Introdução: Os medicamentos psicotrópicos alteram o funcionamento do sistema nervoso central, causando modificações no estado mental do usuário. Este tipo de medicação é utilizado para o combate da ansiedade, agitação, insônia, angústia e depressão. Houve aumento considerável no uso desses medicamentos, em todo o mundo, nos últimos anos, assim como, no Brasil. Objetivo: Verificar o porquê do aumento do uso abusivo de medicamentos psicotrópicos e suas consequências na saúde. Material e métodos: Este estudo tratou-se de uma revisão bibliográfica sistemática. Utilizou-se as bases de dados Scielo e Google Acadêmico, com descritores “Medicamentos psicotrópicos” e “Abuso de medicamentos psicotrópicos” em inglês e português, sem recorte temporal, com a exclusão daquelas que não apresentavam informações sobre o tema. Resultados: O uso de psicotrópicos aumentou consideravelmente nas últimas décadas devido ao ingresso de novos psicofármacos no mercado farmacêutico, renovação automática de receitas e prescrição excessiva destes medicamentos, já que aliviam os sintomas mais rapidamente do que outras abordagens com efeito à longo prazo. Entretanto, os psicotrópicos não levam à cura, necessitando de uso continuo, causando dependência física e psíquica, diminuição da memória, atenção, força muscular e potência sexual, condições que podem acentuar a ansiedade ou a depressão, criando um ciclo negativo. E, uma vez dependente, a suspensão deve acontecer de forma lenta, necessitando de apoio psicológico e psiquiátrico. Esses medicamentos causam tolerância, levando à necessidade de dosagens cada vez maiores, podendo induzir à automedicação e uso abusivo, com aumento das doses ingeridas, para atingir o efeito desejado. Conclusão: O uso abusivo de medicamentos psicotrópicos aumenta constantemente por proporcionar rapidez no alívio dos sintomas e ter muitos incentivos da indústria farmacêutica. Assim, estes são utilizados mesmo nos casos em que o tratamento psicológico à longo prazo é mais recomendado, trazendo riscos à saúde, como dependência e tolerância, resultando no uso abusivo.
O LES desenvolve-se a partir da interação entre fatores ambientais, como exposição ao sol, estresse físico ou emocional, e fatores imunogenéticos. O sistema imunológico tem como função o desenvolvimento de respostas imunológicas a partir da detecção de antígenos. Este processo ocorre em duas etapas, sendo: apresentação de antígenos, através de moléculas de histocompatibilidade (HLA, do inglês human leucocytes antigen), e detecção e interpretação destes por linfócitos T. Assim, o objetivo deste trabalho foi descrever o papel das moléculas de HLA no desenvolvimento de LES. Polimorfismos nas moléculas de HLA podem influenciar no processo de autotolerância de linfócitos T e, consequentemente, desencadear resposta inflamatória contra o próprio organismo, característico de doenças autoimunes. Desta forma, pesquisas imunogenéticas são de suma importância para melhor compreensão da doença, pois fornecem informações sobre marcadores genéticos associados ao LES, proporcionando a análise de fatores de risco ou proteção à doença, entre diferentes etnias, auxiliando na identificação de marcadores genéticos universais e contribuindo, assim, para diagnóstico e tratamento mais precoces.
Introdução: A leucemia é um tipo de câncer que afeta os leucócitos do sangue, comprometendo o sistema imunológico pelo acúmulo de células imaturas na medula óssea e corrente sanguínea. Existem vários tipos de leucemia, diferenciados pelo gênero celular acometido, velocidade de progressão da doença e anormalidades cromossômicas presentes. Objetivo: Descrever a fisiopatologia da leucemia e o papel das anormalidades cromossômicas. Material e métodos: Este estudo tratou-se de uma revisão bibliográfica. Utilizou-se as bases de dados Scielo, PubMed e Google Acadêmico, com os descritores “Leucemia”, “Citogenética” e “Translocação cromossômica”, em inglês, português e espanhol com recorte temporal de dez anos. Resultados: As leucemias ocorrem a partir de mutações genéticas e/ou epigenéticas em leucócitos imaturos. Essas mutações ocasionam erros na produção de proteínas e perda de sinais regulatórios que controlam a divisão celular e apoptose, originando células metastáticas com crescimento desregulado. Consequentemente, há acúmulo de leucócitos imaturos na medula óssea e na corrente sanguínea, prejudicando o sistema imunológico e a formação de elementos hematopoiéticos essenciais. As leucemias são geralmente classificadas em quatro tipos principais, a partir do gênero celular acometido (linfocítica ou mieloide) e velocidade de progressão da doença (aguda ou crônica). Entretanto, existem divergências clínicas e laboratoriais mesmo entre leucemias de um mesmo tipo, devido a diferentes anormalidades cromossômicas observadas. Nos trabalhos analisados notou-se uma grande variabilidade de alterações cromossômicas associadas a cada tipo de leucemia. Pacientes que apresentaram anormalidades cromossômicas definidas, receberam tratamentos personalizados, determinando maior qualidade e expectativa de vida. Conclusão: Todos os tipos de leucemia têm em comum o crescimento desregulado de leucócitos imaturos que se acumulam na medula óssea, prejudicando o sistema imunológico e a formação de elementos hematopoiéticos essenciais. Pacientes que recebem tratamentos personalizados a partir de análises citogenéticas demonstram maior qualidade e expectativa de vida.
MicroRNAs são pequenos fragmentos de RNA, incapazes de codificar proteínas e que participam da regulação pós-transcricional da expressão gênica ao se ligarem em sequências do RNA-mensageiro alvo, induzindo a degradação ou prevenindo a tradução destas sequências. A comprovação de estudos recentes de que alguns microRNAs possam estar envolvidos com a expressão de transportador associado a processamento de antígenos (TAP)-1, pode ser mais um passo no entendimento de diversos mecanismos patológicos como o câncer, por exemplo, auxiliando, assim, em imunoterapias mais eficientes. Dependendo da célula-alvo, os microRNAs são capazes de influenciar diversas etapas da formação tumoral, desde a metástase, progressão da doença, até a frequência de infiltração de células imunes no ambiente tumoral. Essas moléculas também parecem estar envolvidas nos mecanismos de escape imunológico. O objetivo deste artigo foi buscar na literatura a relação entre os microRNAs e a expressão da proteína TAP, descrever a função dessas moléculas, especialmente em patologias como o câncer, sua influência nos mecanismos de evasão imunológica e suas possíveis contribuições para o desenvolvimento de novas tecnologias terapêuticas e diagnósticas.
