Resumo -O objetivo deste trabalho foi avaliar a tolerância à dessecação de sementes de seis espécies frutíferas nativas de Eugenia (E. brasiliensis Lam., E. cerasiflora Miq., E. involucrata DC., E. pyriformis Camb., E. umbelliflora Berg. e E. uniflora L.) e fornecer subsídios para a conservação do poder germinativo dessas sementes durante o armazenamento. As sementes foram submetidas a secagens progressivas, em estufa (40ºC) e em câmaras com sílica-gel (25ºC), até 10% de água. Após cada secagem, as sementes foram avaliadas quanto ao teor de água e germinação. Observou-se relação significativa entre a diminuição do teor de água e a redução do poder germinativo, independentemente do processo de secagem. Apesar de diferenças nos limites de tolerância à dessecação das sementes das diferentes espécies, a redução do teor de água para valores inferiores a 45% sempre prejudicou a germinação dessas sementes, que perderam a capacidade germinativa quando o teor de água foi inferior a 15%.Termos para indexação: Myrtaceae, recalcitrante, secagem, frutífera, tropical. Desiccation tolerance of seeds of species of EugeniaAbstract -The objective of this work was to evaluate the desiccation tolerance of seeds of six fruit species of Eugenia (E. brasiliensis Lam., E. cerasiflora Miq., E. involucrata DC., E. pyriformis Camb., E. umbelliflora Berg. and E. uniflora L.) aiming to provide information for the conservation of the germinability of these seeds during storage. Seeds were dried continuously in oven (40ºC) or closed chambers with silica gel (25ºC) until 10% water content. After drying, samples of the seeds were analyzed for water content and germination. As water content decreased, seeds lost viability, independently of the drying method. Seeds from each species differed on sensitivity to desiccation, but all seeds were intolerant to levels lower than 45% water content and lost viability at levels lower than 15%.
-(Do recalcitrant seeds really exist?). In the 70's, seeds were divided into two categories: recalcitrant and orthodox. In the 80's, it was necessary to create an intermediate category; from the 90's onwards, a gradient between orthodox and recalcitrant categories has been considered by several authors. Currently, the terms orthodox and recalcitrant are appropriate just for technological purposes, not for scientific studies. It seems that the differences between recalcitrant and orthodox seeds lie only on the maturity stage in which they are detached from the mother plant, the recalcitrant ones in a very immature stage. This implies that little progress should be expected to expand the storability of these recalcitrant seeds with the application of any treatment after harvesting. Efforts shall be focused on amplifying the maturation period of these seeds by keeping them linked to the mother plant until the maturation process has been completed. Na década de 1970, as sementes foram divididas em recalcitrantes e ortodoxas. Na década de 1980, foi necessário criar a categoria das intermediárias e, a partir de 1990, um gradiente entre as categorias ortodoxas e recalcitrantes tem sido considerado por vários autores. Atualmente, os termos ortodoxa e recalcitrante são apropriados apenas para fins tecnológicos, não para estudos científicos. As diferenças entre sementes recalcitrantes e ortodoxas parecem residir apenas no estádio de maturidade no qual se desprendem da planta mãe, as recalcitrantes em uma fase muito imatura. Isso implica que pouco progresso deve ser esperado para expandir a capacidade de armazenamento com a aplicação de qualquer tipo de tratamento após a colheita dessas sementes recalcitrantes. Esforços devem ser voltados para ampliar o período de maturidade das sementes, mantendo-as ligadas à planta-mãe até que o processo de maturação tenha sido concluído. -(Desiccation tolerance and storage of seeds of Caesalpinia echinata Lam. (Brazil Wood), a species from the Atlantic Forest). The exploitation of Caesalpinia echinata reduced markedly the original geographic distribution of the species. The knowledge of seed physiology, mainly desiccation tolerance and storability, could be helpful for the conservation of the species and future recovery of degraded areas of the Atlantic Forest. Desiccation study was performed in four categories of seeds selected according to colour and size (I, II, III, and IV stages), being submitted to drying at 40 ºC and 50 ºC until 8% water content was reached. Freshly harvested and dried (9.7%) seeds were stored in cold chamber (7 ± 1 ºC) or in natural environment (22 ± 7 ºC) using packages with three different levels of permeability. Results showed that seeds of C. echinata are tolerant to desiccation (until 7.6% wet basis) and seed sensitivity to drying is dependent on the initial quality of the seed. Seeds stored in natural environment lost their viability in three months while