Five abietane diterpenes were isolated from a hexane extract obtained from the roots of Aegiphila lhotzkyana and identified as uncinatone, cyrtophyllone B, teunvicenone E, sugiol and 12,16-epoxy-11,14-dihydroxy-6-methoxy-17(15 - 16)- abeo-abieta-5,8, 11,13,15-pentaene-3,7-dione. The last compound has only been described for this species. These compounds were tested for their antiproliferative effects on tumor cell lines using the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl- 2H-tetrazolium bromide (MTT) assay and on sea urchin egg development, as well as for their lytic properties on mouse erythrocytes. All diterpenes reduced the viability of tumor cells and inhibited sea urchin egg cleavage. None of the tested compounds presented hemolytic activity.
An NMR study of one new and several known abietane diterpenes isolated from the roots of Aegiphila lhotzkyana is described. In addition to 1D NMR, several 2D shift-correlated NMR pulse sequences ( 1 H-1 H-COSY, NOESY, HMQC and HMBC) were used to establish all the structures, and unambiguously perform the 1 H and 13 C chemical shift assignments of the new natural diterpene and three derivatives, the NMR data for which have not been reported previously. Revision of current data assignment for teuvincenone H is also suggested.
Leaves of Aegiphila lhotzkiana growing in the north-east of Brazil were submitted to hydrodistillation and analysed by GC-MS. The essential oil was characterized by high amounts of sesquiterpene hydrocarbons (87.0%), of which β β β β β-caryophyllene (27.5%), α α α α α-copaene (12.1%), caryophyllene oxide (10.5%) were predominant. Monoterpenes were represented only by oxygenated compounds, but in minor percentages (4.3%). This is the first report of any essential oil from Aegiphila species.
