-(Recalcitrant seeds: intolerant to low temperatures? Recalcitrant embryos of Inga vera Willd. subsp. affinis (DC.) T. D. Penn., a tropical species, are tolerant to subzero temperature). Recalcitrant seeds are intolerant to both desiccation and low temperatures, the most important mechanisms of seed conservation, consequently their storage is difficult. Inga vera subsp. affinis produces the so-called recalcitrant seeds, since they do not survive to water content lower than 35%. In this work it is demonstrated that these seeds are tolerant to temperatures as low as -2 °C. Embryos of I. vera of different stages of maturity were stored at temperatures varying from 8 to -18 °C for until 45 days. Non-dried embryos and both dried to -4 MPa and -6 MPa were stored. Results showed that, despite all embryos died at -18 °C, some cold tolerance was obtained by mature embryos when stored at -2 °C, as long as they are dried to -4 MPa.
As condições hídricas e térmicas do ambiente durante a formação das sementes podem condicionar diversas respostas fisiológicas, como a exigência térmica para a germinação. Neste trabalho analisou-se a germinação de embriões de Inga vera subsp. affinis de origens distintas em resposta a diferentes temperaturas, incluindo-se o cálculo dos graus-dia e da chuva acumulada durante o desenvolvimento e maturação. Os embriões foram obtidos de frutos maduros de matrizes localizadas em São Paulo e Minas Gerais e analisados quanto ao teor de água, conteúdo de massa seca e germinação. Os resultados demonstram variações nas respostas germinativas entre as origens dos materiais, sob temperatura sub-ótima. Dependendo da origem os embriões de I.vera subsp. affinis germinam a partir de 10 ºC ou 15 ºC e apresentam melhor desempenho em tempo a partir de 20 ºC ou 25 ºC. Tais variações podem estar associadas, entre outros fatores, às condições do ambiente de formação e ao grau de maturidade das sementes no momento da dispersão.
Inga vera ssp. affinis embryos at different maturity stages and with various water contents had their CO2 production and O2 consumption analyzed, and then compared to those of Phaseolus vulgaris L. seeds. The results showed that the lower the temperature, the lower the respiratory rates and the more intense the other oxidative processes of I. vera embryos. On the other hand, the same did not happen in P. vulgaris seeds. Oxidative reactions, including respiration, reduced the germinative capacity and compromised the preservation of the embryos and seeds. Oxidative reactions depended on the embryo maturity stage, its moisture level, and the surrounding temperature. Interestingly, the responses obtained for viability loss and changes in O2 consumption rates of I. vera embryos did not correspond to the expected for plant tissues, as verified in P. vulgaris seeds with similar water content.
RESUMO O elevado metabolismo de sementes sensíveis à dessecação (recalcitrantes) de Inga vera Willd. subsp. affinis (DC.) T.D.Pennington é um dos principais fatores que dificultam a manutenção da viabilidade fisiológica das sementes durante o armazenamento. Devido à falta de informações sobre o metabolismo respiratório dessas sementes em diferentes condições de estresse, este estudo teve como objetivo determinar as taxas de respiração de embriões com diferentes conteúdos de água e em diferentes temperaturas após a aplicação dos tratamentos com ABA e PEG. Os resultados mostraram altos níveis de respiração e presença de reações oxidativas. O ABA mostrou poucos efeitos, mas a respiração e os processos oxidativos foram diminuídos pela secagem leve e pelo tratamento com PEG. Portanto, a redução da temperatura de armazenamento é eficiente na redução do metabolismo, permitindo ampliar o armazenamento, mas a escolha do estádio de maturação é decisiva para melhor conservar os embriões de Inga vera.
-Drying, widely used for storing orthodox seeds for prolonged periods, cannot be applied to recalcitrant seeds, which are sensitive to desiccation. Thus, inclusion of species with recalcitrant seeds, like inga, in reforestation programs or even for commercial use has been hindered by the lack of technology that would allow storage of these seeds. The remaining option, cryopreservation, is a method of high cost that requires a high level of technology. Knowledge of the processes involved in sensitivity to desiccation continues to be a great challenge for the seed sector. The aim of this study was to analyze the effects of osmotic treatments on tolerance to desiccation and storage capacity of recalcitrant seeds of inga. Embryos were subjected to osmotic stresses with PEG solution and subjected to progressive drying processes. In another experiment, the effects of these solutions on embryo conservation during storage were analyzed. From the results, it may be concluded that incubation of embryos in a solution with -2.0 MPa increases their tolerance to desiccation.Index terms: osmotic stress, Inga vera, drying, recalcitrant seeds.Indução de tolerância à dessecação e à temperatura subzero em embriões de sementes recalcitrantes de ingá RESUMO -A secagem, amplamente utilizada para armazenar sementes ortodoxas por períodos prolongados, não pode ser aplicada às recalcitrantes, que são sensíveis à dessecação. Assim, a inclusão, em programas de reflorestamento ou mesmo de uso comercial, de espécies com sementes recalcitrantes, como o ingá, tem sido dificultada pela falta de tecnologia que permita armazenar suas sementes, restando a conservação de embriões criopreservados, que é tecnologia de alto custo e requer elevado nível tecnológico. O conhecimento dos processos envolvidos na sensibilidade à dessecação continua sendo um grande desafio para a área das sementes. Neste trabalho, objetivou-se analisar os efeitos de tratamentos osmóticos sobre a tolerância à dessecação e capacidade de armazenamento de sementes recalcitrantes de ingá. Embriões foram submetidos a estresses osmóticos com solução de PEG e submetidos a secagens progressivas. Em outro experimento, analisaram-se os efeitos dessas soluções sobre a conservação dos embriões durante seu armazenamento.