under low temperatures germination was 80% after 18 months.RESUMO -(Tolerância à dessecação e armazenamento de sementes de Caesalpinia echinata Lam. (pau-brasil), espécie da Mata Atlântica). A exploração descontrolada do pau-brasil reduziu sua distribuição original a pequenos remanescentes na atualidade. O conhecimento da fisiologia da unidade de dispersão da espécie pode contribuir para a ampliação de seu cultivo, uso racional e conservação. A sensibilidade das sementes à dessecação foi avaliada em sementes recém-colhidas, separadas em quatro categorias (estádios I, II, III e IV), segundo avaliação visual de tamanho e cor. Sementes de cada estádio foram submetidas a secagem a 40 °C e a 50 °C, até que o teor de água atingisse 8% (base úmida). A capacidade de manutenção da viabilidade das sementes durante o armazenamento foi avaliada em diferentes embalagens (permeável, semi-permeável e impermeável), em ambiente natural (22 ± 7 °C) ou em câmara fria (7 ± 1 °C), com sementes submetidas ou não a secagem inicial, que reduziu o teor de água para 9,7%. Os resultados permitiram concluir que as sementes de C. echinata são tolerantes à dessecação, mas a sensibilidade à secagem pode ser influenciada pela qualidade inicial das sementes. Quando armazenadas sob condições normais de ambiente podem perder a viabilidade em menos de três meses. Sob temperatura baixa foi possível manter a viabilidade das sementes por até 18 meses, com germinação superior a 80%. A espécie em estudo comporta-se como ortodoxa, tolerando dessecação até atingir 7,6% de água, o que pode facilitar o armazenamento e ampliar o potencial de conservação, para fins de reposição das populações naturais.
-(Seed germination of "uvaia" (Eugenia pyriformis Cambess. -Myrtaceae) after cutting). Eugenia pyriformis Cambess. ("uvaia"), a tropical tree species from Brazil, could be used in forest regeneration programs, arboriculture and for fruit industrial use. However, seed production is not enough to produce seedlings in commercial scale. In this work, when seeds of E. pyriformis were longitudinally or transversally sectioned, high percentage of germination was maintained and even those that had been cut to leave 1/4 th of the original seed size have germinated and produced normal seedlings. In a second experiment, seeds were cut into eight parts and at least one of them germinated. Our results showed that it is possible to obtain more than one normal seedling from each seed. Furthermore, higher germination percentage and normal seedling growth have been obtained when seeds were cut in the longitudinal way.Key words -Myrtaceae, propagation, seed germination RESUMO -(Fracionamento e germinação de sementes de uvaia (Eugenia pyriformis Cambess. -Myrtaceae)). Eugenia pyriformis Cambess. (uvaia), espécie arbórea tropical do Brasil, pode ser utilizada em programas de reflorestamento e em áreas urbanas e seus frutos apresentam potencialidade de uso industrial. Contudo, apesar da elevada porcentagem de germinação, a produção de sementes dessa espécie é insuficiente para produção de mudas em escala comercial. No presente trabalho, sementes de E. pyriformis foram cortadas longitudinalmente ou transversalmente em duas partes e mantiveram a elevada porcentagem de germinação e de produção de plântulas normais, mesmo quando a fração resultante do corte tinha apenas 1/4 da semente. Em um segundo experimento, as sementes foram cortadas em até oito partes e pelo menos uma das frações de cada semente continuou apresentando germinação. Os resultados mostraram que, através do fracionamento, é possível obter mais de uma plântula normal a partir de cada semente. Caesalpinia echinata seeds stored in laboratory environmental conditions lose their viability in one month whilst under low temperatures germination is maintained for 18 months of storage. These seeds are tolerant to desiccation, keeping their viability up to 0.08 gH 2 O.gDW -1 . Since soluble carbohydrates are believed to be involved with desiccation tolerance and seed storability, the aim of this work is to analyze the content and composition of soluble carbohydrates in C. echinata seeds during storage in paper bags (PB) and glass flasks (GF) at laboratory room (RT) and cool (CT) temperatures. In freshly harvested seeds, total soluble carbohydrates comprised approximately 10% of the dry weight, decreasing to ca. 8% over 18 months of storage at RT. In seeds stored at CT, sugars varied differently decreasing initially and being restored at the end of the analysis period. The main neutral sugars in seeds from all treatments were sucrose, fructose and glucose. Raffinose and stachyose were present as traces. Free myo-inositol and other cyclitols were also detected. The main tendency observed was the variation in levels of both glucose and fructose in relation to sucrose, the highest levels of monosaccharides which were found