Taro (Colocasia esculenta (L.) Schott) is widely distributed in tropical and sub-tropical areas. However, its origin, diversification and dispersal remain unclear. While taro genetic diversity has been documented at the country and regional levels in Asia and the Pacific, few reports are available from Americas and Africa where it has been introduced through human migrations. We used eleven microsatellite markers to investigate the diversity and diversification of taro accessions from nineteen countries in Asia, the Pacific, Africa and America. The highest genetic diversity and number of private alleles were observed in Asian accessions, mainly from India. While taro has been diversified in Asia and the Pacific mostly via sexual reproduction, clonal reproduction with mutation appeared predominant in African and American countries investigated. Bayesian clustering revealed a first genetic group of diploids from the Asia-Pacific region and to a second diploid-triploid group mainly from India. Admixed cultivars between the two genetic pools were also found. In West Africa, most cultivars were found to have originated from India. Only one multi-locus lineage was assigned to the Asian pool, while cultivars in Madagascar originated from India and Indonesia. The South African cultivars shared lineages with Japan. The Caribbean Islands cultivars were found to have originated from the Pacific, while in Costa Rica they were from India or admixed between Indian and Asian groups. Taro dispersal in the different areas of Africa and America is thus discussed in the light of available records of voyages and settlements.
Six fatty acids (1-6), two xanthines (7, 8), two flavonol glycosides (9, 10), one monoterpene glucoside (11), one lignan glycoside (12), and four flavanes (13-16) were clarified by a bioassay-guided separation as chemical constituents of Scurrula atropurpurea (Loranthaceae), a parasitic plant of the tea plant Thea sinensis (Theaceae). Among these constituents, it was found that the alkynic fatty acid octadeca-8,10,12-triynoic acid (6) exhibits a more potent inhibitory effect on cancer cell invasion in vitro than flavanes [(+)-catechin (13), (-)-epicatechin (14), (-)-epicatechin-3-O-gallate (15) and (-)-epigallocatechin-3-O-gallate (16)].
Isozyme variation was studied in 94 isolates of Phytophthora colocasiae originating from Indonesia, Papua New Guinea, the Philippines, Thailand and Vietnam. Eight polymorphic enzyme systems (HK, PGM, PGI, Gluco, MDH, ICD, 6PDH, ME) revealed 52 isozyme patterns (zymotypes), each uniquely characterized by the presence or absence of 60 electromorphs. A core sample of 20 isolates was subsequently analysed with RAPD markers. Seven primers were used successfully and all profiles were reproducible. Clear bands were revealed and, in some cases, allowed differentiation between isolates exhibiting identical zymotypes. Results indicate that throughout this vast geographic region, taro leaf blight is caused by numerous and distinct strains that are genetically variable. Variation occurs within and between countries. The geographical distribution of zymotypes shows that none is common to two different countries. Although the differences in pathogenicity are not yet established, different P. colocasiae genotypes are likely to recombine and evolve rapidly as this species is heterothallic. From these results, a long-term breeding strategy is recommended for taro ( Colocasia esculenta ) based on recurrent selection using a wide genetic base composed of carefully selected parents from diverse geographic origins to maximize multigenic resistance in progenies.
Clonally propagated crop species are less adaptable to environmental changes than those propagating sexually. DNA studies have shown that in all countries where taro (Colocasia esculenta (L.) Schott) has been introduced clonally its genetic base is narrow. As genetic variation is the most important source of adaptive potential, it appears interesting to attempt to increase genetic and phenotypic diversity to strengthen smallholders' capacity to adapt to climatic changes. A global experiment, involving 14 countries from America, Africa, Asia and the Pacific was conducted to test this approach. Every country received a set of 50 indexed genotypes in vitro assembling significant genetic diversity. After onstation agronomic evaluation trials, the best genotypes were distributed to farmers for participatory on-farm evaluation. Results indicated that hybrids tolerant to taro leaf blight (TLB, Phytophthora colocasiae Raciborski), developed by Hawaii, Papua New Guinea and Samoa breeding programmes outperformed local
To examine diversity of endophytic microbes in plants, we have devised a method for isolation. We could isolate at least one kind of microbes from all the plants examined. One hundred sixty two fungi and 166 bacteria were obtained from 57 plants in Hokkaido Japan and 86 fungi and 56 bacteria were obtained from 20 plants in Java Indonesia. Among those isolates shown to have interesting bioactivities, 9 filamentous fungi were selected and taxonomic study was carried out on the basis of 18S rDNA base sequence analyses. These experiments suggest that endophytes from various plants have great diversity and useful characteristics for biotechnological applications.
3-Carbamoyl-alpha-picolinic acid, a versatile building block for the synthesis of agrochemicals and pharmaceuticals, was prepared by imidase-catalyzed regiospecific hydrolysis of 2,3-pyridinedicarboximide with intact Arthrobacter ureafaciens O-86 cells. Reactions were carried out in a water-organic solvent, two-phase system containing cyclohexanone at low pH to avoid spontaneous random hydrolysis. Under the optimized conditions, with the periodic addition of 2,3-pyridinedicarboximide (in total, 40 mM), the 3-carbamoyl-alpha-picolinic acid yield reached 36.6 mM in the water phase, with a molar conversion yield of 91.5% and a regioisomeric purity of 94.5%, in 2 h at pH 5.5.
