High-dose benzoate treatment aimed at reducing plasma glycine levels to normal reduces seizures and increases wakefulness in patients with nonketotic hyperglycinaemia (NKH). Since benzoate metabolism is dependent on the available glycine pool, and since the glycine pool is variably affected by the deficiency in the glycine cleavage enzyme system, we examined the importance of interpatient variability in benzoate requirement. To correct for the dietary glycine contribution, the glycine index was introduced as the molar requirement of benzoate dose necessary to normalize plasma glycine levels and subtracting from that the dietary glycine intake, both corrected for weight. The glycine index varied between 3.62 and 4.87 mmol/kg per day in five patients with a poor neurodevelopmental outcome and between 0.92 and 1.90 mmol/kg per day in four patients with a better neurodevelopmental outcome, and was 2.54 mmol/kg per day in a single patient with an intermediate outcome. The glycine index was stable over time within each patient. Exceeding the balance by either increasing food glycine intake or decreasing the benzoate dose resulted in increased glycine levels. Exceeding the glycine tolerance by increasing benzoate resulted in elevated and toxic levels of benzoate. The glycine index is a stable, individually specific parameter in patients with NKH. It has clinical consequences for the dose of benzoate required and the role of dietary management. Through its correlation with neurodevelopmental outcome, the glycine index points to potential genetic factors that could contribute to the psychomotor retardation in NKH.
No município de Colniza, Mato Grosso, a principal limitação para expansão pecuária é a ocorrência de "morte súbita" em bovinos, com registros de mortalidade próxima a 50% dos animais. Em visitas realizadas em áreas de ocorrência do problema, nos anos de 2004, 2011 e 2012, constatou-se que havia coincidência entre a ocorrência de "mortes súbitas" no rebanho e a presença de Amorimia pubiflora nas pastagens. As mortes ocorrem durante todo ano, porém acentuam-se no início do período das chuvas, quando há maior quantidade de brotação nas áreas de pastoreio. A intoxicação foi reproduzida em ovinos e bovinos através da administração de folhas jovens coletadas em dois períodos do ano, e, em ovinos, através de folhas maduras e dos frutos. Nos ovinos que morreram, as primeiras manifestações clínicas foram observadas entre 34min e 17h34min após a administração da planta e a evolução clínica foi de 3min a 15h20min, com uma fase final superaguda de 3 a 21min. As principais alterações clínicas encontradas foram taquicardia, evidenciação da jugular, tremores musculares, apatia e relutância à movimentação. Todos os sinais acentuavam-se após a movimentação. A fase final superaguda foi caracterizada por relutância para caminhar, cifose, tremores e contrações musculares generalizadas, principalmente de membros, cabeça e pescoço. Notou-se também taquipneia com respiração abdominal, decúbito esternal e rapidamente lateral ou quedas em decúbito lateral, opistótono, nistagmo e cianose de mucosa oral, seguidos de morte. As folhas jovens, independentemente do período da coleta, foram mais tóxicas; causaram a morte de ovinos a partir de 2g/kg e de um bovino que ingeriu 3g/kg. Já as folhas maduras revelaram-se tóxicas e causaram morte na dose de 20g/kg e os frutos ocasionaram a morte de um ovino que ingeriu 5g/kg. Concluímos que monofluoracetato de sódio (MFA), encontrado na concentração de 0,015% nas folhas em brotação de A. pubiflora, é o princípio tóxico responsável pela "morte súbita" causada por Amorimia pubiflora. Esse estudo mostra a importância de A. pubiflora para a região Centro-Oeste do Brasil, principalmente para a pecuária bovina do município de Colniza, MT. Essa planta é tóxica, também, para ovinos e o quadro clínico é similar ao descrito para bovinos.

