and wet penetration of menthol to three type of sample (charcoal, stone, and brick), durability, and compressive strength. The results show that the menthol solidification process starts at the edges and forms a shape like whiskers. Menthol penetration is strongly influenced by the shape and size of the sample and also water content in the samples. For durability testing, the effect of temperature is very significant on the durability of menthol consolidation. The higher the temperature, the faster the menthol sublimation process will be. Whereas for compressive strength is highly influenced by particle size of the consolidated sample, the smaller consolidated sample the higher compressive strength produced. From this study it can be concluded that menthol can be used as a temporary consolidant material for the fragile heritage findings.

Indonesia mempunyai Benda Cagar Budaya (BCB) yang berlimpah. BCB tersebut dapat mengalami degradasi disebabkan mikroorganisme. Penelitian ini bertujuan untuk memberikan pengetahuan mengenai teknik molecular yang digunakan untuk deteksi biodeteriogen pada benda cagar budaya. Teknik Molekuler yang dapat digunakan adalah Fingerprinting, meliputi DGGE (Denaturing Temperature Gradient Gel Electrophoresis), SSCP (Single Strand Conformation Polymorphism), ARDRA (Amplified Ribosomal DNA Restriction Analysis), dan Clone Library. Teknik ini mempunyai beberapa kelebihan, seperti tingkat presisi yang tinggi, lebih cepat digunakan, dapat digunakan untuk mengetahui peran biodeteriogen pada lingkungan.
