Two outbreaks of contagious agalactia by Mycoplasma agalactiae occurred in Paraíba State, Northeastern Region of Brazil are reported. The disease was characterized by mastitis, agalactia and polyarthritis in does and polyarthritis and conjunctivitis in kids and lambs. Fever and anorexia were also observed. Morbidy was from 26.1% to 100% in does, 36.5 to 100% in kids and 49% in lambs. In one farm 14.3% of the lactating goats and 6.4% of the kids died or were euthanized. In the other, 3.3% of the does, 36.5% of the kids and 22.9% of the lambs died and 84 affected goats were euthanized to control the disease. M. agalactiae was isolated from milk, joint exudates, nasal swabs and ear washings. The colonies were characteristic of Mycoplasma and the agent did not ferment both glucose and arginin. It was typed as Mycoplasma agalactiae by immunoperoxidase and PCR. This is the first report of M. agalactiae infection in Brazil, but the source of the infection remains unknown.
Newcastle disease is characterized by respiratory manifestations in association with nervous and/or digestive symptoms. Its prevention is done by vaccination with live attenuated (lentogenic strains) and/or killed vaccines. The lentogenic strains can lead to strong post-vaccination reaction, principally due to the presence of other pathogenic agents. Among them, Mycoplasma synoviae is worldwide important, mainly in Brazil. The dissemination of this agent in poultry flocks has been achieved due to difficulties in diagnosis and disease reproduction, virulence variations among different M.synoviae strains, and attribution of typical M.synoviae disease manifestation to other disease agents. This experimental study in SPF chicks (Gallus gallus), previously infected by M.synoviae and thereafter vaccinated against Newcastle disease, was done with the objective of evaluating M.synoviae pathogenicity through assessment of post-vaccinal respiratory reactions and serologic responses to Newcastle disease virus vaccine in the absence of environmental factors. A total of 86 three days old chicks were used, being 57 infected by eye and nostril drop, with chicken activated M. synoviae strain WVU 1853. Seven days later, 21 mycoplasma infected birds plus 29 not mycoplasma infected ones were vaccinated against Newcastle disease. As results, the not infected and vaccinated birds yielded, significantly, higher and longer lasting serologic responses to Newcastle disease vaccine virus than those infected and vaccinated. Similarly, the infected and vaccinated birds yielded lower serologic reactions to M.synoviae than those only mycoplasma infected. No post-vaccinal respiratory reaction was observed in the vaccinated birds.
Mycoplasmas were searched in the ear canal flushing of 60 bovine in Brazil. The prevalence obtained was 80%. The percentages of typified species were 12.5%, for M. alkalenses; 2.1%, M. arginini; 8.35%, M. bovirhinis; 2.1%, M. bovis; 25.0%, M. conjunctivae; 14.6%, M. mycoides subsp. mycoides LC and 10.4% M. capricolum.
This work was conducted on rats in two premises located in Niterói and Rio de Janeiro, Brazil. One is classified as conventional controlled and the other, conventional. The objective of the present study was to detect the presence of Mycoplasma pulmonis in animals with symptoms of respiratory disease and low reproductive performance. In the conventional controlled premises, 16 rats of Wistar-Furth strain were necropsied while in the conventional premises necropsy was performed on 12 rats of Hooded Lister strain. The clinical samples of lungs, trachea, oropharynx, middle ear, uterus and ovaries were subjected to culturing while the sera were tested for antibody detection. From 28 rats, 57.14% (16/28) were culture positive for M. pulmonis, being 81.25% (13/16) from the conventional controlled premises, and 25.00% (3/12) from the conventional premises. The ELISA test was carried out in 20 animals of both colonies. In the conventional controlled premises, 92.86% (13/14) were positive for M. pulmonis, and 7.14% (1/14) were suspicious, while in the conventional premises, 100% (6/6) of the samples were positive. The results confirmed that M. pulmonis was the etiologic agent of the disease that affected the rats under study, and that the ELISA positivity rated higher than culture.
