In this study, we determined whether solid-state fermentation could degrade major allergens and reduce potential allergenicity of soybean meal (SBM). Solid-state fermentation was realized through a mixture of Lactobacillus casei, yeast, and Bacillus subtilis. High-performance liquid chromatography, size exclusion-high-performance liquid chromatography, and capillary liquid chromatography/tandem mass spectrometry coupled with electrospray ionization were used to examine the total amino acids and molecular weight distribution of the fermented soybean meal (FSBM). In addition, the potential allergenicity of FSBM was assessed by conducting in vitro competitive inhibition ELISA and oral sensitization and challenge of a BALB/c mice model. The results indicated that the total amino acid content increased and soy protein was degraded into polypeptides with low molecular weights that were derived from the hydrolysis of the allergen sequences N232-D383, G253-I265, E169-S215, G68-G98, A365-I375, and V153-A167. Moreover, the FSBM group exhibited a lower in vitro immunoglobulin E (IgE)-binding capacity than the SBM group. The BALB/c model indicated that the FSBM group manifested milder damage to the intestine, lower mMCP-1 and IgE levels, and higher IFN-γ levels as compared to the SBM group. These findings suggested that the potential allergenicity of SBM was reduced by the solid-state fermentation induced by the mixture of Lactobacillus casei, yeast, and Bacillus subtilis.

-Ergothioneine (EGT) widely existing
in mushrooms has various physiological activities. In this study,
the protective effects of EGT on dextran sulfate sodium (DSS)-induced
colitis mice were investigated.
Results
: It was observed
that EGT administration, especially at the high dose level, prevented
the body weight loss, the colon shortening, and the increase in disease
activity index and spleen index caused by DSS. Moreover, EGT supplementation
attenuated DSS-induced gut barrier damage by enhancing the expression
of tight-junction protein and recovering the loss of gut mucus layer.
Furthermore, EGT considerably decreased the colonic myeloperoxidase
(MPO) activity induced by DSS, but no significant differences were
observed in the concentrations of IL-6 and TNF-α in colon tissues.
Additionally, EGT downregulated the populations of CD4
+
T cells and macrophages, indicating that EGT stabilized the immune
response caused by DSS.
Conclusion
: Together these results
suggest that EGT can alleviate DSS-induced colitis and provide important
insights concerning the potential anticolitis activity of such food
products.
In recent years, the prevalence of allergic diseases has increased significantly, causing great concern, and wheat, as one of the top 8 food allergens, is a common allergy trigger. Nevertheless, reliable estimates of the positivity rate of wheat allergens in the allergic population in China are still lacking. The systematic review and meta-analysis aims to evaluate the positive detection rate of wheat allergens in the Chinese allergic population and further provide a reference for the prevention of allergy. CNKI, CQVIP, WAN-FANG DATA, Sino Med, PubMed, Web of Science, Cochrane Library, and Embase databases were retrieved. Related research and case reports about the positive rate of wheat allergen in the Chinese allergic population published from inception to June 30, 2022, were searched, and meta-analysis was performed using Stata software. The pooled positive rate of wheat allergens and 95% confidence interval were calculated by random effect models, and the publication bias was evaluated using Egger’s test. A total of 13 articles were included for the final meta-analysis, in which wheat allergen detection methods involved only serum sIgE testing and SPT assessment. The results showed that the wheat allergen positivity detection rate in Chinese allergic patients was 7.30% (95% CI 5.68–8.92%). Subgroup analysis showed that the positivity rate of wheat allergens was influenced by region, but hardly by age and assessment method. The positive rates of wheat allergy in the population with allergic diseases were 2.74% (95% CI 0.90–4.58%) and 11.47% (95% CI 7.08–15.87%) in southern and northern China, respectively. In particular, the positive rates of wheat allergens were greater than 10% in Shaanxi, Henan and Nei Mongol, all of which belong to the northern region. These results suggest that wheat allergens are an important cause of sensitization in allergic populations from northern China, and therefore attention should be paid to early prevention in high-risk populations.

Subgroup analysis showed that the positivity rate of wheat allergens was influenced by region, but hardly by age and assessment method. The positive rates of wheat allergy in the population with allergic diseases were 2.67% (95% CI: 0.86%-4.49%) and 12.68% (95% CI: 8.27%-17.10%) in southern and northern China, re-spectively. In particular, the positive rates of wheat allergens were greater than 10% in Shaanxi, Henan and Nei Mongo, all of which belong to the northern region.
Conclusions
These results suggest that wheat allergens are an important cause of sensitization in Chinese allergic populations, mainly in the northern allergic population. Therefore, attention should be paid to the early screening and effective prevention and treatment of high-risk groups.