Computerized microtomography is the gold standard examination for the evaluation of the three-dimensional bone structure. This experiment was developed to evaluate the structure and bone quality of Caiman yacare with metabolic bone disease using high resolution computerized microtomography (μCT). The animals were distributed into four groups: G1 -hyperphosphatemic diet with sun exposure deprivation (n=4), G2 -hyperphosphatemic diet with sun exposure (n=4), G3 -balanced diet with sun exposure deprivation (n=4), and G4 -balanced diet with exposure to sunlight (n=4). The parameters for the trabecular bone (Trabecular Number, Trabecular Thickness, Trabecular Separation, Bone Pattern Factor, Fractal Dimension, Euler Number, Structural Model Index, Degree of Anisotropy, Eigenvalues 1, 2 and 3, and Centroides X, Y and Z), and cortical bone (Number of Closed Pores, Volume of Closed Pores, Surface of Closed Pores, Closed Porosity, Volume of Open Pores, Open Porosity and Total Porosity). The overall results showed that the structure and bone quality of group G3 and G4 were better than those of groups G1 and G2, and that the diet factor influenced more than the sun exposure factor. The computerized microtomography allowed to evaluate the quality of the cortical and trabecular bones of the Pantanal alligator tibia with osteometabolic disease. The diet and sun exposure factors influenced individually the results of the μCT parameters between the groups, demonstrating the functional and structural complexity. Thus, these parameters can contribute to the interpretation of the mechanical behavior of bones and correlate them with the risk of lesions and fractures associated with osteometabolic diseases.
RESUMO: Casos de anemia hemolítica em bovinos de corte em sistema de criação extensiva em Mato Grosso e Rondônia são acompanhados desde 2008. Os animais acometidos apresentam fraqueza, mucosas pálidas ou ictéricas, urina enegrecida, depressão e anorexia. Esse quadro muitas vezes evolui para morte, principalmente quando os animais são movimentados. Durante esse período aproximadamente 429 bovinos morreram em diferentes surtos eem alguns casos a mortalidade chegou a 50%. Foi realizada investigação para as principais causas infecciosas como leptospirose, babesiose, tripanossomíase, anaplasmose e hemoglobinúria bacilar, bem como para as possíveis causas toxicas relatadas como etiologia de anemia hemolítica em bovinos por meio de exames hematológicos, bioquímica sanguínea, moleculares e histopatológicos. Não se observou nas propriedades acometidas, nenhuma das plantas já descritas no Brasil como causadora de anemia hemolítica em animais de interesse pecuário no momento das investigações dos surtos. Apesar da etiologia desses casos ainda não ser conhecida, as evidências epidemiológicas, clínicas e patológicas da doença sugerem que a mesma seja causada por uma planta tóxica de ação hemolítica, presente nas propriedades acometidas, mas que ainda não foi confirmada experimentalmente.
Current research is focusing on integrated longitudinal assessment of animal welfare at farm-level. Housing and management systems may influence pain, discomfort, fear, hunger and abnormal behaviour of farm animals. Poor health records and increased levels of haptoglobin have been shown to correlate with an unfavourable environment but, as yet, few data are available regarding variation between individual animals. Hence, a project was carried out using 78 pig farms (farrow-to-finish), 19-20 in each season, with data on housing and management being collected during visits. At slaughter, pathological findings and blood samples were taken from 60 pigs from each farm. Blood samples were analysed for Lawsonia intracellularis (PIA), Mycoplasma hyopneumoniae, salmonella, and haptoglobin values (HAP) (10 samples). Data were analysed with descriptive statistics and analysis of variance. Housing and management characteristics were considered separately and integrated according to Berns (1996). Pigs from higher-ranking farms (ie those scoring higher for housing [space score] and management [sanitary barriers, cleaning, disinfection, climatic control, breeding protocol]) showed lower HAP levels (P < 0.04), with lower within-farm variability (P < 0.06). HAP levels were higher in pigs infected with PIA (P < 0.04) or having lung lesions (P < 0.02). A negative correlation was found between fasting before transport and lung lesions, HAP levels being lower when pigs with lung lesions were fasted. Haptoglobin sampling in the slaughterline is, therefore, relevant for integrative welfare assessment of slaughter pigs at individual level and for longitudinal monitoring at farm level.
