Diffusion-ordered spectroscopy (DOSY) is an effective method for the analysis of intact mixtures, but the quality of results is critically limited by resolution in the NMR dimension. A new experiment integrating diffusion weighting into the PSYCHE method for pure shift NMR spectroscopy allows DOSY spectra to be measured with ultrahigh NMR resolution at improved sensitivity.
Approximately 255 million people consume illicit drugs every year, among which 18 million use cocaine. A portion of this drug is represented by crack, but it is difficult to estimate the number of users since most are marginalized. However, there are no recognized efficacious pharmacotherapies for crack-cocaine dependence. Inflammation and infection in cocaine users may be due to behavior adopted in conjunction with drug-related changes in the brain. To understand the metabolic changes associated with the drug abuse disorder and identify biomarkers, we performed a 1 H NMR-based metabonomic analysis of 44 crack users' and 44 healthy volunteers' blood serum. The LDA model achieved 98% of accuracy. From the water suppressed 1 H NMR spectra analyses, it was observed that the relative concentration of lactate was higher in the crack group, while long chain fatty acid acylated carnitines were decreased, which was associated with their nutritional behavior. Analyses of the aromatic region of CPMG 1 H NMR spectra demonstrated histidine and tyrosine levels increased in the blood serum of crack users. The reduction of carnitine and acylcarnitines and the accumulation of histidine in the serum of the crack users suggest that histamine biosynthesis is compromised. The tyrosine level points to altered dopamine concentration.
The long-range scalar coupling constant between proton and fluorine nuclei, J , is observed to be larger than J in the pyrimidinyl moiety of voriconazole. A set of smaller molecules is chosen (fluorobenzene, N-methyl-2-fluoropyridine, N-methyl-3-fluoropyridine, 3-fluoropyridine, 5-pyrimidine, and 2-fluoropyridine) to evaluate the influence of the nitrogen atom in the experimental J values. Spectral aliased pure shift heteronuclear single quantum coherence spectroscopy (SAPS-HSQC) is applied to determine the relative sign between the J and J scalar couplings. Theoretical calculations show that the J and J coupling constants can be described mainly by a Fermi contact (FC) transmission mechanism. A decomposition analysis of J in terms of localized molecular orbital (LMO) contributions allows us to determine that the interaction involving the nitrogen lone pair (LPN) is the main reason for the larger J compared to J . Our analysis indicates that delocalization of LPN has a positive contribution to the long-range coupling, while a negative one is observed for J .
BACKGROUND: Entomopathogenic fungi can provide a set of ecological services, such as suppressing arthropod pests and plant pathogens. In this study, novel indigenous Beauveria caledonica (Bc) strains were isolated from naturally infected banana weevils (Cosmopolites sordidus) occurring in commercial banana plantations in Brazil.RESULTS: The prevalence of infection by Bc strains on field-caught C. sordidus ranged from 1.3% to 12.9%. Similar to the Beauveria bassiana strains tested, none of the Bc strains caused more than 50% weevil mortality at a concentration of 1 × 10 8 conidia ml −1 . Bc strain CMAA1810 caused the highest mortality in C. sordidus and had enhanced insecticidal activity when formulated with an emulsifiable oil. In paired co-culture assays, this same strain showed a significant growth-inhibitory effect on the causal agent of Fusarium banana wilt (Fusarium oxysporum f. sp. cubense, Foc) of twofold magnitude compared with the control. Cell-free crude filtrates derived from the red-pigmented culture broth of Bc (CMAA1810) strongly reduced Foc conidial viability, and this inhibitory activity was inversely related to the age of the Bc culture. Crude concentrated filtrates from 4-dayold cultures exhibited the strongest antifungal activity (13-fold) compared with untreated Foc conidia. The abundant compound identified in the crude filtrate of Bc was oosporein (1,4-dibenzoquinone) present at a concentration of 0.829 ± 0.018 mg g −1 dry matter, and the antifungal activity of the filtrate was demonstrated.CONCLUSION: These results indicated that Bc strains might have the potential to manage both C. sordidus and Foc, two of the major phytosanitary problems in banana crops worldwide. Further research under field conditions using suitable formulations of virulent Bc strains in combination with the metabolite oosporein is needed to evaluate their efficacy in the management of C. sordidus and Foc in banana plantations.
