This study focuses on the standardization of techniques across laboratories to enable multiple datasets to be compared and combined in order to obtain reliable and robust wide-scale patterns of diversity. A set of protocols using a core collection of simple sequence repeat (SSR) markers, reference lines and standard alleles, plus a common system of allele nomenclature, was adopted in the study of maize genetic diversity in a network of laboratories in Asia. Pair-wise allele comparisons of the reference lines, done to assess the general agreement between datasets from four laboratories, showed error rates (raw) ranging from 5.8% to 9.7%, which were reduced to less than 8% after adjustments of correctable errors, and further reduced to less than 6% after the exclusion of all markers with greater than 10% individual error rates. Overall, 45% of the total mismatches were due to frameshift errors, 39% to wrong allele size, 15% to failed amplification and 1% to "extra" alleles. Higher genetic similarity values of the reference lines were achieved using fewer markers with data of higher quality rather than with more markers of questionable quality. Cluster analysis of the merged datasets showed the lines from southern China to be highly diverse, falling into six of the seven clusters observed and all well represented by tester lines. The lines from Indonesia fell into five of six groups, with two main groups represented by tester lines. The CIMMYT lines developed for the Asian region showed a relatively narrow genetic base, falling in two out of seven and in three out of six clusters in China and Indonesia, respectively. In contrast to the case in southern China where 95% of the lines clustered separately from the CIMMYT lines, lines in the Indonesian breeding program show a closer relationship with the CIMMYT lines, reflecting a long history of germplasm exchange.
<p>Information on genetic diversity, homozygosity, drought stress and low N tolerance of maize inbred line are useful for parental selection in developing maize varieties tolerant to drought and low N. The objectives of this study were (a) selection for homozygoes lines and analysis of genetic diversity among 51 maize inbred lines applying simple sequence repeats (SSRs) using thirty six markers (b) selection for inbred lines (homozygosity over 80%) for drought and low nitrogen (N) tolerance. Experiment was conducted using split-split plots design with three replications. Water treatments were as the main plots (well-watered and drought stress conditions), subplots were nitrogen fertilization at rate of 75 and 150 kg N/ha and the sub-sub plots were 51 inbred lines. The results showed that there were broad ranges of genetic variability among inbred lines with genetic similarity coefficient values ranging from 0.22 to 0.87 and polymorphism information content average was 0.57. Thirty inbreds having homozygosity over 80% were spread into six heterotic groups. Drought tolerance inbreds were in heterotic groups C and F, namely DTPYC9-F46-3-9-1-1-B and 1044-30, the drought medium tolerance in heterotic group A and B, namely CML 161/NEI 9008 and MR 14. Inbred for low-N fertility tolerance was in the heterotic group D namely G20133077, while medium tolerance to low-N fertility inbreds were in heterotic group A, B, C, D, and F, and they were CML 161/NEI 9008, CY 11, CY 15, CY 6, CLRCY039, Nei9008, DTPYC9-F46-1-2-1-2-B, G2013627, G2013649, 1044-30. Inbreds tolerance to both medium drought and to low-N fertility were in heterotic group C and F they were DTPYC9-F46-1-2-1-2-B and 1044-30. Inbred lines of maize tolerant to drought and to low-N fertility can be used as parent to develop hybrid or synthetic varieties, posessing stress tolerances, by cross recombination between heterotic groups.</p>
Abstract. The demands for alternative sources of energy are currently growing because people now are more aware of the many negative impacts fossil fuel gives to the environment. Plant based renewable energy provides potential sources of energy with advantages of cleaner fuel effect and capability of integration with food crop production. Sorghum have been considered to be a highly potential source of food, feed and fuel, especially sweet stalk sorghum that posses both functions as source of food from its grain and fuel made from its stalk juice. Sorghum varieties are well known to have excellent adaptability in marginal areas, especially drought prone areas where other food crops are unable to thrive. The current paper aimed to share ongoing research on many functional uses of sweet stalk sorghum varieties released at Indonesian Cereals Research Institute (ICERI). Among many varieties that had been released were two sweet stalk sorghum varieties SUPER-1 and SUPER-2 released in 2013 that stands out in biomass yield and bioethanol production. Based on various researches conducted at different location and planting season, yield potential of biomass ranged at 30 -40 t/ha with higher yield occurred during dry season. Stalk juice sugar content in brix were found to be higher in dry season ranged at 13.6 % to 18.4 %, and the amount of juice stalk was obtained at about 30-50 % from total biomass yield. Furthermore, bioethanol production from stalk juice after fermentation was at the range of 8 to 10 % from total stalk juice volume. Modification of processing equipment of bioethanol have also been carried out and was able to increased the concentration of ethanol being distilled from 85% -92% to 90% -95%. Another result obtained was able to decreased fermentation time from 14-21 days to 6-10 days. Furthermore, the yield of ethanol from juice was also from an average of 4.95% to 6.75%.
Linkage of 23 Microsatellite Marker on Chromosome 6 and 7 to Downy Mildew Resistance on Maize. Roberdi, Hajrial Aswidinnoor, Asep Setiawan, Sutrisno, Marcia B. Pabendon, and M. Azrai. Downy mildew caused by Peronosclerospora is one of most important maize disease in several countries, including Indonesia. Parental and progenies selection based on conventional breeding is time consuming and laborious. Development of molecular biology produces many DNA markers used for selection, one of them is microsatellite. The aim of this research to identify microsatellite markers associated with downy mildew resistance on maize progeny MR-4 X AMATLCOHS-9-1-1-1-1-1-2-B, on chromosome 6 and 7. This research was consisted of two activities, phenotypic and genotypic analysis. Phenotypic analysis used 175 progenies BC 1 F 2 and both of parents. This analysis included planting of spreading row, inoculums preparation, inoculation of spreader rows, test material planting, inoculation of test material and observation. Genotypic analysis used 175 progenies BC 1 F 1 and both of parents. This analysis included DNA genome isolation, PCR analysis, electrophoresis, gel staining and data scoring. Percentage of downy mildew infections on MR-4 was 76%, while these on AMATLCOHS-9-1-1-1-1-1-2-B was 16%, and on 175 progenies had range from 10.1-100%. Out of 23 SSR, 12 markers could be mapped in chromosome 6 and 11 markers in chromosome 7. QTL analyses showed that chromosome 7 contain one QTL in position between phi082 and phi116I marker as far as 18.6 cM with 2.6 LOD value.
The compulsory requirement for releasing new high yielding maize varieties is resistance to downy mildew. The study aimed to determine the level of homozygosity, genetic diversity, and genetic distance of 30 S3 genotypes of maize. Number of primers to be used were 30 polymorphic SSR loci which are distributed over the entire maize genomes. The S3 genotypes used were resistant to downy mildew with homozygosity level of >80%, genetic distance between the test and tester strains >0.7, and anthesis silking interval (ASI) between inbred lines and tester lines was maximum 3 days. The results showed that 30 SSR primers used were spread evenly across the maize genomes which were manifested in the representation of SSR loci on each chromosome of a total of 10 chromosomes. The levels of polymorphism ranged from 0.13 to 0.78, an average of 0.51, and the number of alleles ranged from 2 to 8 alleles per SSR locus, an average of 4 alleles per SSR locus. The size of nucleotides in each locus also varied from 70 to 553 bp. Cophenetic correlation value (r) at 0.67 indicated that the Unweighted Pair-Group Method Using Arithmetic Averages (UPGMA) was less reliable for differentiating genotypes in five groups. Of the total of 30 genotypes analyzed, 17 genotypes had homozygosity level of >80% so it can be included in the hybrid assembly program.