RESUMOO uso de animais em experimentos biomédicos com padrão genético e sanitário definido fornece ao pesquisador, modelos importantes para a compreensão de mecanismos biológicos. Entretanto, nesses modelos os ratos podem se infectar por agentes patogênicos, como os Micoplasmas, os quais introduzidos na criação, afetam a produção e os resultados experimentais. Este estudo foi desenvolvido em ratos, positivos para Mycoplasma pulmonis, obtidos de dois biotérios, um em Niterói e outro no Rio de Janeiro. Um foi classificado como convencional controlado e o outro como convencional. O objetivo foi caracterizar as lesões encontradas em pulmões e traquéias de animais que apresentavam sintomas de doença respiratória e baixa produtividade. No biotério convencional controlado foram necropsiados 16 ratos da linhagem Wistar-Furth e no biotério convencional, 12 ratos da linhagem Hooded Lister. As amostras de pulmão, traquéia e útero foram submetidas a análise anatomopatológica. No exame anatomopatológico, os animais estudados apresentavam graus variados de lesões, basicamente, nos pulmões e na traquéia. Dentre os diferentes graus de lesões macroscópicas observados nesses órgãos, as lesões discretas (46,43% e 25,00%, respectivamente) e acentuadas (46,43% e 32,15%, respectivamente) foram as mais encontradas em ambos os órgãos enquanto que, no exame microscópico, as lesões mais encontradas foram as acentuadas nos pulmões (64,70%) e, discretas (41,18%) e moderadas (41,18%) nas traquéias. Para os padrões sanitários mencionados e, na ausência de outros métodos, o exame anatomopatológico pode ser utilizado para o diagnóstico da Micoplasmose Respiratória Murina.
RESUMOA mastite subclínica caprina ocasiona prejuízos econômicos em decorrência do descarte, dos gastos com medidas terapêuticas e da redução da quantidade e qualidade do leite e seus derivados. Nesse estudo, 129 amostras de leite de cabra in natura, provenientes de 11 propriedades foram avaliadas pelo teste da caneca telada, California Mastitis Test (CMT), exame bacteriológico, pesquisa de Mycoplasma spp. e pela determinação dos parâmetros físico-químicos. No teste da caneca telada e no CMT, 3,1% e 4,6% das amostras foram positivas, respectivamente. No exame bacteriológico, 57,4% das amostras foram positivas e o patógeno mais frequente foi Staphylococcus coagulase negativa com 56% das cepas resistentes à penicilina e 100% de sensíveis à gentamicina. Mycoplasma spp. não foi identificado nas amostras. O diagnóstico da mastite subclínica pelo CMT e pelo exame bacteriológico diferiu de forma significativa e não houve associação entre o número de UFC/mL obtidas no exame microbiológico e o resultado do CMT (Qui-quadrado p < 0,05). Os parâmetros físico-químicos diferiram significativamente entre os rebanhos (ANOVA, TukeyKramer, p < 0,05) e a gordura foi o constituinte que demonstrou maior amplitude, sendo que 63,4% dos rebanhos apresentaram os valores abaixo do exigido pela legislação brasileira. Não houve associação significativa entre a presença de mastite diagnosticada pelo exame bacteriológico e os valores obtidos para os parâmetros físico-químicos (t-Student p > 0,05). Com base nos resultados obtidos, recomenda-se a associação do exame bacteriológico quando na utilização do CMT para diagnóstico da mastite subclínica caprina. ABSTRACT MAJOR CHANGES IN MILK BY MASTITIS CAUSATIVE AGENTS IN GOAT HERD FROM THE STATES OF MINAS GERAIS AND RIO DE JANEIRO, BRAZIL.Goat mastitis causes significant economic losses due to the discarding of milk, costs of drugs and veterinary care, reducing the quantity and quality of milk and dairy products. In this study, 129 raw milk samples from 11 goat farms were investigated by the Tamis test, California mastitis test (CMT), bacteriological exam, presence of Mycoplasma spp. and physicochemical parameters. Seven (4.6%) and four samples (3.1%) were positive by CMT and Tamis test respectively. Bacteriological exam was positive from 57.4% of samples and coagulase-negative Staphylococcus was the most frequent bacteria isolated showing 56% of the strains resistant to penicillin and no resistance to gentamicin. Negative results were obtained from traditional culture as well as by PCR for Mycoplasma spp. The diagnosis of mastitis, the bacteriological exam and the CMT results differed significantly and no association was observed (chi squared, p < 0.05). Fat was the constituent with more variations and 63.4% of the herds did not meet the minimum requirement according to Brazilian government criteria for this constituent.
