Acrylamide (AA) is a common chemical, produced during food processing and widely used in various industries and laboratory processes. Thus, AA causes a significant risk for human and animal health. Recently published studies have suggested that reproductive toxicity of AA and glycidamide (GA) was mainly due to the oxidative stress which can lead to cell apoptosis. The present experiment was conducted to investigate the effect of oxidative stress on the apoptosis of mouse Leydig (TM3) and Sertoli (TM4) cells induced by AA and its metabolite GA. TM3 and TM4 cells were exposed to AA (10 µM and 1 mM) and GA (1 µM and 0.5 mM) for 24 h. Following the exposure time, the Leydig and Sertoli cells were evaluated for measurement of cell viability, lactate dehydrogenase activity, lipid peroxidation and hydrogen peroxide levels, apoptosis/necrosis rate, and mRNA expression levels of apoptotic genes (caspase3, Bcl-2, Bax, and p53). The present study showed that AA and GA exposure caused decrease in cell viability and increase in excessive oxidative stress and apoptosis in both cell types. In conclusion, our in vitro results demonstrate that oxidative stress probably plays a major role in AA- and GA-induced apoptosis of Leydig and Sertoli cells.

Objective: Autophagy is a form of physiological programmed cell death which is observable after hormonal withdrawal. In this study, the FM3A murine breast tumor cell line was treated with epirubicin alone and with medroxyprogesterone acetate (MPA) or tamoxifen, to determine if structural and kinetic signs of autophagy may also occur in an enhanced manner during epirubicin sensitization via hormonal agents. Methods: One-week soft agar colony growth, 96-hour values of plating and pulse thymidine labeling and electron microscopical examinations were performed following treatment with MPA and tamoxifen alone or with epirubicin. Results: Tamoxifen induced signs of autophagy, which was enhanced when it was combined with MPA. Epirubicin also induced autophagy of secretory granules, which coalesced to form an intracytoplasmic lumen. Combining MPA with epirubicin enhanced the formation of apoptotic blebs and chromatin fragmentation. When epirubicin was combined with tamoxifen, peculiar nuclear structures were formed. Conclusions: Hormonal agents may modulate anthracycline activity towards specific patterns in eliciting cell death, via autophagy and/or as yet unknown nucleolus-specific interactions.

The aim of this study was to determine the total metal accumulation (aluminium, copper, manganese, lead, cadmium and iron) in different organs and eggs of Astacus leptodactylus (Eschscholtz, 1823) and sediments total metal contents (aluminium, copper, manganese, lead, cadmium, iron, zinc, chromium, nickel) in Lake Terkos. Water and sediment samples were collected from two stations at two different depths (1 and 2 m) of Lake Terkos in May 2008. Crayfish samples were collected by trammel net at the same region. Primary hydrographic conditions, such as temperature (13.6-19.4 degrees C), salinity (0.27-0.34 per thousand), dissolved oxygen (7.04-12.30 mg l(-1)) and pH (7.42-8.51), were recorded for each sampling point. Moreover, the total organic carbon (1.65-5.44%) and the total calcium carbonate contents (19.44-41.16%) of sediment samples were determined. According to the Turkish Food Codex (J Zool 26:283-288, 2002), the maximum allowable Pb and Cd levels in crayfish are 0.5 mg/kg wet weight. Accordingly, the Pb and Cd levels determined in A. leptodactylus samples are below this limit. However, when compared with the acceptable metal limits defined by WHO, Australian National Health and Medical Research Council and Ministry of Agriculture in United Kingdom (UK), it is clear that the Cu level is at the limit and the Cd results exceed the limit. When the metal contents in sediment samples from Lake Terkos are examined, it is seen that the Al, Fe, Mn, Ni and Cu contents are lower while Zn, Cr, Cd and Pb contents are higher than the crustal average values. The high values draw attention to the land-based domestic and industrial inputs. Lake Terkos sediments have high enrichment factors (EF) of Zn, Cr, Cd and Pb metals which corroborate this result. The low EFs of Fe, Ni and Cu are due to the natural (terrigeneous) inputs. Additionally, there is no Al, Fe, Ni and Cu metal enrichment in these lake sediments because of the low contamination factor (CF) values. However, it is moderately contaminated by Zn, Cr and Pb, and heavily contaminated by Cd.

Since transcriptome analysis provides genome-wide sequence and gene expression information, transcript reconstruction using RNA-Seq sequence reads has become popular during recent years. For non-model organism, as distinct from the reference genome-based mapping, sequence reads are processed via de novo transcriptome assembly approaches to produce large numbers of contigs corresponding to coding or non-coding, but expressed, part of genome. In spite of immense potential of RNA-Seq-based methods, particularly in recovering full-length transcripts and spliced isoforms from short-reads, the accurate results can be only obtained by the procedures to be taken in a step-by-step manner. In this chapter, we aim to provide an overview of the state-of-the-art methods including (i) quality check and pre-processing of raw reads, (ii) the pros and cons of de novo transcriptome assemblers, (iii) generating non-redundant transcript data, (iv) current quality assessment tools for de novo transcriptome assemblies, (v) approaches for transcript abundance and diferential expression estimations and inally (vi) further mining of transcriptomic data for particular biological questions. Our intention is to provide an overview and practical guidance for choosing the appropriate approaches to best meet the needs of researchers in this area and also outline the strategies to improve on-going projects.
