Leishmaniasis, Chagas' disease and schistosomiasis (bilharzia) are parasitic diseases with wide distribution on the American continent, affecting millions of people. In the present study, biological assays for antiprotozoal and molluscicidal activities were carried out with ethanolic extracts of plant species from the Brazilian part of the Upper Paraná River. Crude extracts were obtained by percolation with absolute ethanol from the leaves of Cayaponia podantha Cogn., Nectandra falcifolia (Nees) Castiglioni and Paullinia elegans Cambess., as well as from the aerial parts of Helicteres gardneriana St. Hil. & Naud. and Melochia arenosa Benth., all belonging to genera used in folk medicine. Trypanocidal activity of plants was assayed on epimastigote cultures in liver infusion tryptose. Anti-leishmanial activity was determined over cultures of promastigote forms of the parasite in Schneider's Drosophila medium. Microscopic countings of parasites, after their incubation in the presence of different concentrations of the crude extracts, were made in order to determine the percentage of growth inhibition. C. podantha and M. arenosa, at a concentration of 10 µg/mL, showed 90.4 ± 11.52 and 88.9 ± 2.20% growth inhibition, respectively, of epimastigote forms of Trypanosoma cruzi, whereas N. falcifolia demonstrated an LD 50 of 138.5 µg/mL against promastigote forms of Leishmania (Viannia) braziliensis. Regarding molluscicidal activity, the acute toxicity of the extracts on Biomphalaria glabrata was evaluated by a rapid screening procedure. M. arenosa was 100% lethal to snails at 200 µg/mL and showed an LD 50 of 143 µg/mL. Screening of plant extracts represents a continuous effort to find new antiparasitic drugs.

From cultivation of a strain of P. tinctorius isolated from Pinus taeda, four known triterpenes lanostane derivatives, 3β,22ξ-dihydroxy-24-methyllanosta-8,24(28)-diene, 3β,22ξ-dihydroxy-24-ethyllanosta-8,24(28)-diene, 3β-hydroxylanosta-8,24-diene (lanosterol) and 3β-hydroxylanosta-7(8),9,24-triene (agnosterol), were isolated. These compounds were identified on the basis of chemical and physical methods, mainly mass spectrometry and 1D and 2D nuclear magnetic resonance.
RESUMO: "Avaliação das atividades moluscicida e cercaricida (Schistosoma mansoni) dos extratos de Croton floribundus e do ácido caurenóico". Extratos de Croton fluribundus (Euphorbiaceae), ácido caurenóico e dois derivados do ácido caurenóico foram avaliados como moluscicida, cercaricida e também foi verificada a letalidade destas amostras frente a larvas de Artemia salina Leach. Nestes ensaios foram observadas significantes atividades moluscicida e cercaricida associadas a uma reduzida toxicidade frente ao camarão de água salgada. Unitermos: Biomphalaria glabrata, Croton floribundus, Artemia salina, ácido caurenóico, Schistosoma mansoni, cercárias.ABSTRACT: Lethality of the extracts of Croton floribundus (Euphorbiaceae), a medicinal plant from south Brazil, and of the kaurenoic acid, an isolated compound, and two of its derivatives against adult Biomphalaria glabrata snails, Schistosoma mansoni cercariae and Artemia salina Leach. brine shrimp larvae are reported. Both extracts and the isolated compound showed significant molluscicidal and cercaricidal activities and reduced toxicity in brine shrimp assays.

The methanol crude extract of the leaves of Ficus radicans Roxb. 'Variegata' (Moraceae) and the n-hexane, ethyl acetate and aqueous methanol fractions resulting from its fractionation were evaluated for their anti-inflammatory, molluscicidal and free-radical scavenging activities. The crude extract and fractions exhibited significant inhibition of inflammation in both croton oil (CO)-induced ear oedema in mice (p<0.001) and carrageenan-induced rat paw oedema models (p<0.01). The molluscicidal assay against Biomphalaria glabrata showed a weak activity for the n-hexane fraction (DL(50)= 400 µg mL(-1)). A moderated 1,1-diphenyl-1-picrylhydrazyl (DPPH) free-radical scavenging activity was observed for the ethyl acetate fraction (IC(50)= 66.2 µg mL(-1)). Fractionation of the extracts through chromatographic methods afforded the coumarins 7-methoxycoumarin, 7-hydroxy-6-methoxycoumarin and methoxy-3,4-dihydrocoumarin, the steroids β-sitosterol and β-sitosterol 3-O-β-glucopyranoside, as well as a cinnamic acid derivative and a flavonoid identified as trans-4-methoxy-2-β-D-glucopyranosyloxy cinnamic acid and quercetin 3-O-β-D-xylopyranosyl-(1 → 2)-α-L-rhamnopyranoside, respectively. The compounds were identified on the basis of their NMR spectral data and comparison with those previously reported in the literature.

The phytochemical investigation of Coussarea platyphylla led to the isolation of triterpenes betulonic and betulinic acid, monoterpenes monotropein and monotropein salt, the diterpene trans-phytol and esteroids. The structures of the isolated compounds were assigned on the basis of spectroscopic data, including two-dimensional NMR methods. PARTE EXPERIMENTAL Procedimentos experimentais geraisOs espectros de RMN (uni e bidimensionais) foram obtidos em espectrômetro Varian, modelo Mercury plus BB, operando a 300 MHz para 1 H e 75,5 MHz para 13 C. Os deslocamentos químicos foram dados em ppm, tendo como referência interna o tetrametilsilano TMS (δ= 0,0 ppm) ou o próprio solvente. Os solventes utilizados foram D 2 O, CD 3 OD e CDCl 3 . Para as cromatografias em coluna (CC) utilizou-se gel de sílica 60 (0,063-0,200 mm, Merck) ou Sephadex LH-20 como fase estacionária. Para as cromatografias em camada delgada (CCD) e em camada delgada preparativa (CCDP) empregou-se gel de sílica 60 G e 60 GF254 (Merck). A visualização dos compostos em CCD foi realizada por irradiação com luz ultravioleta em 254 e 366 nm e/ou por pulverização com solução de H 2 SO 4 /MeOH (1:1), H 2 SO 4 / anisaldeído/ácido acético (1:0,5:50 mL) seguido de aquecimento.
