The aim of this study was to evaluate the effects of different temperatures in vacuum drying of Arthrospira Spirulina sp. on the physical properties and on the bioactive compounds, and to compare with the traditional oven dryer. The samples were evaluated according to phycocyanin content, phenolic compounds, lipid oxidation, color parameters and rehydration. The phenolic methanol extracts were analyzed by FTIR, and thermal analysis (DSC), scanning electronic microscopy and X-ray diffraction of the samples were also performed. The results showed that the characteristics of the dried products were influenced by the different temperatures of the drying methods. Vacuum drying at 40C was the more suitable to dry Arthrospira Spirulina sp. in relation to the characteristics products due to the minor losses of phycocyanin content, the great source of phenolic compounds, the minor lipid oxidation, the good rehydration capacity and the highest thermal stability. PRATICAL APPLICATIONSArthrospira Spirulina sp. is a microalgae which is a promising source of compounds with biological activity that could be used as functional ingredients, due to presence of pigments, phenolic compounds and fatty acids. However, this algae is very perishable and it should be dried after cultivation in order to prevent fungal deterioration. Phycocyanin content, phenolic compounds and lipid oxidation of the Arthrospira Spirulina sp. are affected by temperature and time drying processing. Thus, the vacuum drying process is a technique which leads to a product with best quality due to the low air temperatures and less oxygen content present in environment of dryer under subatmospheric pressure. Therefore, selection of the most appropriate technique and the drying conditions are very important to avoid major losses of bioactive compounds during the drying process of Arthrospira Spirulina sp.
RESUMO -As microalgas têm sido amplamente cultivadas e estudadas devido à sua rica composição química para fins alimentícios, farmacêuticos, biocombustíveis, biofilmes, dentre outras aplicações. A secagem é uma operação unitária que tem sido empregada para conservação dos nutrientes da biomassa, sendo a secagem a vácuo pouco explorada. Diante disso, este trabalho teve como objetivo de avaliar a secagem a vácuo da microalga Spirulina LEB-18 em relação ao conteúdo de ficocianina, compostos fenólicos e atividade antioxidante. A microalga foi cedida pelo Laboratório de Engenharia Bioquímica da FURG, sendo cultivada em fotobiorreatores abertos, utilizando meio de cultivo Zarrouk. A biomassa era prensada atingindo uma concentração de sólidos de 18% (g/g). Os experimentos foram realizados em uma estufa a vácuo à pressão de 100 mmHg (13,3 kPa) nas temperaturas 40, 50 e 60°C. Os resultados mostraram que com o aumento da temperatura houve uma redução de 26% a 70% no conteúdo de ficocianina, sendo que na menor temperatura (40°C) obteve uma maior preservação da ficobiliproteína. Em relação às demais respostas, compostos fenólicos e atividade antioxidante, os resultados foram similares à amostra in natura independente da temperatura utilizada. INTRODUÇÃOAs microalgas são uma das fontes mais promissoras para desenvolvimento de novos produtos alimentares e alimentos funcionais, sendo utilizadas para melhorar o valor nutritivo dos alimentos, devido à sua composição química equilibrada. Há um grande número de espécies de microalgas sendo cultivadas, sendo as cianobactérias as mais estudadas e utilizadas (Batista et al., 2013).A Spirulina é uma microalga fotossintética, filamentosa, espiralada, multicelular e de coloração azul-esverdeada que tem sido produzida em ampla escala (Koru, 2012). Possui elevado conteúdo de proteína (50-60%), ácidos graxos essenciais, vitaminas e minerais, sendo utilizada para fins farmacêuticos, suplementos alimentares, biocombustíveis, dentre outros. Na região sul do Brasil, situado na Lagoa da Mangueira, há uma planta piloto que possui uma capacidade de produzir 50 kg por mês de biomassa para enriquecer alimentos, nos quais são distribuídos como snacks para crianças na região (Morais et al., 2010). A microalga Arthrospira (Spirulina), além do seu elevado teor de proteínas, apresenta uma rica fonte de pigmentos como a ficocianina, que possui ação antioxidante e anti-inflamatória, além de ser utilizada como corantes naturais em substituição aos sintéticos. Em vista da microalga ser termicamente instável, este trabalho tem como objetivo de analisar diferentes técnicas de secagem e métodos de ruptura para obter um maior rendimento de ficocianina. A Arthrospira Spirulina LEB-18 foi cedida pelo Laboratório de Engenharia Bioquímica-LEB da FURG, com concentração de 20% (m/m) de sólidos. As técnicas de secagem utilizadas foram o leito de jorro e a bandeja com escoamento paralelo do ar, e os métodos de ruptura foram moagem, micro-ondas e autoclave. As amostras de Arthrospira Spirulina foram analisadas quanto ao conteúdo de ficocianina, e seus extratos aquosos foram analisados no espectro de infravermelho (FTIR) para identificação dos grupamentos característicos da ficocianina. Os produtos desidratados foram avaliados quanto sua estrutura morfológica pela microscopia eletrônica de varredura (MEV). A secagem em leito de jorro combinada com o rompimento em micro-ondas apresentou melhores resultados em relação às amostras in natura e as secas sem ruptura. Os espectros de FTIR apresentaram os grupamentos característicos da ficocianina. INTRODUÇÃOAs cianobactérias são organismos aquáticos e fotossintéticos conhecidos pelos seus ricos pigmentos e nutrientes, contendo elevado conteúdo proteico, sendo empregados em suplementos alimentares. Dentre as cianobactérias destaca-se a Arthrospira Spirulina, que possui atividade anticâncer e antimicrobiano via produção de compostos antioxidantes e pigmentos como ficocianina, ficocianobilina, aloficocianina dentre outros produtos (NUHU, 2013).A ficocianina é um dos principais pigmentos da Arthrospira Spirulina que tem sido amplamente estudada devido à sua aplicação como corantes alimentícios e uso em cosméticos, em substituição aos corantes sintéticos (MARTELLI et al., 2014). A ficocianina é uma proteína pertencente à família das ficobiliproteínas caracterizada pela intensa coloração azul, e sua estrutura consiste de uma proteína e componentes não-proteicos conhecido como as ficocianobilinas (FERNÁNDEZ-ROJAS, HERNÁNDEZ-JUÁREZ & PEDRAZA-CHAVERRI, 2014).
