Both increases and decreases in methyl CpG-binding protein 2 (MeCP2) levels cause neurodevelopmental defects. We found that MeCP2 translation is regulated by microRNA 132 (miR132). Block of miR132-mediated repression increased MeCP2 and brain-derived neurotrophic factor (BDNF) levels in cultured rat neurons and the loss of MeCP2 reduced BDNF and miR132 levels in vivo. This feedback loop may provide a mechanism for homeostatic control of MeCP2 expression.
Combination therapy could be of use for the treatment of fungal infections, especially those caused by drug-resistant fungi. However, the methods and approaches used for data generation and result interpretation need further optimizing. The fractional inhibitory concentration index (FICI) is the most commonly used method, but it has several drawbacks in characterizing antifungal drug interaction. Alternatively, some new methods can be used such as the ⌬E model (difference between the predicted and measured fungal growth percentages) and the response surface approach, which uses the concentration-effect relationship over the whole concentration range instead of just the MIC. In the present study, in vitro interactions between tacrolimus (FK506) and three azoles-fluconazole (FLC), itraconazole (ITR), and voriconazole (VRC)-against Candida albicans were evaluated by the checkerboard microdilution method and time-killing test. The intensity of the interactions was determined by visual reading and the spectrophotometric method in a checkerboard assay, and the nature of the interactions was assessed by nonparametric models of FICI and ⌬E.
Colony counting and colorimetric viable detection methods (2,3-bis {2-methoxy-4-nitro-5-[(sulfenylamino) carbonyl]-2H-tetrazolium hydroxide} [XTT] reduction test)were used for evaluating the combination antifungal effects over time. Synergistic and indifferent effects were found for the combination of FK506 and azoles against azole-sensitive strains, while strong synergy was found against azole-resistant strains analyzed by FICI. The ⌬E model gave more consistent results with FICI. The positive interactions were also confirmed by the time-killing test. Our findings suggest a potential role for combination therapy with calcineurin pathway inhibitors and azoles to augment activity against resistant C. albicans.
DBIL was associated with a dose-response increased risk for CHD incidence. However, a U-shaped association existed between TBIL, indirect bilirubin and incident CHD risk. Key messages Direct bilirubin is independently associated with incident coronary heart disease (CHD) in a dose-response manner. A similarly consistent U-shaped association was found between total bilirubin, indirect bilirubin and incident CHD. The potential protective effect of total bilirubin within the normal range on incident CHD should be mainly attributed to mild-to moderate elevated levels of indirect bilirubin.
Cyclosporin A showed potent synergism when combined with the three azoles, especially against azole-R C. albicans strains, and there was good agreement between various methods used in this study.
Remotely sensed nighttime lights (NTL) datasets derived from the Defense Meteorological Satellite Program's Operational Linescan System (DMSP/OLS) have been identified as a good indicator of the urbanization process and have been widely used to study such demographic and economic variables as population distribution and density, electricity consumption, and carbon emission. However, one issue must be considered in the application of NTL data, i.e., saturation in the bright cores of urban centers. In this study, we evaluate four correction methods in China's cities: the linear regression model and the cubic regression model at the regional level, and the Human Settlement Index (HSI) and the Vegetation Adjusted NTL Urban Index (VANUI) at a pixel level. The results suggest that both correction methods at the regional level improve the correlation between NTL data and socioeconomic variables. However, since the methods can only be used on saturated pixels, the correction effects are limited, as the saturated area in Chinese cities is rather small. Proper methods for reducing saturation effects should thus vary with different study areas and research purposes.
In China, rapid urbanization has increased the demand for urban land and intensified the conflict between limited land resources and urban development. In response, high urban density has been proposed to realize sustainable urban development. Achieving this goal requires an examination of the dynamics of urban density in China. Nighttime light (NTL) data from the Defense Meteorological Satellite Program/Operational Linescan System (DMSP/OLS) are a good indicator of human activity. We applied NTL data to measure urban density in 70 major cities in China during 1992-2010. Based on temporal changes in NTL, we identified seven classes of urban density and clustered the distributions of urban density in 70 cities into six types. The dynamics of urban density were then obtained from the GDP density as an index of city development. The curves of urban density distribution gradually changed from a concave increase to W-shaped and S-shaped to a concave decrease, indicating that the current urban land use in China is unsustainable and that the shortage of land resources must be addressed. An examination of the distribution of urban density in Hong Kong revealed a different pattern and a potential solution for cities in mainland China.
