Breast cancer metastasis to the bone, potentially facilitated by chemotactic and angiogenic cytokines, contributes to a dramatic osteolytic effect associated with this invasive behavior. Based on the intrinsic ability of calcium sensing receptor (CaSR) to control hormonal secretion and considering its expression in the breast, we hypothesized that CaSR plays a chemotactic and proangiogenic role in highly invasive MDA-MB-231 breast cancer cells by promoting secretion of multiple cytokines. In this study, we show that MDA-MB-231 cells stimulated with R-568 calcimimetic and extracellular calcium secreted multiple cytokines and growth factors that induced endothelial cell migration and in vitro angiogenesis. These effects were dependent on the activity of CaSR as demonstrated by the inhibitory effect of either anti-CaSR blocking monoclonal antibodies or calcilytic NPS-2143. Moreover, CaSR knockdown prevented the proangiogenic effect of CaSR agonists. Importantly, CaSR promoted secretion of pleiotropic molecules like GM-CSF, EGF, MDC/CCL22, FGF-4 and IGFBP2, all known to be chemotactic mediators with putative angiogenic factor properties. In contrast, constitutive secretion of IL-6 and β-NGF was attenuated by CaSR. In the case of normal mammary cells, secretion of IL-6 was stimulated by CaSR, whereas a constitutive secretion of RANTES, Angiogenin and Oncostatin M was attenuated by this receptor. Taken together, our results indicate that an altered secretion of chemotactic and proangiogenic cytokines in breast cancer cells is modulated by CaSR, which can be considered a potential target in the therapy of metastatic breast cancer.
Las sirtuinas son una familia de enzimas dependientes de nicotinamida adenina dinucleótido (NAD+) expresadas de manera ubicua. Sus principales funciones se ven reflejadas en: regular el silenciamiento génico, estabilidad cromosómica, y metabolismo al eliminar las modificaciones de acil-lisina de una variedad de proteínas. La primera sirtuina conocida fue SIR2 (regulador de información silencioso 2), aislada de Saccharomyces cerevisiae, a la cual la familia debe su nombre. De dicha proteína se presentan 7 homólogos en mamíferos, conocidos como SIRT1-7, las cuales se diferencian según su localización subcelular, actividad catalítica y funciones atribuidas. Las sirtuinas han ganado considerable atención por su impacto en la fisiología de los mamíferos, proporcionando blancos nuevos para el tratamiento de diferentes enfermedades del humano. En esta revisión, ilustramos la historia de las sirtuinas, desde levadura hasta el humano, así como su papel en la fisiología y patología humana.
El Síndrome de Korsakoff es una enfermedad ocasionada por el alcoholismo y causa más frecuentes de amnesia. Se ven afectados especialmente la memoria y el aprendizaje. Las alteraciones se manifiestan como una amnesia anterógrada (crear recuerdos nuevos). También puede llegar a presentarse amnesia retrógrada (dificultad de acceso a recuerdos del pasado). A nivel mundial y principalmente en la región de la Huasteca el consumo de alcohol es un problema de Salud Pública. Una población sensible a este fenómeno son los adolescentes o adultos jóvenes. Debido al alto consumo de bebidas embriagantes en el municipio de Huejutla de Reyes Hidalgo se realizará esta investigación para identificar patrones asociados al consumo de alcohol y detectar las posibles consecuencias en la memoria y el aprendizaje en estudiantes de nivel superior de la Universidad Autónoma del Estado de Hidalgo Escuela Superior Huejutla.
La retina, el órgano visual localizado en la parte posterior del glóbulo ocular, contiene diversas células, desde fotorreceptores especializados, hasta células neuronales, células ganglionares, cuyas necesidades metabólicas son suplidas por la microvasculatura retiniana. Dicha microvasculatura, compuesta por células endoteliales y pericitos, es altamente sensible a los niveles de glucosa en la circulación. Pacientes con Diabetes Mellitus tipo 2, con un pobre control glicémico, sufren daños microvasculares en diversos tejidos, incluyendo la retina. La condición de hiperglicemia descontrolada afecta negativamente a la microvasculatura de la retina, y a largo plazo conlleva a complicaciones irreversibles de vista como resultado de la retinopatía que se desarrolla. Un diagnóstico temprano de la retinopatía ayudaría a disminuir el daño ocasionado, y las comorbilidades que resultan. En esta revisión se otorga información básica sobre la retina, las condiciones patológicas que desencadena la Diabetes Tipo 2, y la utilidad del uso de proteómica de lágrimas como herramienta diagnostica de la retinopatía.
