Background Breast cancer is highly prevalent among women worldwide. It is classified into three main subtypes: estrogen receptor positive (ER+), human epidermal growth factor receptor 2 positive (HER2+), and triple negative breast cancer (TNBC). This study has evaluated the effects of aspirin and metformin, isolated or in a combination, in breast cancer cells of the different subtypes. Methods The breast cancer cell lines MCF-7, MDA-MB-231, and SK-BR-3 were treated with aspirin and/or metformin (0.01 mM - 10 mM); functional in vitro assays were performed. The interactions with the estrogen receptors (ER) were evaluated in silico. Results Metformin (2.5, 5 and 10 mM) altered the morphology and reduced the viability and migration of the ER+ cell line MCF-7, whereas aspirin triggered this effect only at 10 mM. A synergistic effect for the combination of metformin and aspirin (2.5, 5 or 10 mM each) was observed in the TNBC cell subtype MDA-MB-231, according to the evaluation of its viability and colony formation. Partial inhibitory effects were observed for either of the drugs in the HER2+ cell subtype SK-BR-3. The effects of metformin and aspirin partly relied on cyclooxygenase-2 (COX-2) upregulation, without the production of lipoxins. In silico, metformin and aspirin bound to the ERα receptor with the same energy. Conclusion We have provided novel evidence on the mechanisms of action of aspirin and metformin in breast cancer cells, showing favorable outcomes for these drugs in the ER+ and TNBC subtypes.
The present study aimed to characterize the effects of quinoxaline-derived chalcones, designed on the basis of the selective PI3Kγ inhibitor AS605240, in oral cancer cells. Three lead compounds, namely N9, N17 and N23, were selected from a series of 20 quinoxaline-derived chalcones, based on an initial screening using human and rat squamous cell carcinoma lineages, representing compounds with at least one methoxy radical at the A-ring. The selected chalcones, mainly N9 and N17, displayed marked antiproliferative effects, via apoptosis and autophagy induction, with an increase of sub-G1 population and Akt inhibition. The three chalcones displayed marked in vitro antitumor effects in different protocols with standard chemotherapy drugs, with acceptable toxicity on normal cells. There was no growth retrieval, after exposure to chalcone N9 alone, in a long-term assay to determine the cumulative population doubling (CPD) of human oral cancer cells. A PCR array evaluating 168 genes related to cancer and inflammation, demonstrated striking actions for N9, which altered the expression of 74 genes. Altogether, our results point out quinoxalinic chalcones, mainly N9, as potential strategies for oral cancer treatment.
Ciência e justiça precisam interagir para resolver diferentes situações. Essa interação, no entanto, precisa ocorrer de forma eficiente para não gerar ainda mais dúvidas. A avaliação científica no contexto jurídico deve ser compreendida e atingir seu objetivo. Para isso, muito se tem discutido sobre a natureza e finalidade da avaliação científica forense. Neste trabalho, objetivou-se avaliar o que é ciência forense e qual o alinhamento conceitual desta área do conhecimento frente aos cenários nacional e internacional. Realizamos uma revisão bibliográfica sobre o assunto para compreender os aspectos fundamentais desse tema. Estudos conceituais sobre ciência forense são praticamente inexistentes na literatura nacional. A ciência forense foi contextualizada em seus aspectos multi, inter e transdisciplinar. Os resultados quantitativos da revisão bibliográfica mostraram a necessidade urgente de desenvolver o tema no cenário nacional e seu alinhamento com a compreensão mundial. Aspectos históricos do desenvolvimento da ciência forense foram utilizados na discussão sobre a conceituação do termo. No entanto, internacionalmente há esforços constantes para definir o propósito e o escopo da ciência forense. A Declaração de Sydney foi lançada recentemente como um documento orientador. Concluímos que a discussão sobre o conceito de ciência forense deve ser constante para aprimorá-lo em nível nacional e alinhá-lo ao pensamento internacional. Espera-se que este trabalho estimule a discussão do tema, fortalecendo o desenvolvimento da ciência forense e reforçando sua importância para a administração da justiça.
A ciência forense auxilia a compreensão da dinâmica fática a partir do emprego de métodos, que possibilitam uma maior controlabilidade à produção da prova, buscando reduzir o subjetivismo inerente a outros meios de prova. Neste cenário, o que se busca responder é: como a compreensão e a mitigação dos erros forenses pode auxiliar na garantia dos direitos ao contraditório, à ampla defesa e à prova lícita? Como hipótese de trabalho tem-se que o conhecimento sobre os erros forenses é indispensável para a produção de uma prova pericial de maior qualidade, que leve ao julgador um conhecimento especializado de maior confiança, resultando em uma valoração probatória mais coerente e direcionada aos fatos, buscando reduzir o arbítrio e os erros judiciais. Para isso, utilizou-se o método hipotético-dedutivo, com o emprego da técnica de revisão bibliográfica e consulta à jurisprudência. Foi possível demonstrar, ao final, que a comunidade científica deve refletir, discutir e prezar por mais rigor na ciência forense, priorizando uma comunicação clara dos resultados, de modo a integrar ciência, justiça e sociedade de maneira harmônica.
A frase do matemático e PhD Alan Turing traduz, de maneira simples, o entendimento sobre as ciências e, em especial, a forense: mesmo que nós só possamos compreender (“enxergar”) a uma curta distância, já temos a noção de que existe muito trabalho a ser realizado e esta edição demonstra exatamente isso. Com uma série de artigos em formato de revisão bibliográfica, esse volume demonstra a importância de olhar para aquilo que já foi produzido e compreender, de maneira mais assertiva, qual a direção a ser tomada e como as diversas técnicas podem colaborar com os problemas encontrados na prática.
