Generally, floral papillae in Maxillaria have little taxonomic value. Nevertheless, the absence of papillae from members of the M. cucullata alliance, the occurrence of clavate papillae with distended apices in the M. rufescens alliance and the presence of papillose trichomes in some species may yet prove to be useful.
Pseudopollen is found on the labella of several species of Maxillaria and is formed by the fragmentation of uniseriate, multicellular, moniliform trichomes. The resultant component cells are rich in protein. Since flowers of pseudopollen‐forming species generally lack nectar, it is probable that pseudopollen gradually replaced nectar as the pollinator reward. However, direct evidence for this hypothesis is lacking. The present paper examines the labellar micromorphology of five Brazilian species of Maxillaria assigned to the M. discolor alliance. The flowers of two of these species, namely M. violaceopunctata and M. villosa, produce both food hairs and a lipoidal, labellar secretion which is rich in aromatic amino acids. Moreover, in the case of M. violaceopunctata, the secretion was found to contain reducing sugars. This may represent an intermediate stage in the transition from nectar‐producing to pseudopollen‐forming flowers. Labellar food-hairs in Polystachya Hook. exhibit considerable morphological diversity. The commonest type of trichome is uniseriate, two to four-celled, with a clavate or subclavate terminal cell. This type occurs amongst representatives of most sections examined. Other trichomes are bristle-like with tapering or fusiform terminal cells, whereas representatives of section Polystachya have uniseriate, moniliform trichomes that fragment with the formation of rounded or elliptical component cells. Most contain protein and, while some contain starch, lipid is invariably absent. The presence of particular types of labellar trichomes does not coincide with variations in vegetative morphology. Thus, current taxonomic treatment of the genus indicates that trichome types, with perhaps the sole exception of moniliform, pseudopollen-forming hairs found in section Polystachya only, have limited taxonomic value. However, the remarkable similarity between pseudopollen-forming hairs of Polystachya and those of the Neotropical genus Maxillaria in terms of morphology, cellular dimensions and food content indicates that pseudopollen may have arisen several times and evolved in response to similar pollinator pressures.
The labella of Maxillaria acuminata Lindl., M. cerifera Barb. Rodr. and M. notylioglossa Rchb.f., all members of the M. acuminata alliance, produce a viscid wax-like secretion. Histochemical analysis revealed that the chemical composition of the secretion is similar in all three species, consisting largely of lipid and protein. Light microscopy and low-vacuum scanning electron microscopy were used to investigate the secretory process. In a fourth taxon, M. cf. notylioglossa, transmission electron microscopy showed that lipid bodies are associated with smooth endoplasmic reticulum or occur as plastoglobuli within plastids. Lipid bodies vary in appearance and this may reflect differences in chemical composition. They become associated with the plasmalemma and eventually accumulate between the latter and the cell wall. The wall contains no pits or ectodesmata, and it is speculated that lipid passes through the wall as small lipid moieties before eventually reassembling to form lipid globules on the external surface of the cuticle. These globules are able to coalesce forming extensive viscid areas on the labellum. The possible significance of this process to pollination is discussed.
Aims: To determine the effects of fresh and freeze-dried extracts of Allium sativum on the physiology and morphology of Candida albicans. Methods and Results: Inhibition of growth in glucose-yeast extract-peptone was measured using a multiwell plate reader. Scanning and transmission electron microscopy investigations indicated loss of structural integrity. Gas chromatography-mass spectrometry of extracts was employed to separate and quantify putative inhibitory sulphur-containing components. Conclusions: Fresh garlic extract has a greater efficacy than garlic powder extract as indicated both by its effects on morphology and inhibition of growth. Significance and Impact of the Study: The ubiquitous opportunistic pathogen C. albicans is sensitive to garlic; resistance to the broad spectrum of active principles present is unlikely so that its anticandidal effects may provide an important alternative route to chemotherapy.
Whole garlic (Allium sativum L.) extract and some of its components were assayed for antigiardial activity. Whole garlic extract gave an IC 50 at 24 h of 03 mg ml N1 . Most of the components assayed were inhibitory to the organism, especially allyl alcohol and allyl mercaptan, with IC 50 values of 7 µg ml N1 and 37 µg ml N1 respectively. Studies with calcofluor white indicated that whole garlic and allyl alcohol collapse the transmembrane electrochemical membrane potential (∆ψ) of the organism, as indicated by uptake of the fluorochrome. Electron microscopy allowed the morphological changes that occur with garlic inhibition to be recorded. Both the surface topography and internal architecture of the organism changed during incubation with the biocides. Both whole garlic and allyl alcohol resulted in fragmentation of the disc and an overexpression of disc microribbons, internalization of flagella, vacuole formation and an increase in distended vesicles. Allyl mercaptan, however, only gave an increase in distended vesicles, suggesting that this biocide has a different mode of action.
A role for the cAMP-dependent pathway in regulation of the cell wall in the model yeast Saccharomyces cerevisiae has recently been demonstrated. In this study we report the results of a phenotypic analysis of a Candida albicans mutant, characterized by a constitutive activation of the cAMP pathway due to deletion of PDE2, the gene encoding the high cAMP-affinity phosphodiesterase. Unlike wild-type strains, this mutant has an increased sensitivity to cell wall and membrane perturbing agents such as SDS and CFW, and antifungals such as amphotericin B and flucytosine. Moreover, the mutant is characterized by an altered sensitivity and a significantly reduced tolerance to fluconazole. The mutant's membrane has around 30% higher ergosterol content and the cell wall glucan was 22% lower than in the wild-type. These cell wall and membrane changes are manifested by a considerable reduction in the thickness of the cell wall, which in the mutant is on average 60-65 nm, compared to 80-85 nm in the wild-type strains as revealed by electron microscopy. These results suggest that constitutive activation of the cAMP pathway affects cell wall and membrane structure, and biosynthesis, not only in the model yeast S. cerevisiae but also in the human fungal pathogen C. albicans.
