Summary1. How thermal tolerance estimated in the laboratory can be extrapolated to natural settings remains a contentious subject. Here, we argue that the general premise that a single temperature can accurately describe upper or lower tolerance limits is incorrect. 2. Survival probability is determined by both the intensity and the duration of a thermal stress, and the association between these variables can be adequately conveyed by a thermal tolerance landscape. Employing this framework, we demonstrate that the temperature range that an organism can tolerate is expected to narrow down with the duration of the thermal challenge. 3. Analyses suggest that a trade-off exists between tolerances to acute and chronic exposition to thermal stress, and that changes in temperature means or extremes may result in drastically different selective pressures and subsequent evolutionary responses. 4. After controlling for the duration of the thermal challenge, we also uncover latitudinal effects on upper lethal temperatures in insects that remained unnoticed in previous broad-scale comparative analyses. 5. Ultimately, critical thermal limits have been adopted in the ecological literature for logistic reasons and are inadequate descriptors of thermal tolerance on conceptual grounds. We consider that tolerance landscapes provide a more suitable framework to study temperature tolerance and its potential impact in ecological settings.
Models of host-parasite coevolution predict pronounced genetic dynamics if resistance and infectivity are genotype-specific or associated with costs, and if selection is fueled by sufficient genetic variation. We addressed these assumptions in the black bean aphid, Aphis fabae, and its parasitoid Lysiphlebus fabarum. Parasitoid genotypes differed in infectivity and host clones exhibited huge variation for susceptibility. This variation occurred at two levels. Clones harboring Hamiltonella defensa, a bacterial endosymbiont known to protect pea aphids against parasitoids, enjoyed greatly reduced susceptibility, yet clones without H. defensa also exhibited significant variation. Although there was no evidence for genotype-specificity in the H. defensa-free clones' interaction with parasitoids, we found such evidence in clones containing the bacterium. This suggests that parasitoid genotypes differ in their ability to overcome H. defensa, resulting in an apparent host × parasitoid genotype interaction that may in fact be due to an underlying symbiont × parasitoid genotype interaction. Aphid susceptibility to parasitoids correlated negatively with fecundity and rate of increase, due to H. defensa-bearing clones being more fecund on average. Hence, possessing symbionts may also be favorable in the absence of parasitoids, which raises the question why H. defensa does not go to fixation and highlights the need to develop new models to understand the dynamics of endosymbiont-mediated coevolution.
Summary1. An increasing body of knowledge suggests that the estimation of critical upper thermal limits (CT max ) is highly dependent on the experimental methodology employed. Here, we employ a theoretical approach to analyse how estimates of CT max (knock-down temperatures and times) are affected by measurement protocol. 2. Our model is able to reproduce the results of empirical studies on Drosophila melanogaster, suggesting that it adequately mimics organismal responses during assays. With simulated data sets, we also show that many experimental protocols result in unreliable and often highly biased estimates of CT max in Drosophila and possibly in other ectotherms. 3. The confounding effects of stochasticity, resource depletion (or fatigue) and short-term acclimatory responses are expected to be higher in longer assays, and therefore, short assays should be generally preferred. The experimental protocol of choice must also take into consideration the range in which measurement accuracy is not affected and the potential problems of thermal inertia in larger organisms. 4. Our findings justify previous concerns that the methodology may have a greater impact on estimates of CT max than the biological process under study, and explain why many studies on the subject have often reported inconsistent and even contradictory results.
Susceptibility to global warming relies on how thermal tolerances respond to increasing temperatures through plasticity or evolution. Climatic adaptation can be assessed by examining the geographic variation in thermal-related traits. We studied latitudinal patterns in heat tolerance in Drosophila subobscura reared at two temperatures. We used four static stressful temperatures to estimate the thermal death time (TDT) curves, and two ramping assays with fast and slow heating rates. Thermal death time curves allow estimation of the critical thermal maximum (CT(max)), by extrapolating to the temperature that would knock down the flies almost "instantaneously," and the thermal sensitivity to increasing stressful temperatures. We found a positive latitudinal cline for CT(max), but no clinal pattern for knockdown temperatures estimated from the ramping assays. Although high-latitude populations were more tolerant to an acute heat stress, they were also more sensitive to prolonged exposure to less stressful temperatures, supporting a trade-off between acute and chronic heat tolerances. Conversely, developmental plasticity did not affect CT(max) but increased the tolerance to chronic heat exposition. The patterns observed from the TDT curves help to understand why the relationship between heat tolerance and latitude depends on the methodology used and, therefore, these curves provide a more complete and reliable measurement of heat tolerance.
