Piracicaba 2016 3 RESUMO Caracterização de variações genéticas naturais em tomateiro controlando a competência celular para assumir diferentes vias de desenvolvimento O estudo de variações genéticas naturais afetando a capacidade de organogênese in vitro em tomateiro (Solanum lycopersicum) é promissor devido a existência de uma série de espécies selvagens relacionadas ao tomateiro, que apresentam alta capacidade organogênica in vitro. A caracterização de tais variações é relevante não apenas com o objetivo de manipulação do desenvolvimento vegetal, mas também com o intuito de entender o significado ecológico e evolutivo de tal característica. O objetivo desse trabalho foi caracterizar três loci de tomateiro, cujos alelos vindos de seu parente selvagem S. pennellii aumentam a capacidade de regeneração de gemas caulinares e radiculares in vitro, e analisar o envolvimento de tais loci na fase de aquisição de competência para regeneração. Nós apresentamos no primeiro capítulo a caracterização genética e fisiológica dos loci Rg3C, Rg7H e Rg8F. Os alelos de S. pennellii foram introgredidos na cultivar modelo Micro-Tom (MT), criando as linhagens quase isogênicas (Near Isogenic Lines-NILs) MT-Rg3C, MT-Rg7H e MT-Rg8F. No segundo capítulo nós analisamos comparativamente as NILs MT-Rg3C e MT-Rg1. Uma vez que Rg1 foi proposto como gene chave na aquisição de competência, e assim como Rg3C está localizado no cromossomo 3, acredita-se que Rg3C seja provavelmente ortólogo ao gene Rg1 de S. peruvianum. Após a introgressão dos loci na cultivar MT, as NILs, assim como esperado, apresentaram alta taxa de regeneração tanto de gemas caulinares, quanto de radiculares in vitro, confirmando que os loci foram devidamente introgredidos. A análise do tempo de aquisição de competência e indução, juntamente com a caracterização molecular das NILs, indicam que os genes localizados nos loci Rg3C, Rg7H e Rg8F afetam a regeneração in vitro através de rotas distintas. Enquanto Rg3C diminui o tempo necessário tanto para a aquisição de competência quanto para indução de gemas caulinares, os outros dois loci parecem influenciar apenas a aquisição de competência, no caso de Rg8F, ou a indução de gemas caulinares, no caso de Rg7H. Além disso, apesar de MT-Rg3C apresentar alta ramificação, MT-Rg7H e MT-Rg8F não diferiram de MT nesse aspecto, o que evidencia que a formação de gemas caulinares in vitro não está necessariamente relacionada ao aumento da ramificação. As análises comparativas entre MT-Rg3C e MT-Rg1 indicam fortemente que Rg1 e Rg3C sejam dois alelos de um mesmo gene controlando a alta capacidade de regeneração. Através do cruzamento dos dados de mapeamento disponíveis para esses dois alelos foi possível diminuir o número de genes candidatos à Rg1/Rg3C para apenas 27 genes, que são apresentados nesse trabalho. The leaves of the wild tomato Solanum galapagense harbor type-IV glandular trichomes (GT) that produce high levels of acylsugars (AS), conferring insect resistance. Conversely, domesticated tomatoes (S. lycopersicum) lack type-IV trichomes on the leaves of mature plants, preventing high AS production, thus rendering the plants more vulnerable to insect predation. We hypothesized that cultivated tomatoes engineered to harbor type-IV trichomes on the leaves of adult plants could be insect-resistant. We introgressed the genetic determinants controlling type-IV trichome development from S. galapagense into cv. Micro-Tom (MT) and created a line named “Galapagos-enhanced trichomes” (MT-Get). Mapping-by-sequencing revealed that five chromosomal regions of S. galapagense were present in MT-Get. Further genetic mapping showed that S. galapagense alleles in chromosomes 1, 2, and 3 were sufficient for the presence of type-IV trichomes on adult organs but at lower densities. Metabolic and gene expression analyses demonstrated that type-IV trichome density was not accompanied by the AS production and exudation in MT-Get. Although the plants produce a significant amount of acylsugars, those are still not enough to make them resistant to whiteflies. We demonstrate that type-IV glandular trichome development is insufficient for high AS accumulation. The results from our study provided additional insights into the steps necessary for breeding an insect-resistant tomato.