in seeds stored at CT. The values of glucose and fructose were practically constant in seeds stored in paper bags for 18 months at CT, decreasing consistently in the other treatments, mainly at RT. Sucrose contents remained relatively stable. Changes in soluble sugars during storage suggest that the loss of germinability of seeds of C. echinata could be associated with low levels of glucose and fructose in relation to sucrose.Keywords: seed longevity, desiccation tolerance, pernambuco, soluble sugars, cyclitols. RESUMO Modificações nos carboidratos solúveis durante o armazenamento de sementes de Caesalpinia echinata LAM. (pau-brasil), uma leguminosa arbórea da Mata Atlântica em risco de extinçãoSementes de Caesalpinia echinata (pau-brasil) perdem a viabilidade em um mês quando armazenadas no ambiente de laboratório, enquanto a capacidade germinativa é mantida quando armazenadas sob temperturas baixas. O presente trabalho teve como objetivos analisar o conteúdo e a composição dos carboidratos de sementes de C. echinata armazenadas em câmara fria (CT) e em temperatura ambiente do laboratório (RT), em duas embalagens distintas (permeável e impermeável), visando a avaliar o envolvimento desses compostos com a capacidade germinativa das sementes. -(Recalcitrant seeds: intolerant to low temperatures? Recalcitrant embryos of Inga vera Willd. subsp. affinis (DC.) T. D. Penn., a tropical species, are tolerant to subzero temperature). Recalcitrant seeds are intolerant to both desiccation and low temperatures, the most important mechanisms of seed conservation, consequently their storage is difficult. Inga vera subsp. affinis produces the so-called recalcitrant seeds, since they do not survive to water content lower than 35%. In this work it is demonstrated that these seeds are tolerant to temperatures as low as -2 °C. Embryos of I. vera of different stages of maturity were stored at temperatures varying from 8 to -18 °C for until 45 days. Non-dried embryos and both dried to -4 MPa and -6 MPa were stored. Results showed that, despite all embryos died at -18 °C, some cold tolerance was obtained by mature embryos when stored at -2 °C, as long as they are dried to -4 MPa.
The uncommon ability of monoembryonic seeds of Eugenia to develop seedlings after cutting or even after the removal of more than half of their storage tissue has recently been reported. Some Eugenia species are included among those at risk of disappearing, so the regeneration of plantlets from fragments of seeds could be an important mechanism both for the production of more than one normal seedling per seed and to understand the survival strategies of these species. Because the period in which this ability is present has not been studied, we analyzed the regenerative capacity of seeds of five Brazilian native species of Eugenia, considering two levels of maturity and three germination phases. Mature seeds have a greater potential to regenerate than immature ones; germinating seeds showed that the longer the root protrusion, the smaller the potential to regenerate new seedlings. However, our results clearly demonstrate that the ability to regenerate new roots or complete seedlings is present during a long period from the beginning of development until the late phases of germination. Cutting is potentially important to obtain plants on a larger scale. Because of intense predation of these seeds, this ability could be linked to an essential strategy for survival even after the removal of more than half of the storage tissues. As an alternative strategy to orthodox seeds, whose preservation is based on tolerance to desiccation, the recalcitrant seeds of Eugenia probably adopted the great reserve quantity and the regenerative capacity, allowing them to undergo successive germinations.
Há grande demanda por pesquisas com espécies arbóreas nativas do Brasil, principalmente quanto à qualidade fisiológica de suas sementes. Visando avaliar o grau de tolerância à dessecação e a capacidade de armazenamento das sementes de grumixameira (Eugenia brasiliensis), frutos maduros foram coletados em Mogi-Guaçu, SP. Após a colheita, as sementes foram extraídas, lavadas e secas a 36ºC, reduzindo seu teor de água inicial de 48,9% (base úmida) para até 23,6% (última secagem), totalizando cinco níveis de secagem. Amostras de sementes de cada nível de secagem foram armazenadas em sacos plásticos, a 7ºC, até 270 dias. Os resultados mostraram que a redução do teor de água para valores inferiores a 43,1% prejudicou tanto a germinabilidade quanto a capacidade de conservação em armazenamento das sementes. Sementes com 48,9% de água apresentaram 60% de germinação após 180 dias de armazenamento e 19% após 270 dias, quando mantidas em sacos plásticos e em câmara fria. Concluiu-se que sementes de E. brasiliensis podem ser armazenadas por 180 dias a 7ºC e são sensíveis à secagem a 36ºC.