While there are substantial studies suggesting that characteristics of wine are related to regional microbial community composition (microbial terroir), there has been little discussion about what factors affect variation in regional microbial community composition. In this study, we compared the microbial community composition of leaves and berries of a grape variety (Carmenere) from six different Chilean vineyards within 35 km of each other. In order to determine relationships between spatial proximity and microbial compositional dissimilarity, we sequenced amplicons of the internal transcribed spacer (ITS) region for fungi and 16S rRNA gene for bacteria. Results showed that both the fungal and the bacterial community compositions of the studied vineyards differed, but this difference was much clearer in fungi than in bacteria. In addition, while bacterial community dissimilarity was not correlated with geographic distance, the leaf and berry fungal community dissimilarities between locations increased with geographic distance. This indicates that spatial processes play an important role in structuring the biogeographic pattern of grape-associated fungal communities at local scales, which might in turn contribute to the local identity of wine.
The potential of populations to evolve in response to ongoing climate change is partly conditioned by the presence of heritable genetic variation in relevant physiological traits. Recent research suggests that Drosophila melanogaster exhibits negligible heritability, hence little evolutionary potential in heat tolerance when measured under slow heating rates that presumably mimic conditions in nature. Here, we study the effects of directional selection for increased heat tolerance using Drosophila as a model system. We combine a physiological model to simulate thermal tolerance assays with multilocus models for quantitative traits. Our simulations show that, whereas the evolutionary response of the genetically determined upper thermal limit (CTmax) is independent of methodological context, the response in knockdown temperatures varies with measurement protocol and is substantially (up to 50%) lower than for CTmax. Realized heritabilities of knockdown temperature may grossly underestimate the true heritability of CTmax. For instance, assuming that the true heritability of CTmax in the base population is h2 = 0.25, realized heritabilities of knockdown temperature are around 0.08–0.16 depending on heating rate. These effects are higher in slow heating assays, suggesting that flawed methodology might explain the apparently limited evolutionary potential of cosmopolitan D. melanogaster.
Mediterranean biomes are biodiversity hotspots, and vineyards are important components of the Mediterranean landscape. Over the last few decades, the amount of land occupied by vineyards has augmented rapidly, thereby increasing threats to Mediterranean ecosystems. Land use change and agricultural management have important effects on soil biodiversity, because they change the physical and chemical properties of soil. These changes may also have consequences on wine production considering that soil is a key component of terroir. Here, we describe the taxonomic diversity and metabolic functions of bacterial and fungal communities present in forest and vineyard soils in Chile. To accomplish this goal, we collected soil samples from organic vineyards in central Chile and employed a shotgun metagenomic approach to sequence the microbial DNA. Additionally, we studied the surrounding native forest to obtain a baseline of the soil conditions in the area prior to the establishment of the vineyard. Our metagenomic analyses revealed that both habitats shared most of the soil microbial species. The most abundant genera in the two habitats were the bacteria Candidatus Solibacter and Bradyrhizobium and the fungus Gibberella. Our results suggest that the soil microbial communities are similar in these forests and vineyards. Therefore, we hypothesize that native forests surrounding the vineyards may be acting as a microbial reservoir buffering the effects of the land conversion. Regarding the metabolic diversity, we found that genes pertaining to the metabolism of amino acids, fatty acids, and nucleotides as well as genes involved in secondary metabolism were enriched in forest soils. On the other hand, genes related to miscellaneous functions were more abundant in vineyard soils. These results suggest that the metabolic function of microbes found in these habitats differs, though differences are not related to taxonomy. Finally, we propose that the implementation of environmentally friendly practices by the wine industry may help to maintain the microbial diversity and ecosystem functions associated with natural habitats.
