Whole body vibration involves the exposure of the entire human body to direct contact with environmental vibration. Chronic mechanical vibrations, combined with the physical attributes of the human body, can amplify the incoming energy and present the potential for negative health effects. Vibration exposure can, thus, result in adverse health effects such as spinal injuries, abdominal neurological and cardiovascular disorders. These can manifest indirectly as an accident causal factor. The aim of our research is to study the impact of vibration fluctuations of different frequencies on the structural and functional condition and mechanisms of bone remodelling. An experimental study was, therefore, conducted on mature male rats. For assessment of bone metabolism in the venous blood of rats, osteocalcin level was determined, while fragments of rats’ lumbar vertebrae were subsequently taken for histologic examination. Our work revealed that with the increase of vibration frequency, an increase of osteocalcin level in the blood of experimental animals comes about. Moreover, we noted after terminating vibration fluctuations on the 56th day of the experiment, osteocalcin levels are gradually reduced. In addition, in the course of histological study of specimens of lumbar vertebrae bone tissue, even as early as of the 28th day of the experiment, evidences of acute impairment of the bone tissue and initial signs of its remodelling are clearly traced. Indeed, on the 56th day, the remodelling processes represented by enhanced regeneration in the zone of the cartilage plate, increased in proliferation activity. We also saw hyperplasia of chondrocytes, hypertrophy of the respective zones of cartilage tissue, zones of forming immature bone tissue on the areas of previous damage, focal replacement fibrosis and angiomatosis. Hence, with increasing vibratory acceleration of 0,5 g, the rate of bone metabolism grows, osteoblast activation processes are accelerated and the impairment of collagen and calcium loss is increased. All this leads subsequently to the occurrence of osteoporosis.
Study of the influence of vibration oscillations of different frequency, amplitude and vibration acceleration on the structural and functional state and mechanisms of muscle tissue remodelling. An experimental study was conducted on sexually mature male rats. The rats of the four experimental groups were subjected to vertical vibration oscillations of 15, 25, 50 and 75 Hz, respectively. It has been established that pathological changes in muscle tissue in the form of different variants of damage and remodelling tend to increase, which correlates with the frequency of vibration, amplitude and vibration acceleration level, as in the 2nd group, where the maximum permissible vibration levels did not exceed the established allowable norms, and in other groups of animals, where the permissible levels of total vibration were exceeded. By increasing vibration acceleration for more than 1.25 m/s2 (0.13 g, frequency more than 25 Hz and amplitude of 2 mm), severe damages are observed in the form of alterative changes of muscle fibres with the disappearance of transverse strain, homogenization of sarcoplasm, fragmentation with dissociation fibres on separate beams, partial and subtotal myocytolysis, and necrosis of separate fibres. Inflammation is rapidly increasing with the increase in the frequency of vibration and the level of vibration acceleration for more than 5.0 m/s2 (0.51 g).

Під дією вібраційних коливань можуть виникати зрушення у співвідношенні різних груп жирних кислот, змінюватися ступені їхньої ненасиченості. Дисбаланс між насиченими, мононенасиченими та поліненасиченими жирними кислотами, який виникає надалі у клітинній стінці, порушує плинність і в'язкість ліпідної фази та спричиняє порушення клітинного метаболізму.Мета роботи -вивчити вплив вібраційних коливань на рівень жирних кислот загальних ліпідів м'язової тканини та жирнокислотного складу фосфоліпідів м'язів і головного мозку експериментальних тварин, які піддавалися впливу вертикальних вібраційних коливань із різною частотою протягом 28 днів.Матеріали та методи. Фрагменти тканин чотириголового м'яза стегна та головного мозку щурів використовували для отримання метилових ефірів жирних кислот, що досліджували методом газорідинної хроматографії.Результати. Встановили, що вміст ліпідів, співвідношення їхніх окремих фракцій та жирнокислотний склад у м'язовій тканині та мозку тварин під час дії вібраційних коливань істотно змінюються. Зі збільшенням віброприскорення спостерігається тенденція до збільшення абсолютної кількості жирних кислот загальних ліпідів шляхом зростання рівня насичених і мононенасичених. Ці процеси зумовлені активацією механізмів самозахисту організму за умов відхилень від стабілізованої фізіологічної норми, оскільки адаптація вимагає від організму певних структурно-енергетичних витрат. Прослідковується збільшення відносної кількості насичених і мононе-насичених жирних кислот у фосфоліпідах м'язів, головного мозку й одночасне зниження концентрації поліненасичених жирних кислот.Висновки. Зміни служать свідченням про погіршення структурно-функціональної організації клітинних мембран м'язів і головного мозку щурів унаслідок збільшення в'язкості ліпідів, зміни їхньої внутрішньомембранної динаміки. Aim: To determine the frequency of NOD2 gene c.3019-3020insC (rs5743293) and c.2104C>T (rs2066844) allelic variants in the patients with Crohn’s disease (CD), colorectal cancer (CRC) and in the control groups and to study the association of these mutations with the onset time of the diseases, gender and surgical interventions. Materials and Methods: The diagnoses of CD and CRC were established based on standard clinical examination and laboratory tests. Molecular genetic study of a frameshift 3020insC mutations of NOD2 gene were performed in 54 patients with CD; missense R702W mutations of the NOD2 gene — in 41 CD patients and 38 healthy controls. In CRC group, 3020insC mutation was tested in 48 patients, R702W mutation — in 40 patients and 40 healthy controls. PCR-RFLP technique was used to identify the mutations. Results: The frequency of the minor allele (M) of 3020insC mutation of NOD2 gene in the patients with CD was significantly higher than in the control group (р = 0.01). The age at CD onset in females carrying 3020insC mutation was significantly lower (22.5 ± 1.6 years) when compared with females without the mutation (32.7 ± 2.5 years) (p = 0.002). There was no significant difference in the allele frequencies and genotype distributions of R702W mutation in the patients with CD in comparison with the controls. The mean age at CD onset in the patients carrying R702W mutation was significantly lower (28.4 ± 1.4 years) compared with the patients without the mutation (39.4 ± 2.8 years) (p < 0.01). Surgical interventions for CD was required in 40.0% of 3020insC mutation carriers. Among patients with CRC, only 4.2% carried 3020insC mutation and 20.0% R702W mutation. Our study suggests that R702W and 3020insC mutations are not associated with the risk of CRC in Ukrainian patients. There was no statistically significant difference in mean age at CRC onset in patients with/without R702W mutation. Only one patient with CRC had two mutations. Conclusion: The earlier age at CD onset was associated with 3020insC mutation, but only in female patients. The association between R702W mutation and the earlier age of CD onset was found. Patients with 3020insC mutation showed a trend to a higher frequency of surgical interventions for CD.
