Microbial infection by bacteria has caused severe health problems worldwide. Treatment with antibiotics as the current solution has several drawbacks and triggers the phenomenon of bacterial resistance. Therefore, there is an urgency to look for a natural antimicrobial that is safer and has fewer side effects. One of the most promising antibacterial agents is Spirulina platensis. This research was conducted to evaluate the antibacterial activity of microalgae S. platensis against Propionibacterium acne, Staphylococcus epidermidis, and Enterobacter aerogenes and identify compounds from the active fraction of microalgae. Biomass was extracted with ethanol 96% using the reflux method then partitioned with immiscible solvents such as hexane, ethyl acetate, and water. Partial purification was carried out by chromatography techniques such as thin-layer chromatography and column chromatography. The compounds of active fractions were identified by GC-MS analysis. The result showed that ethyl acetate extract had vigorous antibacterial activity against all tested bacteria. The highest activity (14.4 ± 0.63 mm and 16.9 ± 1.48 mm) was achieved against P. acne; followed by S. epidermidis (13.05± 0.14 mm and 13.15 ± 0.0 mm), and E. aerogenes (11.7 ± 2.05 mm and 12.6 ± 1.90 mm), at concentrations 20,000 ppm and 30,000 ppm, respectively. The results indicated that the extract is more sensitive to Gram-positive bacteria (P. acne and S. epidermidis) than Gram-negative bacteria (E. aerogenes). Purification of the extract resulted in fraction 2 and fraction 6 as the most potential fractions for further analysis and identification. Based on the antibacterial activity, inhibition zones of fractions are wider than extracts. It could be assumed that the purification process enhances the activity of a sample. GC-MS analysis revealed that the dominant compounds of fractions 2 and 6 were bis (2-ethylhexyl) phthalate (67.76%) and 1,2-Benzendicarboxilic acid, bis (2-ethylhexyl) ester (50,88%), respectively. This result indicated that the ethyl acetate fraction of the microalgae S. platensis has the potential as a natural antibacterial.
ABSTRAK: Bakteri merupakan salah satu penyebab penyakit kulit wajah, terutama jerawat. Timbulnya jerawat dikarenakan tersumbatnya pori kulit oleh lemak, sehingga memunculkan adanya komedo. Gejala lebih lanjut, dapat menimbulkan peradangan yang disebabkan infeksi bakteri yang memunculkan penyakit jerawat pada kulit wajah. Studi terdahulu mengemukakan flavonoid yang dimiliki Chroococcus turgidus berpotensi sebagai antibakteri dan antioksidan. Penelitian ini bertujuan untuk mengetahui kemampuan data hambat terhadap bakteri Propionibacterium acnes dan Staphylococcus epidermidis pada sediaan masker peel-off yang mengandung ekstrak etanol C. turgidus dengan variasi konsentrasi 0%, 2%, 3% dan 4%. Evaluasi sediaan yang dilakukan meliputi organoleptik, pH, viskositas, homogenitas, waktu sediaan mengering, daya sebar, sifat mekanik, uji hedonik, dan uji stabilitas cycling test. Hasil penelitian menunjukkan semua formulasi sediaan masker peel-off mempunyai aktivitas antibakteri terhadap bakteri P. acnes dan S. epidermidis. Semakin tinggi ekstrak etanol C. turgidus yang ditambahkan ke dalam sediaan masker peel-off, zona hambat yang terbentuk pada kedua bakteri uji semakin besar. Zona hambat yang terbentuk sesudah cycling test terhadap P. acne adalah 10,15 cm (2%), 11,02 cm (3%) dan 20,34 cm (4%), sedangkan terhadap bakteri S. epidermidis zona hambat yang terbentuk adalah 8,01 cm (2%), 10,60 cm (3%) dan 15,14 cm (4%). Oleh karena itu, ekstrak etanol dari C. turgidus dapat digunakan sebagai salah satu senyawa bioaktif yang berpotensi sebagai antibakteri pada sedian masker peel-off.
ABSTRAK.Fikobiliprotein merupakan kompleks pigmen-protein yang dimiliki oleh sianobakteria dan menunjukkan berbagai aktivitas biologi yang luas. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan toksisitas hayati dari pigmen fikobiliprotein Chroococcus turgidus yang diekstrak menggunakan berbagai pelarut polar, yaitu air, kalsium klorida dan buffer fosfat. Uji aktivitas antioksidan dilakukan dengan metode perendaman radikal bebas DPPH sedangkan uji toksisitas hayati dengan BSLT. Hasil penelitian menunjukkan bahwa pigmen fikobiliprotein berhasil diekstraksi menggunakan ketiga macam pelarut. Hasil terbaik dicapai oleh pelarut air dengan kadar pigmen 0,296 mg/mL. Berdasarkan uji aktivitas, pigmen fikobiliprotein yang diekstrak menggunakan pelarut air bersifat aktif sebagai antioksidan dengan nilai IC 50 sebesar 198,706 µg/mL, sedangkan hasil ekstraksi menggunakan kalsium klorida dan buffer fosfat tidak aktif sebagai antioksidan dengan nilai IC 50 berturut-turut adalah 1255,430 µg/mL dan 1508,130 µg/mL. Hasil uji toksisitas menunjukkan pigmen fikobiliprotein yang diekstrak menggunakan air tidak bersifat toksik dengan nilai LC 50 sebesar 1920,430 µg/mL sedangkan pigmen yang diekstrak menggunakan kalsium klorida dan buffer fosfat bersifat toksik dengan nilai LC 50 berturut-turut sebesar 534,070 µg/mL dan 221,050 µg/mL. Hasil penelitian ini membuktikan bahwa pigmen fikobiliprotein sianobakteria C. turgidus dapat dimanfaatkan sebagai antioksidan alami. Aktivitas toksisitas yang dimilikinya memberikan gambaran untuk pengujian lebih lanjut ke arah potensi dan seleksi senyawa antikanker.
