We examined the effect of dietary eicosapentaenoic acid (20:5n-3, EPA) on growth, survival, pigmentation and fatty acid composition of Senegal sole larvae using a dose-response design. From 3 to 40 days post hatch (dph), larvae were fed live food that had been enriched using one of four experimental emulsions containing graduated concentrations of EPA and constant docosahexaenoic acid (22:6n-3, DHA) and arachidonic acid (20:4n-6, ARA). Proportions of EPA in the enriched Artemia nauplii were described as "nil" (EPA-N, 0.5% total fatty acids, TFA), "low" (EPA-L, 10.7% TFA), "medium" (EPA-M, 20.3% TFA) or "high" (EPA-H, 29.5% TFA). Senegal sole larvae were fed live prey enriched with different amounts of arachidonic acid (ARA,) and eicosapentaenoic acid (EPA, 20:5n-3) to reevaluate the effect of these two fatty acids on flatfish pigmentation. Echium oil, a plant derived oil rich in gamma-linolenic acid (GLA, 18:3n-6) and stearidonic acid (SDA, 18:4n-3) was also used as a component of one of the enrichment emulsions. Although ARA content did not have any effect on growth there was a clear influence on pigmentation that correlated clearly with prostaglandin production. Inclusion of Echium oil, on the contrary, exerted a positive effect on pigmentation rate even though dietary ARA levels were as high as in the other emulsions. The relationships between dietary ARA levels and dietary ARA/EPA ratio, prostaglandin production and pigmentation are discussed.3
This study provides a comprehensive description of the main morphological and histological events that take place during larval and post-larval development of Senegal sole Solea senegalensis in order to establish a reference for its normal developmental organogenesis. Five stages have been described. Before gill development at the onset of metamorphosis (eye migration process, stage 4c), the skin was the main site of gas and ion exchange, whereas during stages 3 and 4, the skin begins differentiating into the definitive juvenile structure. The timing of development of the endocrine system depends on each organ, the endocrine pancreas and thyroid gland being the first to differentiate (stages 2 and 3, respectively), followed by the interrenal tissue and stannius corpuscles that develop at metamorphosis (stages 4 and 4c, respectively). The differentiation and maturation of the lymphohaematopoietic organs was coupled with the increase in complexity of the cardiovascular system and the presence of mature erythrocytes (stage 4b), which might be attributed to the change in respiration and the development of fully functional gills. In the differentiation of sensory structures, the development of eyes, inner ear, neuromasts and olfactory organs was rapid, with most of these organs becoming fully developed soon after hatching (stage 1). Vision, chemo- and mechano-reception developed very early in ontogeny, in parallel with the development of the central nervous system and changes in feeding habits. Although the general pattern of development in S. senegalensis appeared similar to most marine fish larvae already described, there were species-specific ontogenetic characteristics probably derived from the species' particular environment (subtropical waters) and behaviour (nocturnal, benthic, omnivorous feeding habits). These results on the organogenesis of larvae are a useful tool for establishing the functional systemic capabilities and physiological requirements of larvae to ensure optimal welfare and growth under aquaculture conditions.
Two groups of Senegal sole (Solea senegalensis) larvae were cultured. One used rotifers for the first 10 days after hatching (dph) and enriched Artemia metanauplii from 6 to 30 dph and the other without rotifers, using enriched Artemia metanauplii as the sole food source. The quantity of metanauplii used was the same (group A), twice (group A2) and three times (group A3) the quantity of live prey (in dry weight) of the group fed with rotifers (group R). At the end of the experiment, the growth, in terms of total length and individual dry weight was significantly higher for the larvae on group A2 whilst rotifer fed larvae (R) showed the poorest results. Eye migration was also delayed by approximately 2 days in the group R. No significant differences were found in survival rates. The influence of diet on Senegal sole metamorphosis and its relationship with the size rather than the age of the fish are discussed.
