O controle de qualidade em indústrias farmacêuticas tem sido aprimorado de maneira gradual com a implementação de novas estratégias e tecnologias, de modo a possibilitar a obtenção de produtos cada vez mais eficazes e seguros. Com surgimento do conceito de Quality by Design (QbD), a produção de medicamentos passou a ser otimizada por meio da delimitação dos parâmetros de qualidade dos produtos desde os estágios iniciais de desenvolvimento e planejamento da formulação a partir das características do insumo farmacêutico ativo, visando a prevenção de defeitos e melhoria contínua da cadeia produtiva. Sendo assim, o presente estudo constitui uma análise quantitativa de notificações de irregularidades em medicamentos sólidos publicadas no site oficial da Agência Nacional de Vigilância Sanitária (ANVISA) durante o período de 2017 a 2019, a partir do uso da metodologia de Análise de Modos de Falhas e Efeitos (FMEA) no contexto de QbD. Ao todo, foram analisadas 421 notificações de produtos farmacêuticos irregulares, das quais 28,5% correspondiam a queixas técnicas, sendo 60,0% delas referentes a medicamentos sólidos. Após a avaliação dos dados, verificou-se que aproximadamente 80,0% das não-conformidades encontradas estavam relacionadas a aspecto, dissolução, embalagem, doseamento e pureza, sendo o QbD uma estratégia eficiente para o gerenciamento dos riscos relacionados à produção, visando a redução de custos e implementação de estratégias que possibilitem a diminuição gradativa do número de queixas técnicas destes produtos.
Diante o crescimento da indústria farmacêutica nos últimos anos, diferentes abordagens podem ser adotadas no delineamento de processos de fabricação de medicamentos, visando a obtenção de produtos cada vez mais seguros e eficazes. A utilização de diferentes estratégias e tecnologias são necessárias para assegurar a identificação de todos os potenciais pontos de falhas e a otimização dos controles implementados nas etapas de manufatura. Neste contexto, o conceito de Quality by Design (QbD), amplamente promovido pela agência Food and Drug Administration (FDA) e pela Conferência Internacional de Harmonização (ICH) vem se destacando no Brasil, por consistir em uma abordagem fundamentada no desenvolvimento sistemático de produtos por meio da utilização de diversas ferramentas, com base no gerenciamento de riscos de qualidade aliado à redução de custos, a fim de promover a melhoria contínua da cadeia de produção de medicamentos. O estudo apresentou uma análise quantitativa das causas de falhas potenciais notificadas pelo site oficial da ANVISA durante o período de 2020 a 2022, através da avaliação do RPN elaborada pela ferramenta FMEA dentro do contexto de QbD. Foram analisadas 453 queixas técnicas, sendo 71 queixas (15,7%) sobre medicamentos líquidos, sendo aproximadamente 77,5% referentes a problemas farmacotécnicos como embalagem, aspecto e formulação. O conceito QbD para gestão de riscos e princípios de fabricação, visa a redução de custos e implementação de estratégias de controle no desenvolvimento do processo e da formulação, que possibilitem a diminuição gradativa do número de queixas técnicas destes produtos, assim como a redução de falhas e erros humanos.
