A mandioca (Manihot esculenta Crantz) é uma planta arbustiva que apresenta uma ampla diversidade genética e várias são as técnicas existentes para a avaliação dessa diversidade, dentre estas destaca-se os marcadores moleculares. Diante disso, objetivou-se neste estudo caracterizar etnovariedades de mandioca cultivadas no norte do estado de Mato Grosso, por meio de marcadores moleculares ISSR. Para a extração do DNA vegetal das 15 etnovariedades de mandioca, utilizou-se o método CTAB e as amplificações foram via PCR com 15 primers ISSR. Nas análises estatísticas utilizou-se o programa Genes. Foram amplificados um total de 159 fragmentos, dos quais 67,92% foram polimórficos. O PIC variou entre 0,00 e 0,62, enquanto a dissimilaridade genética estimada pelo coeficiente de Jaccard, variou entre 0,08 a 0,39. O dendrograma obtido pelo método de agrupamento UPGMA formou três grupos, já pelo método de Tocher houve a formação de sete grupos, ambos demonstrando que há diversidade genética entre as etnovariedades analisadas. Dessa forma, é possível concluir, que os primers utilizados foram eficientes na detecção de polimorfismo e que há diversidade genética entre as etnovariedades de M. esculenta.
The characterization of ethnovarieties of cassava cultivated by family farmers is essential for the selection of superior genotypes in breeding programs of the species. The aim of this study was to evaluate the genetic variability of 15 ethnovarieties of cassava cultivated in the municipality of Alta Floresta, Mato Grosso, via the morphological descriptors of the flowers. For the characterization, eight qualitative and quantitative morphological descriptors were used, as described for the species Manihot esculenta. The manioc flowers (female and male) showed variability in the color of the ovary, disc and sepals. Female flowers have varied sizes, from 7.12 to 9.03 mm in length and in width from 2.80 to 3.72 mm. For the male flowers, the variation in length was from 6.43 to 8.12 mm and width from 2.61 to 3.61 mm. The UPGMA grouping, using a cut-off point of 75.74%, allowed for the formation of two genetic groups, of which the first group (GI) was composed of 86.67% of the evaluated ethnovarieties and the second group (GII) was composed by 13.33%. The floral characteristics were efficient for use in phenotypic differentiation and show genetic variability among the ethnovarieties.
<p class="Normal1"><span>O cultivo da mandioca se destaca na agricultura brasileira pelo fácil manuseio, adaptação edafoclimáticas e desempenho produtivo satisfatório. Diante disso, esse trabalho teve como objetivo estimar a divergência genética entre 17 etnovariedades de mandioca e selecionar, com base no desempenho agronômico, as etnovariedades que possam ser utilizadas em futuros programas de melhoramento genético com a espécie. As etnovariedades de mandioca foram avaliadas com base em sete descritores agronômicos quantitativos. Os descritores foram submetidos a análise de variância e agrupados pelo teste de Scott e Knott, ao nível de significância de 5% de probabilidade. A técnica de análise multivariada foi empregada para avaliar a divergência genética entre as etnovariedades, baseada na distância generalizada de Mahalanobis. Todas as análises foram realizadas com auxílio do programa Genes. A Análise de variância mostrou diferença significativa, pelo teste de F, e a análise de agrupamento possibilitou a detecção de variabilidade genética entre as etnovariedades de mandioca por meio de sete características quantitativas. Portanto, houve divergência genética entre as 17 etnovariedades de mandioca cultivadas no município de Alta Floresta, Mato Grosso, Brasil.</span></p>
