Purpose Asthenozoospermia is a common cause of human male infertility characterized by reduced sperm motility. The molecular mechanism that impairs sperm motility is not fully understood. This study proposed to identify novel biomarkers by focusing on sperm tail proteomic analysis of asthenozoospermic patients. Methods Sperm were isolated from normozoospermic and asthenozoospermic semen samples. Tail fractions were obtained by sonication followed by Percoll gradient. The proteins were extracted by solubilization and subjected to two-dimensional gel electrophoresis (2-DE); then, the spots were analyzed using Image Master 2D Platinum software. The significantly increased/ decreased amounts of proteins in the two groups were exploited by matrix-assisted laser desorption-ionization time-of-flight/ time-of-flight (MALDI-TOF-TOF) mass spectrometry. Results Three hundred ninety protein spots were detected in both groups. Twenty-one protein spots that had significantly altered amounts (p<0.05) were excised and exploited using MALDI-TOF-TOF mass spectrometry. They led to the identification of the following 14 unique proteins: Tubulin beta 2B; glutathione S-transferase Mu 3; keratin, type II cytoskeletal 1; outer dense fiber protein 2; voltage-dependent anionselective channel protein 2; A-kinase anchor protein 4; cytochrome c oxidase subunit 6B; sperm protein associated with the nucleus on the X chromosome B; phospholipid hydroperoxide glutathione peroxidase-mitochondrial; isoaspartyl peptidase/L-asparaginase; heat shock-related 70 kDa protein 2; stress-70 protein, mitochondrial; glyceraldehyde-3-phosphate dehydrogenase, testis-specific and clusterin. Conclusion Fourteen proteins present in different amounts in asthenozoospermic sperm tail samples were identified, four of which are reported here for the first time. These proteins might be used as markers for the better diagnosis of sperm dysfunctions, targets for male contraceptive development, and to predict embryo quality.
Background:Platelets contain a significant amount of growth factors that have positive effects on local tissue repair and endometrial receptivity.Case:Here we present a 45-yr-old woman with primary infertility and two failed in vitro fertilization (IVF) cycles who was candidate to receive donor eggs. Five consecutive frozen-thawed embryo transfer cycles with good quality embryos were performed within 2 yr. With the diagnosis of recurrent implantation failure (RIF), the patient was treated for improving endometrial receptivity with intrauterine administration of autologous platelet-rich plasma (PRP), 24 hr before embryo transfer. The patient gave birth to a healthy baby boy weighing 2350 gr in the cesarean section.Conclusion:Extensive literature search suggests that this is the first successful pregnancy after administration of PRP in a woman with RIF. Local administration of PRP before embryo transfer may play a vital role in successful implantation .
ObjectiveDiabetes mellitus (DM) is known to cause many systemic complications as well as male infertility. Astaxanthin (ASTX) is a powerful antioxidant that is involved in a variety of biologically active processes, including those with anti-diabetes effects. The present study investigates the effect of ASTX on the spermatozoa function in streptozotocin (STZ)-induced diabetic rats.MethodsWe divided 30 adult rats into three groups (10 rats per group), with a control group that received corn oil mixed with chow. DM was induced by intra-peritoneal injection of STZ. Eight weeks after the STZ injection, half of the diabetic animals were used as diabetic controls, and the rest were treated with ASTX for 56 days. Then the parameters and chromatin integrity of the epididymal sperm were analyzed using chromomycin A3, toluidine blue (TB), and acridine orange (AO) staining.ResultsThe count, viability, and motility of the epididymal sperm were decreased significantly in the STZ group in comparison with the control group (count and viability, p<0.001; motility, p<0.001;0.01). ASTX increased normal morphology and viable spermatozoa compared to the STZ group (morphology, p=0.001; viability, p<0.001;0.05). The percentage of abnormal chromatins in TB and AO staining was higher in the STZ group compared to the control group (p<0.001;0.001). The mean percentage of TB and AO positive spermatozoa in STZ rats was significantly lower in the STZ+ASTX group (TB, p=0.001; AO, p<0.001;0.05).ConclusionThis study observed that in vivo ASTX treatment partially attenuates some detrimental effect of diabetes. Conversely, ASTX improved sperm viability, normal morphology, and DNA integrity.
The follicular fluid (FF) is produced during folliculogenesis and contains a variety of proteins that play important roles in follicle development and oocyte maturation. Age-related infertility is usually considered as a problem that can be solved by assisted reproduction technology. Therefore, the identification of novel biomarkers that are linked to reproductive aging is the subject of this study. FF was obtained from healthy younger (20–32 years old) and older (38–42 years old) women undergoing intracytoplasmic sperm injection (ICSI) due to male factor infertility. The FF was analyzed by two-dimensional gel electrophoresis (2-DE). The power of two-dimensional gel electrophoresis and the identification of proteins were exploited using matrix-assisted laser desorption-ionization time-of-flight/time-of-flight (MALDI-TOF-TOF) mass spectrometry. Twenty three protein spots showed reproducible and significant changes in the aged compared to the young group. Of these, 19 protein spots could be identified using MALDI-TOF-TOF-MS. As a result of MASCOT search, five unique downregulated proteins were identified in the older group. These were identified as serotransferrin, hemopexin precursor, complement C3, C4 and kininogen. A number of protein markers were found that may help develop diagnostic methods of infertility.