Conservation of plant genetic resources is important to prevent genetic erosion. Seed banks are the most common method of ex situ conservation; however, coffee seeds can not be stored by conventional methods. Cryopreservation is a viable alternative for long-term conservation of species that produce intermediate or recalcitrant seeds, as coffee. The aim of this work was to cryopreserve Coffea arabica L. cv Catuaí Vermelho IAC 144 zygotic embryos, and analyse the effects of dehydration prior cryopreservation and osmotic rehydration after thawing, in embryos germination and seedlings formation after cryopreservation. Prior to cryopreservation, different dehydration times (0, 15, 30, 60 and 120 min) were tested. Dehydrated embryos were cryopreserved in liquid nitrogen for 1 hour, and after thawing were rehydrated by osmotic solutions. Dehydrated and non-cryopreserved embryos were also analysed. The test with 2,3,5 triphenyl tetrazolium chloride (TTC) was used to evaluate the embryos viability. Non-dehydrated embryos did not survive after freezing. Embryos that were dehydrated until 20% of the moisture content did not germinate when osmotic rehydration was not performed. In contrast, cryopreserved embryos with the same moisture content presented 98% germination when they were rehydrated slowly in osmotic solution. According to tetrazolium tests, embryos presented maximum viability (75%) after dehydration for 60 minutes (23% moisture content). Therefore, coffee zygotic embryos (Coffea arabica L. cv. Catuaí Vermelho) can be successfully cryopreserved using physical dehydration in silica gel for 60 minutes (23% moisture content), followed by osmotic rehydration after thawing. RESUMOA conservação dos recursos genéticos vegetais é uma medida importante para prevenção do processo de erosão genética. Bancos de sementes são as formas mais comuns de conservação ex situ, entretanto, sementes de cafeeiro não podem ser armazenadas por métodos convencionais. A criopreservação é uma alternativa viável para conservação em longo prazo de espécies, como o cafeeiro, que produzem sementes intermediárias ou recalcitrantes. O objetivo deste trabalho foi criopreservar embriões zigóticos de Coffea arabica L. cv Catuaí Vermelho avaliando o efeito da desidratação antes da criopreservação, e da reidratação osmótica após o descongelamento. Antes da criopreservação foram testados e determinados os teores de umidade em diferentes tempos de desidratação (0, 15, 30, 60 e 120 min). Após a desidratação os embriões foram criopreservados por uma hora em nitrogênio líquido, e após o descongelamento foram reidratados utilizando solução osmótica. NAC proteins are one of the largest families of plant-specific transcription factors (TFs). They regulate diverse complex biological processes, including secondary xylem differentiation and wood formation. Recent genomic and transcriptomic studies of Tectona grandis L.f. (teak), one of the most valuable hardwood trees in the world, have allowed identification and analysis of developmental genes. In the present work, T. grandis NAC genes were identified and analyzed regarding to their evolution and expression profile during wood formation. We analyzed the recently published T. grandis genome, and identified 130 NAC proteins that are coded by 107 gene loci. These proteins were classified into 23 clades of the NAC family, together with Populus, Eucalyptus, and Arabidopsis. Data on transcript expression revealed specific temporal and spatial expression patterns for the majority of teak NAC genes. RT-PCR indicated expression of VND genes (Tg11g04450-VND2 and Tg15g08390-VND4) related to secondary cell wall formation in xylem vessels of 16-year-old juvenile trees. Our findings open a way to further understanding of NAC transcription factor genes in T. grandis wood biosynthesis, while they are potentially useful for future studies aiming to improve biomass and wood quality using biotechnological approaches.