Background. Silver nanoparticles (AgNPs) are more often used in various products, and consequently the potential deleterious effects associated with exposure to them are of concern. Several lines of evidence have demonstrated that the toxicity of AgNPs affects different organs and leads to some side effects, including weight loss, inflammation and cell death. Objectives. The aim of this study was to evaluate the effect of different concentrations of AgNPs on sperm parameters and testicular histology. Material and methods. In the present study, 28 male adult Wistar rats were categorized into a control group and 3 experimental groups (AgNP-1, AgNP-2 and AgNP-3), intraperitoneally (i.p.) receiving 30, 125 and 300 mg/kg of AgNPs, respectively. Twenty-eight days after injection the epididymes and the testes of each rat were dissected in order to evaluate sperm parameters, sperm chromatin integrity and histomorphometric changes in the testicular tissue. Results. The results showed a significant decrease in sperm count (p < 0.0001), vitality (p < 0.05) and morphology changes (p < 0.001) in the group receiving 300 mg/kg of AgNPs compared to the control group. A significant decrease was also observed in the number of spermatogonia, Sertoli and Leydig cells in the AgNP-2 and AgNP-3 groups (p < 0.05). The evaluation of sperm chromatin did not show any significant differences among the experimental groups (p > 0.05). Conclusions. The data showed some dose-dependent adverse effects of AgNPs on sperm and seminiferous tubules. More experimental investigations are necessary to draw better conclusions regarding the safety of nanoparticles (NPs) on the male reproduction system.
High doses of anabolic-androgenic steroids (AAS) are used by some athletes to increase muscle mass, that is often associated with male infertility. The aim of this study was to investigate the possible cause/s of male infertility using a rat model by analysing sperm quality, including its protamine content and DNA integrity, as well as pregnancy rate. Five groups of male Wistar rats were treated for 10 weeks as follows: nandrolone decanoate (10 mg kg(-1) per week) (ND); running exercise (50 min per day, 5 days a week) (EX); Combination of ND and exercise (ND-EX); nandrolone decanoate solvent (Sham); and control without any injection or exercise (CO). Deterioration in sperm quantity was observed in all test groups (P ≤ 0.01). The frequency of fertile rats was decreased in the ND-EX and ND groups (P ≤ 0.05). Chromomycin-A3 staining showed a protamine deficiency in the epididymal spermatozoa in the ND-EX rats (P ≤ 0.05). Chromatin analysis indicated an abnormal maturation of the sperm nuclei in all test groups compared with the controls (P ≤ 0.05). TUNEL analyses showed a highly significant increase in apoptosis in the EX, ND, and ND-EX groups (P ≤ 0.01). Our data show that a combination of exercise and high doses of nandrolone decanoate negatively influences the DNA integrity and protamine content resulting in lower sperm quality and reduced pregnancy rate.
Background: Multiple sclerosis (MS) is a chronic, idiopathic and the most common inflammatory demyelinating disease of the central nervous system that progresses to severe disability. Prevalence of MS is 2-150 per 100,000 people. The main cause of the disease is not clear and occurs due to a combination of both genetic and environmental factors. Objectives: The purpose of this study was to determine the prevalence rate of MS in the northern cities of Khuzestan Province, Iran, as well as to determine the demographic characteristics, initial symptoms at onset, clinical features and the correlation between the above factors with each other. Patients and Methods: In this descriptive correlational study, patients were recruited through the MS Society as well as the neurology clinics and departments of hospitals in north cities of Khuzestan Province (Iran). Data were collected through completing a face to face questionnaire and were analyzed using SPSS version 17 and the chi-square test. P value < 0.05 was considered statistically significance. Results: A total of 142 patients were participated in this study. Prevalence of MS was 15 per 100,000 people. Seventy-eight percent of the patients had low literacy, and 81% were unemployed. Female to male ratio was 2.08: 1 that became less in younger patients. 69% and 15% of the patients were relapsing remitting and progressive relapsing, respectively. Demographic characteristics of the patients did not show any statistically significant relationship with clinical courses and signs at onset; however, a significant correlation was seen between severe disability and sex (P = 0.001) as well as between severe disability and progressive-relapsing subtype (P = 0.02). Conclusions: The prevalence of MS showed an increasing rate of the disease in recent years. Decreasing of the female to male ratio and patients' low socioeconomic level, need more studies. The increasing rate of patients with progressive relapsing feature and more disabled patients belong to this subtype show the poor prognosis.
